Prognostic significance of reduced handgrip strength in patients with unresectable hepatocellular carcinoma receiving HAIC combined with targeted immunotherapy.

BACKGROUND: Hepatic arterial infusion chemotherapy (HAIC) combined with targeted immunotherapy has emerged as a key therapeutic option for advanced (or unresectable) hepatocellular carcinoma (HCC). Nevertheless, treatment efficacy varies significantly among individuals. Sarcopenia, characterized by loss of muscle mass and strength, may adversely affect therapeutic outcomes and patient prognosis. This study investigates the clinical relevance of sarcopenia in patients undergoing HAIC combined with targeted immunotherapy. METHODS: A total of 265 patients with unresectable HCC who received HAIC

Cytological Features of Pancreatic Medullary Carcinoma Diagnosed by Endoscopic Ultrasound-Guided Fine Needle Aspiration.

Pancreatic medullary carcinoma is an extremely rare tumor classified as a histological subtype of pancreatic ductal adenocarcinoma. We report a case of a pancreatic medullary carcinoma and describe its cytological features. A 75-year-old woman presented with lower back pain, epigastric discomfort, and a tumor-like lesion in the pancreatic body. Cytologic specimens obtained via endoscopic ultrasound-guided fine-needle aspiration revealed relatively uniform small round cells with a high nuclear-to-cytoplasmic ratio, appearing as isolated scattered cells or loosely aggregated small clusters in a

Puerarin-loaded injectable hydrogel mitigates skeletal muscle wasting in colorectal cancer cachexia by targeting inflammatory signaling and improving protein homeostasis.

Cancer cachexia (CC) is a complex, tumor-induced metabolic syndrome characterized by progressive skeletal muscle atrophy, systemic inflammation, and refractoriness to nutritional intervention, contributing to functional decline and increased mortality. Effective targeting of CC-induced skeletal muscle atrophy are major challenges in clinical treatment. Systemic administration of puerarin (PUE) is limited by poor oral bioavailability, rapid clearance, and lack of tissue-targeting capability, resulting in suboptimal therapeutic concentrations at the affected muscle tissue. This study aimed to co

Inhibition of serine racemase prevents retinopathy in diabetic mice.

A single-nucleotide polymorphism in the serine racemase (SRR)-encoding gene, Srr, is associated with an increased risk of type 2 diabetes. Studies, including our own, demonstrate that SRR is linked to a complication of diabetes, diabetic retinopathy (DR), and deletion of Srr protects diabetic animals from developing retinopathy. We investigate the feasibility and mechanisms of SRR inhibition as a strategy to prevent DR. SRR proteins in the retinas of db/db mice were approximately 50 % higher than C57BLKS/J wild-type (WT) mice, while D-serine levels in the aqueous humor were more than two-fold

DCAF1 promotes the growth and metastasis of pancreatic cancer cells by activating the PTEN/PI3K/Akt signaling pathway.

Pancreatic cancer is a common malignant tumor of the digestive tract characterized by high mortality, primarily due to its aggressive proliferation and metastasis. DDB1-CUL4-associated factor 1 (DCAF1) has recently been identified as a cancer ‒ promoting gene in hepatocellular carcinoma and melanoma. However, its role and mechanism of action in pancreatic cancer remain largely unexplored. Analysis of the GEPIA database and experimental validation revealed that DCAF1 is significantly overexpressed in pancreatic cancer tissues and cell lines. Functional assays demonstrated that DCAF1 knockdown i

Coupled Bone-Muscle Degeneration in Chronic Pancreatitis: A Juvenile Porcine Model of Secondary Osteosarcopenia.

Osteosarcopenia, characterized by concurrent bone loss and muscle wasting, significantly impacts mobility and quality of life. While age-related primary osteosarcopenia is well-studied, secondary osteosarcopenia (SOS) caused by chronic diseases remains poorly understood, particularly in young individuals. The present study aimed to comprehensively characterize musculoskeletal alterations associated with SOS using a juvenile porcine model of cerulein-induced chronic pancreatitis. Femoral bone analysis included densitometry, mechanical testing, histomorphometry, and serum bone turnover markers.

Ciliogenesis in pancreatic neuroendocrine tumors: insight into the role of WDR60.

Primary cilia have emerged as key regulators in cancer biology, influencing tumor progression and therapeutic response through diverse signaling pathways. In this study, we identify WDR60, a component of the dynein-2 complex essential for retrograde intraflagellar transport, as a novel modulator of pancreatic neuroendocrine tumor (Pa-NET) behavior. Transcriptomic analysis of the GEO dataset GSE73338 revealed that WDR60 is significantly upregulated in both primary and metastatic Pa-NETs compared to normal pancreatic islets. Moreover, WDR60 expression is higher in G2 compared to G1 Pa-NETs. Func

Piperine Inhibits the Immune Checkpoint CD47 to Enhance Immunity Against Pancreatic Cancer.

Pancreatic cancer is a common malignant tumor of the digestive tract, with insidious onset and rapid metastasis. Most patients have already developed to an advanced stage at the time of diagnosis, and the fatality rate is very high. In recent years, immunotherapy has emerged as a promising potential treatment for pancreatic cancer. Some studies have shown that piperine has the potential to inhibit pancreatic cancer, but the role of piperine in tumor progression and tumor immunity in vivo remains unclear. The effects and mechanisms of piperine on the immunity of pancreatic cancer were studied.

CTHRC1 Derived From Cancer-Associated Fibroblasts Promotes Pancreatic Cancer Progression and Metastasis via the LIF-STAT3 Pathway.

BACKGROUND: Collagen triple helix repeat containing 1 (CTHRC1) is a secreted protein involved in tissue remodeling and fibrotic processes, which also suggests emerging roles in cancer. Studies have shown that it is mainly expressed in the outer membrane fibroblasts of injured arteries and in the neointimal smooth muscle cells, where it promotes cell migration and tissue damage repair. However, the regulatory role of CTHRC1 as a tumor microenvironment factor in pancreatic cancer is not well understood. METHODS: We employed multi-omics analysis combined with cellular and animal experiments to e

Direct in vivo reprogramming to relieve tissue ischemia via induced vasculogenesis.

There is an ongoing need for innovative cell and regenerative medicine therapies to promote vascular repair and regeneration to meet the growing global health concern of ischemic cardiovascular diseases. Direct reprogramming of somatic cells into an induced pluripotent state caused scientists to reconsider prior theories of cell differentiation and develop approaches to directly reprogram cells in vivo into other states or fates to adaptively replace cells lost to aging or disease. While direct in vivo reprogramming of fibroblast to cardiomyocytes, neurons, or pancreatic beta islet cell states

Durotaxis is a driver and potential therapeutic target in lung fibrosis and metastatic pancreatic cancer.

Durotaxis, cell migration along stiffness gradients, is linked to embryonic development, tissue repair and disease. Despite solid in vitro evidence, its role in vivo remains largely speculative. Here we demonstrate that durotaxis actively drives disease progression in vivo in mouse models of lung fibrosis and metastatic pancreatic cancer. In lung fibrosis, durotaxis directs fibroblast recruitment to sites of injury, where they undergo mechano-activation into scar-forming myofibroblasts. In pancreatic cancer, stiffening of the tumour microenvironment induces durotaxis of cancer cells, promoting

A novel FAP-targeting antibody-exatecan conjugate improves immune checkpoint blockade by reversing immunosuppressive microenvironment in pancreatic cancer.

Pancreatic cancer is a highly aggressive malignancy with a dismal prognosis, characterized by a complex tumor microenvironment that promotes immunosuppression and limits the efficacy of immune checkpoint blockade (ICB) therapy. Fibroblast activation protein (FAP) is overexpressed in the tumor stroma and represents a promising target for therapeutic intervention. Here, we developed a novel antibody-drug conjugate (ADC) targeting FAP, and investigated its anti-tumor activity and ability to enhance ICB efficacy in pancreatic cancer. We conjugated a humanized anti-FAP antibody with the potent topo

Tumor microenvironment subtyping in pancreatic ductal adenocarcinoma: New avenues for personalized therapeutic strategies.

In the past decade, single-cell-resolved approaches have uncovered the extensive heterogeneity of pancreatic ductal adenocarcinoma (PDAC), reshaping our understanding of this complex solid tumor. PDAC entities exhibit both intra- and inter-tumor heterogeneity at the tumor and stromal levels, translating into distinct ecosystems and functions, ultimately impacting disease progression and treatment response. Increasing evidence highlights how specific genetic alterations drive unique tumor microenvironment landscapes, affecting fibroblast programming, immune cell contexture and extracellular mat

The tumour microenvironment in pancreatic cancer - new clinical challenges, but more opportunities.

Patients with advanced-stage pancreatic ductal adenocarcinoma (PDAC) predominantly receive chemotherapy, and despite initial responses in some patients, most will have disease progression and often dismal outcomes. This lack of clinical effectiveness partly reflects not only cancer cell-intrinsic factors but also the presence of a tumour microenvironment (TME) that precludes access of both systemic therapies and circulating immune cells to the primary tumour, as well as supporting the growth of PDAC cells. Combined with improved preclinical models of PDAC, advances in single-cell spatial multi
