First report of systemic round cell tumor, suspected lymphoma, in a wild Amami rabbit (Pentalagus furnessi) from Tokunoshima Island.

This study reports the first documented case of neoplasia in a wild Amami rabbit, an endangered species endemic to the Amami-Oshima and Tokunoshima Islands in Japan. In January 2018, a deceased Amami rabbit was found on a rural road on Tokunoshima Island. Necropsy revealed hemorrhage, cervical vertebral dislocation, generalized lymphadenopathy, a 5 × 5 cm mediastinal mass encasing the heart, and splenomegaly. The rabbit's death was presumed to be due to vehicle collisions following emaciation from systemic tumor progression. Histopathological examination showed significant proliferation of tum

Mutational dissection of HCMV gB and gH cytoplasmic tails highlights conserved and divergent features of fusion regulation.

In herpesviruses, the fusogenic activity of the conserved glycoproteins gB and gH is regulated by their cytoplasmic (or intraviral) tails. However, prior to this work, their regulatory mechanisms had only been investigated in Alpha- and Gamma- but not in Betaherpesvirinae. Here, we developed a plasmid-transfection-based split-luciferase assay as a quantitative platform for measuring cell-cell fusion mediated by glycoproteins from human cytomegalovirus (HCMV), a member of Betaherpesvirinae. Using this assay, we confirmed that glycoproteins gB, gH, gL, and gO, along with the PDGFRα receptor, are

New Assay Systems to Characterize the Broad-Spectrum Antiherpesviral and Non-Herpesviral Activity of Cyclin-Dependent Kinase (CDK) 8 Inhibitors.

Background. To date, a number of human pathogenic viruses are still unaddressed by the current repertoire of approved antiviral drugs. In order to widen this spectrum of preventive measures against virus infections, we have focused on additional host targets that exert interesting virus-supportive functions. Inhibitors of cyclin-dependent kinase 8 (CDK8) have been found to exhibit highly pronounced and relatively broad antiviral activity. Objectives. The current research question concerning the potential for broad-spectrum antiviral drug activity should be addressed in detail to understand the

Toward Anti-Herpesviral PROTACs: Assessing the Challenges for Targeted Protein Degradation on the Example of Kaposi's Sarcoma-Associated Herpesvirus Latency-Associated Nuclear Antigen.

Kaposi's sarcoma-associated herpesvirus (KSHV) is a gamma-herpesvirus linked to several malignancies, including Kaposi's sarcoma, primary effusion lymphoma, and multicentric Castleman's disease. Among its numerous encoded proteins, the latency-associated nuclear antigen (LANA) plays a pivotal role in the maintenance of viral latency and oncogenesis. This manuscript focuses on therapeutic strategies aimed at targeting LANA to prevent KSHV-associated diseases. Following the concept of proteolysis-targeting chimeras (PROTACs), heterobifunctional compounds are designed and synthesized, which are a

ER protein CLCC1 promotes nuclear envelope fusion in herpesviral and host processes.

Herpesvirales are an ancient viral order that causes lifelong infections in species from mollusks to humans. They export their capsids from the nucleus to the cytoplasm by a noncanonical nuclear egress route that involves capsid budding at the inner nuclear membrane followed by fusion of this temporary envelope with the outer nuclear membrane. Here, using a whole-genome CRISPR screen, we identify ER protein CLCC1 as important for the fusion stage of nuclear egress in herpes simplex virus 1. We also find that the genomes of Herpesvirales that infect mollusks and fish encode CLCC1 genes acquired

Viral non-coding RNAs hijack host Pumilio proteins to regulate host transcripts.

Viral non-coding RNAs (ncRNAs) often perform multiple functions during persistent infection, but how they interface with host RNA-binding proteins remains incompletely understood. Herpesvirus saimiri (HVS), a T-tropic γ-herpesvirus that induces aggressive lymphomas in primates, during its latency expresses abundant small nuclear RNAs HSUR1 and HSUR2. HSUR1 is known to trigger target-directed microRNA decay (TDMD) of host miR-27a, and HSUR2 can function as a miRNA-dependent adaptor; here we show that both HSURs also bind host Pumilio proteins (PUM1 and PUM2) via perfectly conserved Pumilio resp

Atypical felid alphaherpesvirus 1 dermatitis in a cat.

Feline herpesviral ulcerative dermatitis is associated with ulceration of the face and oral mucosa. Histologically, lesions consist of eosinophilic inflammation in the superficial dermis, with occasional eosinophilic intranuclear viral inclusion bodies in the epidermis, follicular epithelium, and associated glands. We describe a case of atypical felid alphaherpesvirus 1 (FeAHV1; family Orthoherpesviridae, taxon species Varicellovirus felidalpha1) dermatitis in a 6-y-old, castrated male, domestic shorthair cat in which lesions were located on the limbs and shoulder without facial or oral involv

Extended poly(A) tails are a shared feature of herpesvirus mRNAs.

Poly(A) tails are present on most cellular and viral mRNAs, providing a platform for poly(A)-binding proteins that stimulate translation and regulate the deadenylation and stability of transcripts in the cytoplasm. Here we leverage nanopore direct RNA sequencing to analyse the distribution of poly(A) tail lengths on cellular and viral mRNAs across Herpesviridae and other DNA and RNA virus infections. We find that herpesvirus mRNA poly(A) tails are consistently longer than those on cellular and other viral transcripts, presenting a previously unrecognized yet widespread mechanism to advantage h

Recurrent severe viral infection in a child with inherited complete TBK1 deficiency.

TANK-binding kinase 1 (TBK1) acts at the crossroads of various host immune pathways. Autosomal dominant (AD) and recessive (AR) TBK1 deficiencies have been reported in human patients with herpes simplex encephalitis and SARS-CoV-2 pneumonia (AD deficiency) or systemic inflammation (AR deficiency). We describe here a Turkish boy, born to consanguineous parents and homozygous for a loss-of-function mutation of TBK1, who had experienced recurrent vesicular skin eruptions presumably triggered by herpesviral infection and five episodes of viral pneumonia since the age of 2 mo. Whole-exome sequencin

Bovine herpesviral meningoencephalitis: large case study and literature review.

Bovine herpesviral meningoencephalitis (BHM) is an infectious disease of cattle caused by bovine alphaherpesvirus 1 (BoAHV1; Orthoherpesviridae, Varicellovirus bovinealpha1) or 5 (BoAHV5; Orthoherpesviridae, Varicellovirus bovinealpha5). Cases are frequent in South America, but the disease is sporadic in other countries. Infection typically leads to fatal necrotizing meningoencephalitis that most often affects calves <1-y-old and yearlings under stressful situations, including weaning, large concentrations of animals, transport, and introduction of cattle from other locations into a herd. Clinic-radiological classification of herpesviral encephalitis in humans (systematic review).

The development of a comprehensive classification for herpesvirus encephalitis remains an urgent task. Distinct clinic-radiological forms of herpesvirus cerebral lesions have been characterized, including findings from histopathological studies. Differences among these forms have been demonstrated concerning key clinical and paraclinical parameters. The presented classification identifies several distinct forms of herpesvirus encephalitis: temporal, brainstem, limbic, diencephalic encephalitis, rhombencephalitis, leukoencephalitis, ventriculoencephalitis, diffuse glial micronodular encephaliti

Neonatal hearing screening among high-risk newborns in Northwestern Nigeria.

BACKGROUND: Neonatal Intensive Care Unit (NICU) patients are at an increased risk of developing hearing loss. Given the critical role of normal hearing in speech and language development, early detection and treatment of this condition in children are paramount. METHODS: Hearing assessments were performed using transient evoked otoacoustic emission (TEOAE) and automated auditory brain stem response (AABR) tests on 60 neonates. The resulting data were analysed using R version 4.3.3. RESULTS: Out of the 60 neonates enrolled in the study, 57% were males and 43% were females, and their ages rang

Virome profiling of long-tailed marmots reveals tissue tropism and cross-species transmission of a diversity of uncharacterized mammalian viruses.

BACKGROUND: Anthropogenic land conversion and population encroachment into wildlife habitats are amplifying zoonotic risks through intensified human-livestock-wildlife contacts. As ecologically resilient rodents exhibiting dynamic population fluxes, long-tailed marmots (Marmota caudata) are crucial viral reservoirs in Central Asian ecosystems. Their sympatric coexistence with domestic herds creates sustained spillover risks, yet comprehensive virome characterization remains limited. This study employs a whole-virome profiling strategy to comprehensively characterize the viral diversity of 101

An Atypical Presentation of Enteroviral Meningitis in an Immunocompetent Adult Man.

Aseptic meningitis is inflammation of the meninges characterized by cerebrospinal fluid (CSF) pleocytosis and negative bacterial cultures. Viral infections are the most common cause, with enteroviruses (EV) accounting for the majority of cases worldwide. Typical presentations include fever, headache, and meningeal signs; however, atypical cases can occur, leading to diagnostic uncertainty. Early recognition and lumbar puncture (LP) are essential to avoid misdiagnosis. This case highlights an unusual presentation of enteroviral meningitis without fever or meningeal signs in an immunocompetent a

Diseases of the nervous system of equids in Brazil: a review.

In Brazil, CNS diseases in equids were little known until the 1980s. Since then, several diagnostic laboratories have been operating in different universities, initially in the South and Southeast regions and, later, in the Central-West, Northeast and North regions. However, the knowledge accumulated from the diagnoses of nervous system diseases of equids made at these institutions over the years has not been reviewed comprehensively, and many papers are published in Portuguese. Here, we review 18 diseases that occur in the nervous system of equids in Brazil, including some critical infectious

Herpesviruses mimic zygotic genome activation to promote viral replication.

Zygotic genome activation (ZGA) is crucial for maternal to zygotic transition at the 2-8-cell stage in order to overcome silencing of genes and enable transcription from the zygotic genome. In humans, ZGA is induced by DUX4, a pioneer factor that drives expression of downstream germline-specific genes and retroelements. Here we show that herpesviruses from all subfamilies, papillomaviruses and Merkel cell polyomavirus actively induce DUX4 expression to promote viral transcription and replication. Analysis of single-cell sequencing data sets from patients shows that viral DUX4 activation is of

Duck plague virus ICP27 protein suppresses IFN-β production by dual targeting of DNA- and RNA-sensing pathways.

Duck plague virus (DPV), an alphaherpesvirus causing severe economic losses in global waterfowl industries, adopts sophisticated strategies to subvert host antiviral immunity. Here, we identify DPV ICP27 as a pivotal immune evasion protein that concurrently inhibits both DNA (cGAS-STING) and RNA (RIG-I/MDA5-MAVS) innate immune sensing pathways-a novel function unreported in avian herpesviruses. Through co-transfection and infection assays in duck embryo fibroblasts (DEFs), we demonstrate that ICP27 suppresses key immune sensors' transcriptional and protein expression levels (STING, RIG-I) and

Duck Plague Virus Full-Length UL15 Protein Is a Multifunctional Enzyme Which Not Only Possesses Nuclease Activity but Also Exerts ATPase and DNA-Binding Activity.

The genome of the herpesvirus is a linear double-stranded DNA. The viral genome replicates in the host cell to form a concatemeric DNA, which is then cleaved to produce a unit-length genome. This unit-length genome is packaged into procapsid to produce mature virus particles. The terminase large subunit, pUL15, mediates the cleavage and packaging of viral concatemeric genomes. Duck plague virus (DPV) is a member of the α herpesvirus subfamily. Previous studies have demonstrated that the C-terminal region of DPV pUL15 exhibits non-sequence-specific DNA cleavage activity in vitro, but the charac

A targeted mutation in ICP8 attenuates duck plague virus by disrupting replication and proposes a vaccine strategy.

Controlling duck plague (DP) is essential for waterfowl health and industry productivity. The herpesviral single-stranded DNA-binding protein ICP8 is essential for replication compartment assembly, yet its exploitation for vaccine development in avian herpesviruses is limited. In this study, we generated two ICP8 mutants (H825A and F829A/P830A) in duck plague virus (DPV) via BAC-based homologous recombination. Using structural modeling, in vitro replication kinetics, co-immunoprecipitation, and in vivo duckling models, we assessed impacts on replication complex assembly, virulence attenuation,
