Inflammatory Pseudotumour in Common Seal (Phoca vitulina): A Case Report.

Pinnipeds, including seals, sea lions and walruses, are marine mammals susceptible to a variety of pathological conditions. Although neoplastic lesions in these species are receiving increased attention, relatively little information is available compared to neoplastic studies in domestic animals. In this report, a 19-year-old intact male common seal (Phoca vitulina) presented with a chronic ulcer on the fifth phalanx. Radiological examination revealed cortical reaction with significant osseous alterations in this animal and in another companion common seal, although the flipper appeared macro

Pre-assembled complexes and allosteric effects: parallels between eukaryotic phosphorylation cascades and membrane fusion during herpesviral entry.

Unlike most enveloped viruses, Herpesviridae distribute cell entry functions across several viral envelope proteins. The prevailing model posits that, upon interaction with the target cell, the activating signal is transmitted from the receptor-binding to the fusion-mediating component in a signaling cascade that involves sequential interactions. However, herpesvirus entry proteins may form complexes throughout fusion. Here, we propose that-by analogy with certain eukaryotic signaling cascades-transmission of the activating signal involves pre-assembled complexes and allosteric effects.

Development and Validation of Quantitative PCR Assays for Strigid Alphaherpesvirus 1 and 2 in Great Horned Owls (Bubo virginianus).

Great Horned Owls (Bubo virginianus; GHOW) are a true owl species (family Strigidae) widely distributed across the Americas. In GHOWs, herpesviral disease is best known from fatal infections with columbid alphaherpesvirus 1, which is endemic in rock pigeons (Columba livia). Recently, two novel alphaherpesviruses, strigid alphaherpesvirus 1 (StrAHV1) and strigid alphaherpesvirus 2 (StrAHV2), have been identified in GHOWs displaying ocular lesions and upper respiratory signs, respectively. To date, these are the only potentially endemic herpesviruses described in this species. This study aimed t

UpRoi1 - the phage-derived microRNA-size non-coding RNA molecule regulating both phage and its bacterial host development.

The phage Φ24B-encoded small RNA (sRNA), named UpRoi1, was identified following the prophage induction. The UpRoi1 molecule is encoded in the antirepressor region of the phage genome and is notably small, consisting of only 30 nucleotides. This sRNA is comparable in size to microRNAs found in eukaryotic cells, and thus, unusual for prokaryotic sRNAs. Bioinformatic analysis revealed that UpRoi1 has multiple interaction sites within the phage and host genomes, suggesting its complex regulatory role. In turn, RNA-seq analyses confirmed that the UpRoi1 molecule has a broad impact on the host's gen

Potent Inhibitory Effects of Lauric Acid and Glycerol Monolaurate Against CyHV-2 and the Viral Envelope.

Lauric Acid (LA) and Glycerol Monolaurate (GML) are naturally present in several plant-derived oils, particularly coconut and palm kernel oil. These compounds have been widely applied in swine production owing to their proven antiviral activity against Porcine Reproductive and Respiratory Syndrome Virus (PRRSV), Porcine Epidemic Diarrhoea Virus (PEDV), and African Swine Fever Virus (ASFV). In this study, we investigated the potential of LA and GML as antiviral agents against Cyprinid herpesvirus 2 (CyHV-2), the causative agent of Herpesviral haematopoietic necrosis (HVHN), which poses signific

HAMMER: Hairpin-based APOBEC3A-mediated mRNA editing reporter.

APOBEC3A catalyzes cytosine-to-uracil deamination in single-stranded DNA and RNA. Physiologically, APOBEC3A functions in innate immunity and aberrant deamination is associated with cytosine mutations in enzymatically preferred YTCW substrate motifs in multiple cancers. Much less is known about the potential contribution of APOBEC3A-catalyzed RNA editing to virus and cancer evolution. Here, we present HAMMER (hairpin-based APOBEC3A-mediated mRNA editing reporter), a rapid luminescence-based cellular assay for measuring RNA editing by APOBEC3A. Attenuation mechanisms of the P7-P8 live-attenuated cyvirus cyprinidallo2 vaccine potentially involve apoptosis non-inhibition feature: insights into virus pathogenesis.

Cyvirus cyprinidallo2 (CyHV-2) is an alloherpesvirus and the causative agent of herpesviral haematopoietic necrosis in goldfish. Whole-genome sequence comparison of the developed live-attenuated vaccine P7-P8 with virulent CyHV-2 strains revealed seven single-nucleotide polymorphisms, five deletions and one inversion in the ORFs, which may be involved in attenuation. A start codon loss in ORF 113, a putative apoptosis-inhibition gene, was observed in the mutations. In vitro assays indicated that apoptosis-related genes were upregulated in cells inoculated with the vaccine or virulent virus com

Marek's disease virus UL16 tegument protein is essential for secondary envelopment and infectious virion formation.

Marek's disease virus (MDV) is a highly oncogenic alphaherpesvirus whose productive infection and cell-to-cell spread rely on the formation of infectious mature virions, a process mediated by tegument proteins. UL16 is a conserved herpesviral tegument protein, in other herpesviruses, its deletion causes severe replication defects. However, its role in MDV remains unknown. In this study, a UL16-null MDV mutant was generated using BAC-based reverse genetics to investigate UL16's function. Deletion of UL16 completely abrogated the recovery of infectious virus from BAC DNA in chicken embryonic fib

Single-cell transcriptome profiling reveals herpesviral manipulation of host processes in spleen of gibel carp infected with a Cyvirus cyprinidallo2.

The herpesviral haematopoietic necrosis with acute gill hemorrhage has recently become prevalent in gibel carp farming regions and has resulted in significant losses. Although several herpesvirus strains (belonging to Cyvirus cyprinidallo2) have been isolated, the permissive cells in vivo, how the virus manipulates host cell processes, and causes disease remain largely unclear. In this study, the isolate Carassius auratus herpesvirus (CaHV) was used as a model to dissect the interactions between virus and host through single-cell RNA sequencing (scRNA-seq) and functional validation. HSV-2 genome recognition by nuclear cGAS instigates IFN-β production and influences inflammasome activation during de novo infection in HFF cells.

INTRODUCTION: Type I interferon response, specifically, the cGAS-cGAMP-STING axis that results in IFN-β response, is well known for its complex roles early during viral infection. Previous reports suggest that HSV-1 DNA in Thp-1 cells and HIV-2 dsDNA in DCs and macrophages could be sensed by cGAS. The nuclear DNA sensor IFI16's viral DNA sensing leads to its acetylation, cytoplasmic translocation and STING activation and inflammasome activation. Although cGAS is known to be associated with IFI16 in the nucleus, however, during HSV-2 infection, the role of nuclear cGAS in viral DNA sensing, inf

Cyprinid herpesvirus 2 (CyHV-2) ORF53 inhibits NF-κB signalling by targeting p65 degradation.

Cyprinid herpesvirus 2 (CyHV-2) is the pathogen responsible for herpesviral disease in cyprinid fish, inflicting substantial economic losses on the carp aquaculture industry. At present, the immune evasion mechanism of CyHV-2 has not been fully elucidated. In the present study, the open reading frame 53 (ORF53) of CyHV-2 was functionally screened as a negative regulator of the NF-κB signalling pathway from 19 predicted non-membrane protein ORFs. Overexpression of ORF53 suppressed the activation of NF-κB promoter mediated by CyHV-2 infection and poly(dG:dC) stimulation, as well as by overexpres

Crystal structure of Bcl-2 from lymphocystis disease virus 2 in complex with the BH3 domain of zebrafish BaxA.

Lymphocystis disease viruses (LCDVs), members of the Lymphocystivirus genus of the Iridoviridae family, infect various freshwater and marine fish species. They cause the chronic disease lymphocystis, which is non-fatal, but substantially reduces the commercial value of the infected fish. To date, four genotypes of LCDV (LCDV1-4) have been identified, all of which encode the viral homologue of B-cell lymphoma 2 (Bcl-2), a key inhibitor of apoptosis. In this study, we performed biochemical and structural analyses of LCDV2 Bcl-2. Binding assays revealed that LCDV2 Bcl-2 exhibits binding selectivi

Impact of COVID-19 pandemic on viral keratitis.

COVID-19 has altered the epidemiology and clinical course of viral keratitis, with increasing evidence of herpesvirus reactivation after infection or vaccination. HSV keratitis remains most common in younger-to-middle-aged men, while HZO predominates in older adults and around one-fourth of HZO patients report current or prior COVID-19. HSV cases frequently recur and complications such as corneal opacity and neuropathic pain can cause vision outcomes ranging from normal to severe loss. Large recent studies suggest vaccination is associated with higher risk of new-onset and relapsing herpesvira

Extended poly(A) tails are a shared feature of herpesvirus mRNAs.

Poly(A) tails are present on most cellular and viral mRNAs, providing a platform for poly(A)-binding proteins that stimulate translation and regulate the deadenylation and stability of transcripts in the cytoplasm. Here we leverage nanopore direct RNA sequencing to analyse the distribution of poly(A) tail lengths on cellular and viral mRNAs across Herpesviridae and other DNA and RNA virus infections. We find that herpesvirus mRNA poly(A) tails are consistently longer than those on cellular and other viral transcripts, presenting a previously unrecognized yet widespread mechanism to potentially

The Current Status of Herpesviridae as Major Human Pathogens: A 10-Year Diagnostic Evaluation in Germany.

Herpesvirus infections belong to major pathogens in the human population. This study aimed at evaluating diagnostic data for eight human herpesviruses, based on datasets derived from a large European tertiary care center. Specifically, we analyzed 118,692 herpesvirus submittals to the Diagnostic Division of the Virological Institute, University Hospital Erlangen (UKER), Germany, between July 2014 and June 2024. Our points of focus were the following: (i) the frequencies of herpesvirus diagnostic results with positivity rates, (ii) departments representing main sample submitters, (iii) the spec

The antiviral GTPase MxB is packaged into virions and binds via its N-terminal domain to alphaherpesvirus capsids.

The interferon-inducible myxovirus resistance proteins (Mx) belong to the dynamin-like GTPases; MxA is a restriction factor against several RNA viruses, while MxB restricts infections of lentiviruses and herpesviruses. Humans express MxA(1-662) with an N-terminal domain (NTD) of 43 residues, MxB(1-715) with an NTD of 91 residues, and a truncated MxB(26-715) with an NTD of 66 residues. Although the roles of the GTPase and stalk domains during infection are increasingly being elucidated, the functions of the different NTDs remain poorly understood. Using cell lines stably expressing Mx proteins,
