The Current Status of Herpesviridae as Major Human Pathogens: A 10-Year Diagnostic Evaluation in Germany.

Herpesvirus infections belong to major pathogens in the human population. This study aimed at evaluating diagnostic data for eight human herpesviruses, based on datasets derived from a large European tertiary care center. Specifically, we analyzed 118,692 herpesvirus submittals to the Diagnostic Division of the Virological Institute, University Hospital Erlangen (UKER), Germany, between July 2014 and June 2024. Our points of focus were the following: (i) the frequencies of herpesvirus diagnostic results with positivity rates, (ii) departments representing main sample submitters, (iii) the spec

Herpesvirus Diversity in Stranded Mysticetes of Brazil, Southwestern Atlantic.

Emerging and re-emerging pathogens have been increasingly investigated in cetaceans worldwide, particularly in odontocetes, whereas infectious diseases in mysticetes remain poorly understood. This study surveyed herpesvirus in 51 baleen whales stranded along the Brazilian coast, between 2008 and 2022: humpback whales (Megaptera novaeangliae, n = 46), Bryde's whales (Balaenoptera brydei, n = 3), a southern right whale (Eubalaena australis, n = 1), and a common minke whale (Balaenoptera acutorostrata, n = 1). A total of 159 tissue samples were analyzed using nested PCR assays targeting the herpe

The antiviral GTPase MxB is packaged into virions and binds via its N-terminal domain to alphaherpesvirus capsids.

The interferon-inducible myxovirus resistance proteins (Mx) belong to the dynamin-like GTPases; MxA is a restriction factor against several RNA viruses, while MxB restricts infections of lentiviruses and herpesviruses. Humans express MxA(1-662) with an N-terminal domain (NTD) of 43 residues, MxB(1-715) with an NTD of 91 residues, and a truncated MxB(26-715) with an NTD of 66 residues. Although the roles of the GTPase and stalk domains during infection are increasingly being elucidated, the functions of the different NTDs remain poorly understood. Using cell lines stably expressing Mx proteins,

Risk of Neurodevelopmental Disorders Following the Herpesviral Infection in Preschooler Children.

The associations between the herpesviral infections and neurodevelopmental disorders, including autism and attention deficit hyperactivity disorder (ADHD), remained unclear. Using the Taiwan National Health Insurance Research Database, the present study included 27,922 children < 6 years who were exposed to herpesviral infections between 1997 and 2011, as well as 111,688 age- and sex-matched children who were not exposed. All participants who developed autism and ADHD, as well as started to use ADHD medications during the follow-up (from enrollment to the end of 2013) were identified. Human he

Extensive necrotizing fasciitis in an immunocompetent neonate leading to rectovulvar and rectovaginal fistulae: A case report.

BACKGROUND: Necrotizing fasciitis (NF) is a rare but life-threatening soft tissue infection in neonates, associated with high morbidity and mortality. Perineal involvement in female neonates is particularly uncommon and may lead to devastating complications. CASE PRESENTATION: A 5-week-old previously healthy female infant presented with a 10-day history of a generalized rash that later localized to the perianal region and limbs, developing into ulceration. The condition evolved into extensive perineal necrosis complicated by rectovulvar and rectovaginal fistulae. Initial clinical features wer

The definition of response and inadequate response to topical corticosteroid treatment in atopic dermatitis and related skin inflammatory diseases: A GA2LEN ADCARE statement paper.

BACKGROUND: Topical corticosteroids (TCS) remain the first-line treatment for atopic dermatitis (AD) and related inflammatory skin diseases, yet no standardized definition of response exists. This gap contributes to heterogeneity in clinical practice and complicates trial design. We therefore aimed to develop consensus-based definitions of response and inadequate response to TCS therapy through a structured international eDelphi process. METHODS: A PubMed search (1974-July 2025) identified 403 relevant publications. Candidate statements were drafted from the evidence and refined by the ADCARE

Keratinocyte Priming by Staphylococcus aureus Reduces HSV-1 Susceptibility.

BACKGROUND: Individuals with atopic dermatitis (AD) are at increased risk for skin infections, including eczema herpeticum (EH), a severe condition caused by herpes simplex virus type 1 (HSV-1). While AD skin is often colonized by Staphylococcus aureus (S. aureus), its role in EH susceptibility remains unclear. Here we aim to investigate differences in the skin microbiome of AD patients with (ADEH+) and without (ADEH-) EH and examine the impact of S. aureus and S. epidermidis on HSV-1 infection in an in vitro keratinocyte model. METHODS: 16S microbiome sequencing was performed on skin samples

Severe Eczema Herpeticum With Ocular Involvement Leading to Orbital Cellulitis: A Pediatric Case Report.

Eczema herpeticum (EH) is a superimposed cutaneous viral infection that occurs in the context of preexisting dermatoses, most commonly atopic dermatitis (AD). It typically involves the face, neck, and upper trunk. Although ocular involvement is relatively uncommon, it may present diagnostic and therapeutic challenges and pose a significant risk to vision. We report a pediatric case of EH with severe ocular involvement in a young child with underlying AD. Despite early oral antiviral and antibiotic therapy, the condition progressed rapidly to orbital cellulitis. Clinical improvement was observe

NK Cell Disfunction in Atopic Dermatitis: A Missing Link Between Type 2 Inflammation, Microbial Dysbiosis and Antiviral Immunity.

Atopic dermatitis (AD) is a prevalent chronic inflammatory skin disorder driven by epidermal barrier defects and dysregulated Th2 cell responses. While therapies primarily address adaptive immunity, the role of Natural Killer (NK) cells remains underappreciated. This review analyzes NK cell dysfunctions in AD pathogenesis, evaluating their contributions to compromised skin immunity, microbial dysbiosis, and secondary infections. Accumulating evidence reveals a systemic deficiency of mature, cytotoxic CD56dim and NKp80+ NK cell subsets in peripheral blood, correlating with disease severity. Wit

The Use of CSF Multiplex PCR Panel in Patients with Viral Uveitis.

Background/Objectives: Polymerase chain reaction (PCR) testing of ocular fluids is an essential diagnostic method for identifying infectious causes of uveitis. However, multiplex PCR kits specifically developed for ophthalmic use are not commercially available in many regions, including Korea. Given the biochemical similarity between cerebrospinal fluid (CSF) and aqueous humor, this study evaluated the diagnostic utility of a commercially available CSF multiplex PCR panel for detecting herpesviruses in patients with suspected viral uveitis. Methods: We retrospectively reviewed the medical reco
