Hypomethylation of GNA15 Promotes Pancreatic Ductal Adenocarcinoma Progression and Macrophage M2 Polarization via STAT3-CXCL8 Axis.

The complexity of pancreatic ductal adenocarcinoma (PDAC) progression, coupled with the lack of effective immunotherapy, underscores the imperative to deepen our understanding of its mechanisms and identify suitable immune-targeted interventions. The G protein alpha subunit 15 (GNA15) is significantly upregulated in PDAC and correlates with poor prognosis in patients with PDAC. Mechanistically, our study demonstrates that TET3 upregulates GNA15 expression through demethylation in PCs, and the highly expressed GNA15 activates the phosphorylation of STAT3 via the GP130-JAK signaling pathway, the

High-Dimensional SERS Mapping of Single Circulating Tumor Cells Deciphers the Phenotypic Landscape and Stratifies Metastatic Risk in Pancreatic Cancer.

The profound phenotypic heterogeneity of circulating tumor cells (CTCs) presents a major analytical challenge, demanding technologies capable of high-plex, quantitative single-cell profiling. Here, we introduce PRISM (Phenotypic Resolution via Immuno-SERS Mapping), a biosensing platform engineered to meet this challenge. The platform integrates a high-efficiency dual-antibody (anti-EpCAM/anti-CSV) capture substrate with a suite of seven spectrally orthogonal SERS nanoprobes, enabling crosstalk-free, multiplexed quantification of epithelial, mesenchymal, and stem-like (E-M-S) markers. Metabolic Analyses of Crosstalk between Pancreatic Stromal and Cancer Cells.

Metabolic crosstalk between cell types is critical to our mechanistic understanding of pancreatic cancer. The lack of methods to investigate metabolic crosstalk has impeded the comprehensive OMIC characterization of these complex cellular systems. To address this need, an NMR metabolomics method employing a selective isotope-labeling strategy is presented to study metabolic crosstalk in routine cell cultures. 2D 1H-13C HSQC NMR experiments were used to simultaneously monitor pancreatic cancer cells and stromal cells, which revealed a cancer-cell-induced metabolic remodeling of stromal cells. Analysis of factors influencing the complications of severe acute pancreatitis combined with hemorrhage.

BACKGROUND: Complications of hemorrhage in patients with severe acute pancreatitis (SAP) significantly impact prognosis. However, current research on this topic remains limited. This study aims to analyze the factors influencing hemorrhage in SAP patients. METHODS: This study retrospectively analyzed clinical data from SAP patients admitted to our hospital between January 2019 and December 2022. Using propensity score matching and multivariate logistic regression analysis, we identified independent factors influencing hemorrhage. Additionally, we developed a nomogram prediction model for blee

Oncogenic KRAS Regulates Secretion of Extracellular Vesicles and Surface Membrane Charge via Regulation of Phosphatidylserine in Pancreatic Cancer.

Extracellular vesicles (EVs) have the potential to be used as a liquid biopsy for cancer detection and treatment response assessment. Although the potential of EVs as disease-specific biomarkers has been promising, the rapid and specific enrichment of EVs from body fluids in a clinical setting remains challenging. To address this limitation, we developed a microfluidic electrophoresis (MEP) device for the label-free, charge-based enrichment of EVs from the serum of patients with pancreatic cancer (PaCa). We first validated the principle of charge-based cancer EV enrichment using cell-line-deri

Survival impact of involvement of lymph node stations 16b1, 9, and 8a during pancreatoduodenectomy for pancreatic and periampullary cancers (PANODE): prospective multicentre study.

BACKGROUND: Current guidelines do not recommend routine sampling of lymph node stations (LNS) 16b1, 9, and 8a during pancreatoduodenectomy for pancreatic ductal adenocarcinoma (PDAC) and other periampullary cancers because prospective multicentre studies are lacking. This study evaluated the association between the involvement of these LNS and overall survival (OS). METHODS: This prospective multicentre study included patients undergoing pancreatoduodenectomy for PDAC, distal cholangiocarcinoma, ampullary cancer, or duodenal cancer between October 2015 and April 2020. Prognostic value of microRNA expression in pancreatic ductal adenocarcinoma: a systematic review and meta-analysis.

BACKGROUND: Pancreatic ductal adenocarcinoma (PDAC) ranks among the deadliest malignancies, with limited survival benefits despite recent therapeutic advances. Few prognostic biomarkers are available for PDAC monitoring in clinical practice. We conducted a meta-analysis to evaluate and synthesise evidence on the prognostic significance of miRNA expression in PDAC. METHODS: The protocol was registered with PROSPERO (CRD42023463542). We conducted a systematic search in PubMed, CENTRAL, and Embase on January 27, 2026, to identify studies assessing miRNA expression levels in patients with PDAC wi

Rapid Single-Cell N-glycosylation Profiling of Circulating Human Immune Cells by Mass Spectrometry Imaging.

The metabolic fitness of immune cells, reflected in part through glycosylation of surface proteins, is a key determinant of their activation, persistence, and effector responses. A recently developed high-throughput antibody capture cell array platform was further optimized to profile N-glycosylation across bulk cell and single-cell populations of peripheral blood mononuclear cells (PBMCs). Using antibodies against lymphocyte, monocyte, and dendritic cell markers (CD4+, CD8+, CD11c+, CD14,+ CD16+, CD3+, CD19+, CD56+, CD68+), N-glycosylation patterns of these immune cell subtypes were mapped ac

Targeted Gastrointestinal Delivery of mRNA Lipid Nanoparticles Enables Systemic Vaccination and Glucagon-like Peptide-1 Metabolic Therapy.

Lipid nanoparticles (LNPs) administered parenterally often show poor localization to multiple tissue types, including the pancreas and gastrointestinal (GI) tract. Furthermore, patients typically prefer orally administered drugs to those that are given parenterally. We therefore investigated whether GI LNP administration to buccal, gastric, small intestinal, colonic, and rectal submucosal tissues could simultaneously circumvent parenteral administration while enhancing LNP delivery to previously inaccessible locations. We found that, in addition to local targeting, gastric LNP administration a

Biomimetic Nanoplatform for Fibrosis Remodeling and Anti-PD-1 Potentiation via Cu+/Ca2+ Ion Dysregulation and TGF-β Inhibition in PDAC.

Pancreatic ductal adenocarcinoma (PDAC) remains refractory to immunotherapy, largely due to the dense fibrotic stroma that promotes immune evasion and restricts drug penetration. Here, we report a multifunctional biomimetic nanoplatform (RPm@CC-LY) that simultaneously remodels the fibrotic tumor microenvironment and potentiates antitumor immunity. RPm@CC-LY achieves efficient tumor accumulation and effectively attenuates fibrosis through sustained release of Transforming growth factor-β (TGF-β) inhibitor LY2157299 (LY). Concurrently, intracellular acid-triggered degradation induces Cu+-mediate

Asia-Pacific Transplant Oncology Symposium 2024: Expert Statements on the Current State and Future Direction of Hepatobiliary Surgical and Transplant Oncology.

BACKGROUND: Hepatobiliary oncology is at an inflection point. Parallel advances in surgical technique, locoregional therapy, systemic treatment, and understanding of tumour biology have expanded the boundaries of curative-intent care yet have also exposed a critical reality that technical resectability is not synonymous with oncologic resectability. Technically successful resection or liver transplantation may be futile when tumour biology is aggressive, leading to early recurrence and limited survival benefit. Modern hepatobiliary surgical and transplant oncology must therefore move beyond an

Is it possible to predict resectability in pancreatic cancers? A retrospective study in a tertiary center.

OBJECTIVES: Pre-operative radiological or laboratory evaluations can provide some insight into resectability in pancreatic ductal adenocarcinoma (PDAC). However, these methods still fall short, as patients may be deemed unresectable during perioperative exploration. Our study seeks to identify novel predictors that can accurately determine the resectability status of PDAC patients. METHODS: Patients who underwent surgical exploration for potentially resectable PDAC were retrospectively analyzed and categorized into two groups: resectable and unresectable during exploration. Univariate and mul

Preoperative Frailty and Risk of Post-ERCP Pancreatitis in Older Adults with Choledocholithiasis.

BACKGROUND: Frailty is increasingly considered when evaluating surgical risk in older adults. However, evidence regarding whether frailty contributes to the risk of post-endoscopic retrograde cholangiopancreatography (ERCP) pancreatitis in elderly patients is limited. OBJECTIVE: We aimed to evaluate whether preoperative frailty was associated with the development of post-ERCP pancreatitis (PEP) in elderly patients undergoing ERCP. METHODS: Patients aged 65 years or older who underwent ERCP for choledocholithiasis were retrospectively included. Frailty was assessed preoperatively using an ele

Pancreatic head solid-pseudopapillary neoplasm in a man treated by robot-assisted laparoscopic partial pancreatectomy: a case report.

BACKGROUND: Solid-pseudopapillary neoplasm (SPN) of the pancreas is a rare tumor with low malignant potential, most commonly occurring in the pancreatic body or tail of young women. Atypical presentations, particularly in male patients and cases involving the pancreatic head, may pose diagnostic and therapeutic challenges. This report highlights surgical management considerations in a patient with inconclusive preoperative findings. CASE PRESENTATION: We report the case of a 37-year-old man with a large lesion initially interpreted as a retroperitoneal mass closely associated with the pancrea

Navigating the Pancreatic Duct: Up-to-Date Radiologic Expedition through Intraluminal Pathologic Entities.

Several congenital, traumatic, infectious, inflammatory, and neoplastic processes, as well as iatrogenic interventions, can alter intraluminal contents of the pancreatic duct, leading to adverse outcomes. Variant ductal anatomy may result in clinically significant complications such as duodenal obstruction, acute and chronic pancreatitis, and increased risk of developing gallbladder or biliary malignancies. The presence of calculi, blood, pus, and other unintended iatrogenic material in the pancreatic duct can be associated with a range of complications. Neoplastic involvement of the pancreati

HMGA2 Stabilizes hnRNPK to Drive YAP-Dependent Liver Metastasis in Pancreatic Cancer.

Pancreatic cancer (PC) is a highly aggressive malignant neoplasm with a propensity for liver metastasis (LM), which significantly contributes to poor patient prognosis, but its molecular mechanism remains incompletely elucidated. In this study, we demonstrated that high mobility group protein A2 (HMGA2) is markedly elevated in LM tissues and circulating tumor cells (CTCs) of PC patients. Mechanistically, HMGA2 binds to heterogeneous nuclear ribonucleoprotein K (hnRNPK) via "AT-hook" functional motif, suppressing its ubiquitination to block proteasomal degradation. This interaction promotes the

Design, Synthesis, and Biological Evaluation of Novel, Fluorinated Camptothecin Derivatives as Potential Topoisomerase I Inhibitors.

Camptothecin (CPT), a classical topoisomerase I (TOP I) inhibitor, shows potent antitumor activity but is limited by toxicity and lactone instability. Herein, structural optimization of the CPT scaffold was conducted via A-ring fluorination and C-7 and C-17 modifications. A series of fluorinated derivatives were synthesized and evaluated for antiproliferative activity. Structure-activity relationship studies identified C-11 fluorination as optimal, whereas C-7 ethyl and C-17 thiocarbonyl substitutions enhanced potency. Compounds W3 (11F-CPT) and W12 (7Et-11F-CPT) exhibited potential inhibitory
