Mutational dissection of HCMV gB and gH cytoplasmic tails highlights conserved and divergent features of fusion regulation.

In herpesviruses, the fusogenic activity of the conserved glycoproteins gB and gH is regulated by their cytoplasmic (or intraviral) tails. However, prior to this work, their regulatory mechanisms had only been investigated in Alpha- and Gamma- but not in Betaherpesvirinae. Here, we developed a plasmid-transfection-based split-luciferase assay as a quantitative platform for measuring cell-cell fusion mediated by glycoproteins from human cytomegalovirus (HCMV), a member of Betaherpesvirinae. Using this assay, we confirmed that glycoproteins gB, gH, gL, and gO, along with the PDGFRα receptor, are

New Assay Systems to Characterize the Broad-Spectrum Antiherpesviral and Non-Herpesviral Activity of Cyclin-Dependent Kinase (CDK) 8 Inhibitors.

Background. To date, a number of human pathogenic viruses are still unaddressed by the current repertoire of approved antiviral drugs. In order to widen this spectrum of preventive measures against virus infections, we have focused on additional host targets that exert interesting virus-supportive functions. Inhibitors of cyclin-dependent kinase 8 (CDK8) have been found to exhibit highly pronounced and relatively broad antiviral activity. Objectives. The current research question concerning the potential for broad-spectrum antiviral drug activity should be addressed in detail to understand the

Toward Anti-Herpesviral PROTACs: Assessing the Challenges for Targeted Protein Degradation on the Example of Kaposi's Sarcoma-Associated Herpesvirus Latency-Associated Nuclear Antigen.

Kaposi's sarcoma-associated herpesvirus (KSHV) is a gamma-herpesvirus linked to several malignancies, including Kaposi's sarcoma, primary effusion lymphoma, and multicentric Castleman's disease. Among its numerous encoded proteins, the latency-associated nuclear antigen (LANA) plays a pivotal role in the maintenance of viral latency and oncogenesis. This manuscript focuses on therapeutic strategies aimed at targeting LANA to prevent KSHV-associated diseases. Following the concept of proteolysis-targeting chimeras (PROTACs), heterobifunctional compounds are designed and synthesized, which are a

ER protein CLCC1 promotes nuclear envelope fusion in herpesviral and host processes.

Herpesvirales are an ancient viral order that causes lifelong infections in species from mollusks to humans. They export their capsids from the nucleus to the cytoplasm by a noncanonical nuclear egress route that involves capsid budding at the inner nuclear membrane followed by fusion of this temporary envelope with the outer nuclear membrane. Here, using a whole-genome CRISPR screen, we identify ER protein CLCC1 as important for the fusion stage of nuclear egress in herpes simplex virus 1. We also find that the genomes of Herpesvirales that infect mollusks and fish encode CLCC1 genes acquired

Viral non-coding RNAs hijack host Pumilio proteins to regulate host transcripts.

Viral non-coding RNAs (ncRNAs) often perform multiple functions during persistent infection, but how they interface with host RNA-binding proteins remains incompletely understood. Herpesvirus saimiri (HVS), a T-tropic γ-herpesvirus that induces aggressive lymphomas in primates, during its latency expresses abundant small nuclear RNAs HSUR1 and HSUR2. HSUR1 is known to trigger target-directed microRNA decay (TDMD) of host miR-27a, and HSUR2 can function as a miRNA-dependent adaptor; here we show that both HSURs also bind host Pumilio proteins (PUM1 and PUM2) via perfectly conserved Pumilio resp

Atypical felid alphaherpesvirus 1 dermatitis in a cat.

Feline herpesviral ulcerative dermatitis is associated with ulceration of the face and oral mucosa. Histologically, lesions consist of eosinophilic inflammation in the superficial dermis, with occasional eosinophilic intranuclear viral inclusion bodies in the epidermis, follicular epithelium, and associated glands. We describe a case of atypical felid alphaherpesvirus 1 (FeAHV1; family Orthoherpesviridae, taxon species Varicellovirus felidalpha1) dermatitis in a 6-y-old, castrated male, domestic shorthair cat in which lesions were located on the limbs and shoulder without facial or oral involv

Extended poly(A) tails are a shared feature of herpesvirus mRNAs.

Poly(A) tails are present on most cellular and viral mRNAs, providing a platform for poly(A)-binding proteins that stimulate translation and regulate the deadenylation and stability of transcripts in the cytoplasm. Here we leverage nanopore direct RNA sequencing to analyse the distribution of poly(A) tail lengths on cellular and viral mRNAs across Herpesviridae and other DNA and RNA virus infections. We find that herpesvirus mRNA poly(A) tails are consistently longer than those on cellular and other viral transcripts, presenting a previously unrecognized yet widespread mechanism to advantage h

Genome-Wide Association Studies Data and Transcriptomics Data Link Herpes Simplex Virus 1 Infection and Parkinson's Disease.

OBJECTIVE: The main goal of this study is to explore the link between herpes simplex virus 1 (HSV-1) infection and Parkinson's disease (PD) from the genome-wide association studies (GWAS) data and find the shared molecular signature for mechanism understanding and drug repurposing from the transcriptomics data. METHODS: We used summary-level GWAS data for causal inference, exploring the association between herpes keratitis (mainly caused by HSV-1) and PD, and used transcriptomics data to study the shared molecular signature for mechanism understanding and drug repurposing. RESULTS: The causa

Recurrent severe viral infection in a child with inherited complete TBK1 deficiency.

TANK-binding kinase 1 (TBK1) acts at the crossroads of various host immune pathways. Autosomal dominant (AD) and recessive (AR) TBK1 deficiencies have been reported in human patients with herpes simplex encephalitis and SARS-CoV-2 pneumonia (AD deficiency) or systemic inflammation (AR deficiency). We describe here a Turkish boy, born to consanguineous parents and homozygous for a loss-of-function mutation of TBK1, who had experienced recurrent vesicular skin eruptions presumably triggered by herpesviral infection and five episodes of viral pneumonia since the age of 2 mo. Whole-exome sequencin

Kaposi Varicelliform Eruption Complicated by Steroids: A Case Report and Review of Literature.

Kaposi varicelliform eruption (KVE) is a severe disseminated cutaneous infection mainly caused by herpes simplex virus (HSV-1 or HSV-2). Herein, we report a case of KVE which was misdiagnosed elsewhere as a case of toxic epidermal necrolysis and received systemic steroids, following which, the patient's condition deteriorated. After being diagnosed as a case of KVE with the aid of tzanck smear, the patient was initiated on acyclovir and responded dramatically to the treatment. This case underscores the critical value of simple bedside tests, such as the tzanck smear, for rapid diagnosis of KVE

Woman with a Painful Rash.

CASE PRESENTATION: A 21-year-old woman with a history of eczema presented to the emergency department with a painful rash over the previous three days spreading from her left axilla to her left arm, left chest and left abdominal wall. The rash consisted of clusters of small, erythematous vesicles on hyperpigmented patches of skin. The patient was treated empirically with intravenous acyclovir for eczema herpeticum with improvement. Polymerase chain reaction testing of the fluid obtained from the rash vesicles later confirmed the presence of herpes simplex virus-1. DISCUSSION: Eczema herpeticu

The real-world, long-term risk of infections associated with dupilumab in atopic dermatitis: A global cohort study.

BACKGROUND: A low risk of infections was found in the randomized placebo-controlled trials of dupilumab in atopic dermatitis (AD). Dupilumab-associated real-life long-term risk of infections remains unclear. OBJECTIVES: To assess the risk of infectious complications in patients with AD managed by dupilumab relative to those treated with methotrexate and cyclosporine. METHODS: Using the TriNetX global dataset, a retrospective cohort study comprised two distinct analyses comparing patients with AD under different treatments: (i) initiators of dupilumab (n = 10,913) versus methotrexate (n = 10,

Collagen XXIII (COL23A1): A novel risk factor for eczema herpeticum.

BACKGROUND: Eczema herpeticum (EH) is a potentially life-threatening disseminated skin infection caused by herpes simplex virus (HSV) in a subset of patients with atopic dermatitis (AD). The occurrence of EH in a subset of patients with AD and its frequent recurrence imply the importance of genetic factors in its pathogenesis. OBJECTIVE: We sought to identify novel genetic risk factors for EH and to study their impact on HSV-1 infection. METHODS: Using whole-exome sequencing we identified a heterozygous single nucleotide polymorphism (SNP) in the COL23A1 gene (encoding collagen type XXIII al

Outpatient and Inpatient Management of Infants and Children With Eczema Herpeticum Diagnosed in the Emergency Department.

BACKGROUND: There are no universally established guidelines for when pediatric EH warrants hospitalization or intravenous (IV) acyclovir over oral therapy. To address this lack of consensus, this study aimed to describe outcomes of outpatient oral acyclovir treatment in pediatric EH cases, including rates of disease exacerbation, hospitalizations, and complications. METHODS: We conducted a retrospective cohort review of pediatric patients under age 7 diagnosed with EH who presented to the pediatric emergency department (ED) between January 1, 2008, and December 31, 2022. Cases were identified

Disproportionality Analysis of Nemolizumab in Patients with Atopic Dermatitis: A Real-World Pharmacovigilance Study.

Background: Nemolizumab is approved for the treatment of moderate-to-severe atopic dermatitis in patients aged ≥12 years, combined with topical corticosteroids and/or calcineurin inhibitors, when the disease is not adequately controlled by topical prescription therapies. Given the limited duration of follow-up in clinical trials and the current lack of postmarketing surveillance data, further investigations to evaluate the long-term safety profile of nemolizumab are urgently needed. Methods: Adverse event (AE) signals were identified using disproportionality analysis with four algorithms: the
