Dietary parsley seed powder attenuates acrylamide-waterborne toxicity hazards on growth, redox status, immunity and gene regulations in Oreochromis niloticus.

This study evaluated the protective role of dietary parsley seed powder (PSP) against acrylamide (ACR) toxicity in Nile tilapia (Oreochromis niloticus). A total of 200 juvenile fish (15.98 ± 0.01 g) were randomly assigned to four dietary groups (0.0, 2.5, 5.0 and 10.0 g PSP/kg diet) and exposed for 56 days to a sublethal ACR concentration (3.46 mg/L). Throughout the experiment, growth performance, blood biochemical indices, immune responses, oxidative stress biomarkers, digestive enzyme activities, histopathology and hepatic gene expression were thoroughly assessed. The results revealed that P

Bone marrow necrosis and rapidly progressive liver metastases after breast-conserving surgery in a 33-year-old woman with triple-negative breast cancer, followed by death 288 days after surgery: a case report.

BACKGROUND: Bone marrow necrosis (BMN) is rare, and malignancy-associated BMN often reflects high tumor burden, rapid progression, and poor prognosis. BMN after definitive treatment for early triple-negative breast cancer (TNBC), followed by visceral progression and coagulation-fibrinolytic abnormalities, is rarely reported. CASE DESCRIPTION: A 33-year-old woman underwent breast-conserving surgery and sentinel lymph node biopsy for a 1.8 cm grade 3 invasive carcinoma of no special type. One of four sentinel nodes was positive (pT1cN1a, cM0); the surgical specimen was estrogen receptor (ER)-ne

Association of Methotrexate-Induced Renal Toxicity in Humanized-Liver Mice With Human AOX1-Derived 7-Hydroxymethotrexate.

High doses of methotrexate are widely used, but its clinical application is limited by renal toxicity. The poorly soluble metabolite 7-hydroxymethotrexate contributes to renal injury, potentially through intratubular precipitation; however, its formation differs across species, complicating the evaluation of human-relevant toxicity in conventional animal models. In this study, methotrexate-induced renal toxicity was investigated using chimeric mice with humanized livers (humanized-liver mice). The pharmacokinetics of methotrexate and its major metabolite, 7-hydroxymethotrexate, were characteri

Ghrelin Receptor Blockade Restricts Hepatic Echinococcus granulosus Infection by Suppressing Host Cell Proliferation and Enhancing Fibrotic Encapsulation.

INTRODUCTION: Cystic echinococcosis (CE) is a parasitic liver disease caused by Echinococcus granulosus (E.g) infection, characterized by hepatic fibrosis and liver necrosis. Ghrelin and its receptor GHSR have been proven to regulate the progression of hepatic E.g infection, and GHSR knockout inhibits infection progression. Nevertheless, the specific regulatory mechanism remains poorly understood. METHODS: In this study, mice with hepatic E.g infection were intraperitoneally injected with ghrelin protein and the GHSR antagonist [D-Lys3]-GHRP-6 for 90 days, and the related molecular and pathol

Dual presentation of metastatic gestational trophoblastic neoplasia in mother and infant: an infrequent diagnostic challenge.

Gestational trophoblastic neoplasia (GTN) rarely presents concurrently in mother and infant, with very few cases reported worldwide. We describe an infant who presented with vomiting, lethargy and hepatomegaly. Emergency ultrasound revealed hepatomegaly, and contrast-enhanced CT showed large bilobar hepatic lesions with central necrosis and multiple bilateral pulmonary nodules. Despite supportive care, the infant's condition deteriorated, resulting in death. Post-mortem liver biopsy confirmed metastatic GTN. Shortly thereafter, the mother developed neurological symptoms; brain MRI revealed a h

Circulating spexin and adiponectin as early biomarkers of insulin resistance in pediatric obesity.

BACKGROUND: Insulin resistance (IR) is a central pathological feature of childhood obesity. Spexin (SPX), an emerging endogenous metabolic regulatory peptide, plays a key role in regulating energy metabolism. This study aimed to evaluate the association between IR and serum levels of SPX and adiponectin (ADP) and assess their biomarker potential for predicting IR in children with obesity. METHODS: In total, the study enrolled 104 children (40 normal weight and 64 obese) aged 7–18 years. Further, children with obesity were categorized as having IR (n = 37) or not (n = 27) using a homeostatic mo

Identification of key genes related to metabolic cell death in hepatic ischemia-reperfusion injury from transcriptome data and mechanism research using single-cell data.

BACKGROUND: Ferroptosis and cuproptosis are closely associated with hepatic ischemia-reperfusion injury (HIRI). However, the significance of metabolic cell death-related genes (MRGs) in HIRI still awaits exploration. This study examined the molecular mechanisms through which key genes contribute to metabolic cell death in HIRI. METHODS: GSE12720, GSE14951, and GSE171539 datasets and 478 MRGs were included. First, candidate genes were screened through differential expression analysis combined with MRGs. Then, key genes were identified by using machine learning algorithms combined with expressi

Pathophysiological and behavioral consequences of dichlorvos-induced oxidative stress in rodent models: A systematic review of mechanisms and protective strategies.

DDVP, a widely used organophosphate insecticide, exerts toxicity primarily through acetylcholinesterase inhibition and redox imbalance, culminating in multi-organ injury. This systematic review compiles findings from rodent studies to outline the toxicodynamic profile of DDVP, emphasizing oxidative disturbances, antioxidant depletion, biochemical alterations, tissue pathology, behavioral outcomes, and protective interventions. Oxidative stress emerges as a central mechanism, evidenced by increases in ROS (up to 433 % in brain cortex), MDA (200 % in hepatic mitochondria), PCO (105 % in brain co

TNF-α Inhibitors and HBV Reactivation Risk in HBsAg-/Anti-HBc+ Patients: A Systematic Review and Meta-Analysis.

BACKGROUND: Tumour necrosis factor-α (TNF-α) inhibitors are integral in the treatment of immune-mediated inflammatory diseases but may precipitate hepatitis B virus reactivation (HBVr) in HBsAg-/anti-HBc+ patients. APASL guidance presumes a greater risk with higher-potency agents, but the evidence is limited and conflicting. We conducted a systematic review and meta-analysis to assess whether TNF-α inhibitor potency alters HBVr risk in HBsAg-/anti-HBc+ individuals and to identify predictors. METHODS: PubMed, Embase, and the Cochrane Central Register of Controlled Trials were searched on May 2

Long-chain polyphosphates induce glomerular microthrombi and exacerbate LPS-induced acute kidney injury in mouse.

Polyphosphates (PolyPs) are evolutionarily conserved anionic polymers mediating pleiotropic functions in eukaryotes and prokaryotes, depending on their chain length. Bacteria typically synthetize long chains, while human platelets harbor exclusively medium chains. PolyP-mediated lung and liver-injury have been reported in experimental mouse models but their effects on the kidney remain undefined. Here, we assessed kidney histopathology and cytokine levels following intravenous administration of medium-chain (P100) and long-chain (P700) PolyPs and their synergistic effects with lipopolysacchari

Objective TCM syndrome differentiation in rheumatoid arthritis patients with cerebral infarction: The auxiliary diagnostic value of autoantibodies (anti-CCP IgG, AKA, and GPI) and hemoglobin.

The co-occurrence of rheumatoid arthritis (RA) and cerebral infarction is highly prevalent in clinical practice. While integrated traditional Chinese medicine (TCM) and Western medicine offers unique advantages in treatment, the objective indicators associated with TCM syndromes in this specific patient population remain unclear, hindering precise syndrome differentiation. This study, utilizing a screened cohort from 920 hospitalized patients with RA and cerebral infarction, aimed to address this research gap. Based on strict inclusion, exclusion, and elimination criteria, 142 patients were se

Biomarkers of inflammation and risk of adverse events in patients with cirrhosis.

Acute kidney injury (AKI) is a frequent and severe complication in patients with cirrhosis. Identifying patients at high risk for AKI progression and in-hospital complications could enable timely targeted interventions. Given the central role of inflammation in AKI and cirrhosis, we investigated whether inflammatory biomarkers could provide prognostic insights. In this multicenter, prospective cohort study, we enrolled 188 patients with cirrhosis and AKI from 4 tertiary care centers. Urine and plasma samples were analyzed for 4 biomarkers: urine monocyte chemoattractant protein-1 (MCP-1), urin

A web-based prognostic nomogram incorporating VETC-associated imaging markers for predicting postoperative recurrence in hepatocellular carcinoma.

BACKGROUND: Vessels that encapsulate tumor clusters (VETC) is a powerful predictor of aggressive hepatocellular carcinoma (HCC) and associated with poor outcomes of HCC. Imaging surrogates of VETC potentially help predict postsurgical recurrence. PURPOSE: To explore the noninvasive predictive potential of contrast-enhanced computed tomography (CE-CT) for VETC of HCC (VETC-HCC). A web-based prognostic nomogram model including VETC-associated imaging markers was subsequently created to predict postoperative recurrence-free survival (RFS) in HCC patients. METHODS: A retrospective evaluation was p

Integrated chemometric and pharmacological evaluation of processing-induced attenuation of hepatotoxicity in Polygonum multiflorum.

ETHNOPHARMACOLOGICAL RELEVANCE: Polygonum multiflorum (P. multiflorum) is the dried root tuber of P. multiflorum Thunb., a perennial herbaceous plant belonging to the Polygonaceae family. Traditionally, it is believed to possess effects such as nourishing blood, replenishing yin, and moistening the intestines to relieve constipation. Research indicates that the processing procedure significantly alters the chemical composition, pharmacological activity, and hepatotoxicity of substances; however, the underlying mechanisms responsible for attenuating toxicity and enhancing efficacy remain to be

Long-term chromium exposure and its residual impact on growth, biochemical changes and immune gene expression of climbing perch (Anabas testudineus).

Accumulation of heavy metal in fish not only causes morphological, biochemical, haematological and genetic changes but also poses serious health risks to their consumers. The present study investigated the effects of chromium (Cr) exposure on the alteration of physiological parameters and the expression pattern of three candidate genes: metallothionein 2 (MT-2), interleukin 1 beta (IL-1β) and tumour necrosis family 1 (TNF) in a climbing perch (Koi fish). The potassium dichromate (KCr2O7) was used as a source of Cr for fish feed and fed at three experimental doses (1, 2 and 5 ppm of Cr/kg feed)

Laparoscopic Extended Left Hepatectomy and Portal Vein Thrombectomy for Advanced Hepatocellular Carcinoma After Second-Line Conversion Therapy.

BACKGROUND: Hepatocellular carcinoma with portal vein tumor thrombus (HCC-PVTT) is correlated with poor prognosis.1 Recent advances in first-line therapies have enabled some patients with HCC-PVTT to become candidates for curative resection.2,3 However, over 50% of patients experience disease progression during initial treatment,4 and subsequent second-line options remain limited. Furthermore, reports on radical resection in patients with HCC-PVTT after second-line therapy are scarce. Herein, this case highlights the successful downstaging of HCC-PVTT with second-line therapy, followed by lapa

Temperature-Gradient Transcriptomic Atlas Reveals the PPAR Signaling Pathway Dysfunction in Heat Stroke Induced Liver Injury.

Heat stroke (HS) is the most severe form of hyperthermia, with mortality exceeding 50% in severe cases. The liver is highly vulnerable to HS-induced injury, often triggering multi-organ failure. Although rapid cooling remains the primary treatment, the molecular mechanisms underlying hepatic damage remain elusive, highlighting an urgent need for mechanistic insights, especially given global extreme heat events. We established a HS model by gradually increasing the core temperature of mice from 40°C to 43°C. Mice were sacrificed at each target temperature to collect blood and liver tissues for
