A live-cell medium-throughput assay for quantifying lipid accumulation in hepatocytes.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a global health burden contributing to the high prevalence of associated metabolic diseases such as cardiovascular disease and thus global mortality rates. MASLD is diagnosed by ≥ 5 % lipid accumulation in the liver, and candidate therapies are often evaluated for lipid lowering effects. Current cell-based lipid imaging approaches rely on fixed-cell staining with dyes such as BODIPY 493/503, limiting the ability to dynamically monitor lipid accumulation in living hepatocytes. To gain a better understanding of the effect of dif

Microvasculopathy in spinal muscular atrophy: The endothelium as a target of SMN deficiency and an amplifier of disease.

Spinal muscular atrophy (SMA) is caused by deficiency of the ubiquitous survival motor neuron (SMN) protein. Degeneration of anterior horn motor neurons defines the disease, but many organs are affected, including heart, liver, kidney, and bone. The vascular compartment is among them: digital necrosis in infants with the fewest SMN2 copies, capillary rarefaction in SMA mice, and decreased von Willebrand factor with fibrinogen extravasation in human post-mortem spinal cord. Whether these reflect endothelial injury by SMN deficiency itself or capillary adaptation to atrophic, denervated tissue h

Advances in assessment systems and non-pharmacological intervention strategies for cancer-related fatigue following transarterial chemoembolization in hepatocellular carcinoma: a narrative review.

PURPOSE: Transarterial chemoembolization (TACE) is a primary treatment for intermediate to advanced hepatocellular carcinoma (HCC); however, cancer-related fatigue (CRF) is a prevalent and distressing symptom following TACE that significantly impairs patients' recovery and health-related quality of life. This review aimed to systematically summarize the current evidence on CRF screening, assessment, and non-pharmacological interventions specifically tailored to the post-TACE HCC population, and to propose an integrated, nurse-led clinical management framework. METHODS: A narrative review was

PAK1 Promotes Gluconeogenesis Via the ACSS2/KAT2A/PCK1 Complex to Attenuate Steatotic Liver Ischemia-Reperfusion Injury.

Ischemia-reperfusion (IR) injury is particularly severe in steatotic livers, substantially increasing the risk of graft failure. The growing use of steatotic grafts amid donor shortages has therefore heightened the clinical importance of this problem. The conserved serine/threonine kinase p21-activated kinase 1 (PAK1) regulates cell migration, growth, and redox homeostasis, but its role in steatotic liver IR injury remains unclear. To address this question, we examined the association between PAK1 expression in human steatotic liver grafts and clinical outcomes in 74 liver transplantation (LT)

A naturally occurring polyphenolic compound for liver cancer treatment.

As a polyphenolic compound naturally found in nature, piceatannol is an analogue of resveratrol. These compounds share a common structural motif but have various substituents. Resveratrol shows anti-inflammation and anti-tumor properties, but such biological activities of the active resveratrol metabolite - piceatannol are unclear. In this study, the anti-tumor properties of piceatannol were investigated through cell viability, colony formation, migration assay, detection of reactive oxygen species (ROS) and mitochondrial membrane potential (MMP) change, reverse transcription quantitative poly

Evidence Suggests Linoleic Acid Exerts Health Benefits Beyond Cardiovascular Disease: A Brief Review of Clinical and Observational Studies.

PURPOSE OF REVIEW: Questions challenging the healthfulness of diets high in linoleic acid (LA) and seed oils, which are major sources of this essential fatty acid, have recently arisen. These questions are especially relevant to adherents of plant-based diets as their LA intake is higher than the general population. Some maintain that LA increases inflammation and oxidative stress and for these reasons, despite its ability to lower circulating LDL-cholesterol levels when replacing saturated fat, does not lower cardiovascular disease (CVD) risk. Because most of the discussion about LA has focus

Passively Targeted Polymeric Nanocarrier Delivering STAT1 siRNA to the Liver Ameliorates Steatohepatitis and Fibrosis in the Thioacetamide Mouse Model.

Liver fibrosis is a major complication of chronic liver injury, driven in part by Signal Transducer and Activator of Transcription 1 (Stat1). This study evaluated passively liver-targeted polymeric nanocarriers delivering Stat1 siRNA (siSTAT1) to treat steatohepatitis and fibrosis in a thioacetamide (TAA) mouse model. Two polyplex nanocarriers with different core-corona ratios, EN15 and EN76, were tested in TAA-treated mice and activated human and murine hepatocytes. In vitro, both formulations enabled efficient cellular uptake and Stat1 silencing, reducing TAA-induced lipid accumulation. Thro

Effect of conditions and diseases associated with systemic inflammation on the plasma concentration of growth differentiation factor 15 (GDF15) in dairy cows.

Plasma growth differentiation factor 15 (GDF15) was analyzed in dairy cows during the transition period, following treatment with the endotoxin lipopolysaccharide (LPS), during heat stress, and in the presence of infectious diseases. Events associated with these conditions include systemic inflammation and reduced appetite. GDF15 may be relevant to these conditions given its discovery as an overexpressed cDNA in an activated immune cell line and its emergence as an anorexic signal. An i.v. LPS bolus to early lactating cows increased plasma GDF15 from a baseline concentration of 45.2 ± 7.7 pg/m

Gut Microbiota-Derived Indole-3-propionic Acid Attenuates Titanium Dioxide Nanoparticle-Induced Liver Injury by Targeting Kupffer Cells via the Gut-Liver Axis.

Nanotitanium dioxide (nTiO2) is widely used as a food additive, leading to chronic dietary exposure and rising health concerns. Although adverse effects on the liver and gut microbiota have been individually investigated, whether gut microbiota mediates nTiO2-induced liver injury remains unclear, especially given the well-established role of the gut-liver axis in hepatic physiology and pathology. Herein, we explored the long-term hepatotoxic effects of oral nTiO2 in mice and the underlying mechanisms through the lens of gut-liver axis crosstalk. Our results showed that chronic exposure to nTiO

Intestinal Macrophage-Mediated Exosomal Repackaging Establishes Bioactive Reservoirs for Long-Acting Oral Peptide Therapy.

Oral peptide delivery systems are conventionally engineered to improve intestinal absorption, but they rarely address the progressive and systemic nature of chronic inflammatory disorders such as type-2 diabetes (T2D), which requires both durable glycemic control and immunometabolic management. Here, we report an oral delivery system based on porous calcium carbonate/sodium alginate (PCC/SA) microparticles loaded with exenatide-chitosan oligosaccharide (EXE-COS). The PCC/SA carrier facilitates the efficient intestinal absorption and transepithelial transport of EXE-COS. Notably, in the lamina

Dietary Supplementation of Auraptene, a Major Component in Citrus reticulata Blanco, Prevents Acute Liver Injury Associated with Apoptosis and Inflammation.

Auraptene (AP), a coumarin derivative abundant in citrus peels, exhibits diverse bioactivities; however, its role in acute liver injury and the underlying mechanisms remain elusive. This study investigates the hepatoprotective effects of AP against carbon tetrachloride (CCl4)-induced acute liver injury in mice and explored its mechanistic basis. In vitro, transforming growth factor-beta (TGF-β) was employed to stimulate AML-12 hepatocytes to model the injury microenvironment. The impact of AP on the toll-like receptor 2 (TLR2) signaling pathway was assessed through a combination of siRNA gene

Decoding the Biological Function of Bile Acids for Drug Discovery.

Bile acids are a class of cholesterol-derived metabolites that have long been recognized for their essential roles in dietary lipid absorption and cholesterol homeostasis. Over the past two decades, however, bile acids have emerged as crucial signaling molecules that regulate diverse physiological and pathological processes, including metabolism, inflammation, immunity, host-microbiota interactions, and liver diseases. Dysregulation of bile acid homeostasis has been implicated in a broad spectrum of human disorders, making bile acid signaling pathways attractive targets for therapeutic interve

Lipidomics Reveals Potential Mechanisms of Intestinal-Targeted Astaxanthin in Ameliorating Metabolic Dysfunction-Associated Steatotic Liver Disease in ApoE-/- Mice.

Metabolic dysfunction-associated steatotic liver disease (MASLD) represents a major global health burden with an urgent need for effective interventions. Although astaxanthin (AST) exhibits hepatoprotective potential, its efficacy is limited by poor oral bioavailability. This study evaluated an intestine-targeted astaxanthin delivery system (AST-LPs) in HFHC-diet-induced MASLD mice. AST-LPs administration exhibited superior efficacy over free AST in attenuating hepatic steatosis, improving lipid profiles and glucose tolerance, and suppressing inflammation. Lipidomics analysis revealed that AST

Mannose attenuates CCl4-induced liver fibrosis by reducing oxidative stress through AMPK-dependent NRF2 signaling.

Mannose, a naturally occurring hexose, has been implicated in the regulation of metabolism and inflammation. Although emerging evidence suggests its therapeutic potential in liver diseases, the mechanisms by which mannose modulates liver fibrosis, particularly in relation to oxidative stress, remain poorly defined. This study aimed to investigate the protective effects of mannose in liver fibrosis and elucidate the underlying mechanisms. A CCl4-induced mouse model of liver fibrosis was established with mannose administered in drinking water. Transcriptomic analysis was performed to explore pot

Chronic stress activates a hypothalamic vagal circuit to promote hepatocellular carcinoma progression in male mice.

Hepatocellular carcinoma and depression frequently co-occur, yet the mechanisms linking them remain unclear. Here, we show that chronic stress, systemic inflammation, or neuropathic pain are associated with hepatocellular carcinoma progression in male mice. Chronic stress hyperactivates a glutamatergic paraventricular hypothalamic nucleus → cholinergic dorsal motor nucleus of the vagus → liver circuit, increasing intratumoral acetylcholine. Single-cell transcriptomics identifies acetylcholine-responsive Chrna9+ epithelial cells that interact with macrophages through COL1A1/2-CD44-STAT3 signali

Patients with MASH experience worse health-related quality of life: a multinational real-world study using LiverMultiScan.

PURPOSE: Prevalence of metabolic dysfunction-associated steatotic liver disease (MASLD) and its progressive form, metabolic dysfunction-associated steatohepatitis (MASH), is increasing globally. Assessment of health-related quality of life (HRQoL), alongside non-invasive tests (NITs), may support identification of patients requiring closer clinical management. This multinational real-world randomised controlled trial evaluated associations between HRQoL (using the EQ-5D) and multiparametric MRI (mpMRI) markers of liver disease. METHODS: RADIcAL-1 included 802 patients (453 male) with suspecte

Semaglutide and Tirzepatide Suppress Hepatic Fatty Acid Uptake and Fatty Acid β-Oxidation in High-Fat Diet-Induced Obesity Mice.

Glucagon-like peptide-1 (GLP-1) and dual GLP-1/glucose-dependent insulinotropic polypeptide (GIP) receptor agonists show high efficacy in treating obesity and metabolic dysfunction-associated liver diseases, conditions that collectively represent one of the most challenging One Health crises of our time. However, their hepatic molecular mechanisms remain poorly understood. This study defined targeted hepatic proteomic signatures associated semaglutide (GLP-1 agonist) and tirzepatide (dual GLP-1/GIP) in diet-induced obese (DIO) male C57BL/6J mice using an liquid chromatography-tandem mass spect

Atomistic Mechanisms of Allosteric Dysfunction in Wilson Disease: How Local ATP7B Mutations Propagate to Global Destabilization.

Wilson disease (WD) is a severe metabolic disorder caused by mutations in the copper-transporting ATPase ATP7B. The MBD5-6 tandem module serves as a critical regulatory hub for the protein, harboring several pathogenic missense mutations, including T587M, A595T, and R616Q within the MBD6 domain. However, the atomistic mechanisms by which these localized defects propagate across the MBD5-6 tandem module to disrupt interdomain organization remain unclear. In this study, we employed AlphaFold 3 to generate initial structures of the wild-type and variant ATP7B, followed by three independent 500 ns

Attenuation of IL-8 expression following AHR inhibition in an in vitro model of intestinal inflammation.

BACKGROUND: The aryl hydrocarbon receptor (AHR) is an important regulator of intestinal homeostasis and has been linked to inflammatory bowel disease (IBD). However, its role in epithelial inflammatory responses is not fully understood. This study aimed to evaluate AHR signaling in in vitro models of intestinal inflammation and to investigate the effects of pharmacological AHR inhibition. METHODS: Two inflammatory models were evaluated: a differentiated Caco-2/THP-1 co-culture system and a TNF-α/LPS-stimulated Caco-2 epithelial model. AHR expression was measured by real-time PCR. The model sh
