Pancreatic ductal adenocarcinoma (PDAC) is characterized by its intricate biology governed by spatiotemporal dynamics in the expression and function of specific proteins. Here, DKK3 is identified as a dynamic player with a dual role in PDAC. Using the KRASG12D-driven mouse model with homozygous (DDKC) and heterozygous (DKC) DKK3 knockout, its stage and compartment-specific functions are investigated. Knockout mice exhibited shorter lifespans with a higher incidence of high-grade, desmoplastic, and metastatic cancers. DKK3-deficient acini exhibited a marked increase in acinar-to-ductal metaplas

Fibroblast-driven deposition of extracellular matrix, particularly type III collagen, is linked to poor outcomes in patients with liver fibrosis, fibrotic solid tumors such as hepatocellular carcinoma and pancreatic cancer as well as in patients with other fibrotic diseases. The pro-peptide of type III collagen (PRO-C3) serves as a serum biomarker of active collagen formation. PRO-C3 is elevated and prognostic in patients with liver fibrosis and pancreatic cancer, suggesting that targeting mechanisms behind PRO-C3 elevation could benefit these individuals. In this study, we sought to identify

Microplastics (MPs), as a kind of emerging pollutant, have attracted widespread attention. The effects of MPs of different diameter sizes on chronic pancreatitis and their underlying mechanisms remain to be explored. Therefore, we established a mouse chronic pancreatitis model exposed to two-diameter polystyrene particles (2 or 0.5 μm) for 6 weeks. We found that MPs exposure resulted in severe pancreatic fibrosis in mice, which was negatively correlated with particle size. In-depth mechanistic exploration revealed that after exposure to MPs, MPs can enter pancreatic stellate cells through the

Acute kidney injury (AKI) progression is driven by mitochondrial redox collapse in proximal tubular epithelial cells (PTECs), where reactive oxygen species (ROS) surge and molybdenum (Mo) metabolic dysregulation create an "oxidative storm-defense collapse" cycle. Conventional antioxidant therapies fail to halt AKI chronicity due to their inability to restore Mo-dependent detoxification enzymes (e.g., Mo-containing Amidoxime Reducing Component, mARC). To address this dual pathology, we developed N-acetylcysteine (NAC)-modified molybdenum disulfide quantum dots (NMDs) that implement an endo-exog

Gemcitabine-induced thrombotic microangiopathy (GITMA) is a rare yet devastating complication in patients receiving gemcitabine, especially at cumulative doses above 20,000 mg/m2. We report the case of a 72-year-old woman with metastatic pancreatic adenocarcinoma who developed severe thrombotic microangiopathy (TMA) during gemcitabine and nab-paclitaxel therapy. Her initial presentation included thrombocytopenia, microangiopathic hemolytic anemia, and acute kidney injury. A kidney biopsy confirmed TMA and, given the high likelihood this was related to gemcitabine, therapy was discontinued and

Acute pancreatitis (AP), a potentially fatal disorder driven by macrophage-associated inflammation, involves mitochondrial reactive oxygen species (ROS) overproduction with pancreatic acinar cells (PACs). This ROS surge damages mitochondria, causing mitochondrial DNA (mt-DNA) leakage and PACs apoptosis. Released mt-DNA then activates the pro-inflammatory cyclic GMP-AMP synthase-stimulator of interferon genes (cGAS-STING) pathway in macrophages, exacerbating disease progression. Critically, while scavenging mitochondrial ROS can halt this cycle, existing mitochondria-targeted drugs fail to pene

Cystic fibrosis (CF) is commonly associated with musculoskeletal issues including inflammatory arthritis, CF arthritis. We present a retrospective cohort study which aims to describe the clinical characteristics, prevalence, and demographic associations of CF arthritis using both a clinical and literature cohort. We identified adult CF patients (≥ 18 years) with arthritis from the rheumatology clinic at Rigshospitalet (2020-2022). The clinical cohort (CC) was reviewed through electronic medical records. Literature cases were identified by searching online databases for relevant studies, case r

Pancreatic adenocarcinoma is marked by high mortality and limited treatment options. In mice, HSP90 inhibition limits cancer-associated fibroblasts activation and enhances T cell infiltration, sensitizing tumors to PD-1 blockade. We hypothesized that co-administration of pembrolizumab (anti-PD-1) with XL888 (an HSP90 inhibitor) would be safe and induce measurable immunological changes in the PDAC tumor microenvironment that could influence clinical outcome. We report results from an expansion cohort of patients with advanced PDAC (n = 16), who were enrolled in a single-center, open-label, nonr

Spermine synthase (Sms), a key enzyme in polyamine biosynthesis, catalyzes the conversion of spermidine to spermine using decarboxylated S-adenosylmethionine (dcAdoMet) as an aminopropyl donor. Although Sms is well-characterized in eukaryotes, it is relatively rare in bacteria, where spermine in some species is probably produced by non-specific aminopropyltransferases. In humans, SMS mutations cause Snyder-Robinson syndrome (SRS), an X-linked disorder characterized by intellectual disability, osteoporosis, and neurological dysfunction due to disrupted polyamine homeostasis. Structural studies

The loss of pancreatic beta cell function leads to chronically high blood glucose levels, contributing to diabetes mellitus, one of the leading causes of morbidity and mortality worldwide. Understanding the molecular mechanisms that regulate beta cell function could pave the way for the development of more effective antidiabetic treatments. In this study, we identify the evolutionarily conserved transforming growth factor β-1 stimulated clone D1 (TSC22D1) protein as a previously unknown regulator of beta cell function. TSC22D1 depletion in INS-1E cells enhances the expression of key beta cell

OBJECTIVE: The genetic architecture underlying bone metabolic disorders (BMetDs) remains poorly characterized. Utilizing genomic structural equation modeling (Genomic-SEM) and multiple post-GWAS approaches, we estimated causal single nucleotide polymorphisms (SNPs) associated with BMetDs-independent metabolic variations, identifying 26 novel risk loci (including 14 genome-wide significant loci). Multi-omics analyses-including tissue-specific, cellular-level, and genomic element annotations-were applied to prioritize susceptibility loci and regulatory elements. Additionally, polygenic risk scor

Cuproptosis, a newly identified form of cell death, is closely linked to copper homeostasis and protein lipoylation. Using a multi-omics approach, we firstly reveal that SERPINB3 confers cuproptosis resistance in pancreatic cancer and functions as a theranostic biomarker. Mechanistically, SERPINB3 inhibits FDX1 transcription by activating the MAPK signaling pathway, thereby conferring cuproptosis resistance. We further demonstrated that SERPINB3 directly interacts with MEK1, impeding its chaperone-mediated autophagic degradation, which ultimately leads to sustained activation of the MAPK signa

Multiple case-controlled studies have shown that analyzing fragmentation patterns in plasma cell-free DNA (cfDNA) can distinguish individuals with cancer from healthy controls. However, there have been few studies that investigate various types of cfDNA fragmentomics patterns in individuals with other diseases. We therefore developed a comprehensive statistic, called fragmentation signatures, that integrates the distributions of fragment positioning, fragment length, and fragment end-motifs in cfDNA. We found that individuals with venous thromboembolism, systemic lupus erythematosus, dermatomy

BACKGROUND: Pulmonary tumour thrombotic microangiopathy (PTTM) is a malignancy-associated disease that causes fatal pulmonary hypertension (PH). Due to its rapidly progressive clinical course, antemortem diagnosis is often difficult. Pulmonary tumour thrombotic microangiopathy shares several clinical and imaging characteristics with chronic thromboembolic pulmonary hypertension (CTEPH), making differentiation between these two diseases challenging.
CASE SUMMARY: A 64-year-old woman with pancreatic cancer treated with chemotherapy presented with shortness of breath and cough over the past 5 mo

Pancreatic ductal adenocarcinoma (PDAC) has one of the poorest prognoses of all cancer types. Immune checkpoint inhibitors (ICIs) are currently not indicated for patients with PDAC except for those with high microsatellite instability. In this study, we developed an immunocompetent orthotopic transplant mouse model with Kras and Trp53 mutations, characterized by high fibrosis and an immunosuppressive tumor microenvironment, closely mimicking human PDAC lesions. This model provides a robust platform for investigating strategies for improving ICI efficacy. We observed that ICI monotherapy yielde

The "obesity paradox" suggests a protective effect of obesity on certain diseases including cardiovascular and respiratory illnesses, but results remain inconsistent, largely due to reliance on body mass index (BMI) alone. To clarify this, we used Mendelian randomization (MR) to assess causal effects of multiple obesity-related anthropometric traits on key clinical outcomes. We conducted MR analyses using summary statistics from genome-wide association study to investigate the causal effects of 13 obesity-related anthropometric indicators-including BMI, basal metabolic rate (BMR), hip circumfe

Metabolic dysfunction-associated steatotic liver disease (MASLD) is the most common liver disease worldwide and is closely linked to obesity, insulin resistance, type 2 diabetes, and dyslipidemia. It ranges from simple steatosis to steatohepatitis (metabolic dysfunction-associated steatohepatitis), which can progress to fibrosis, cirrhosis, or liver cancer. Beyond liver-related issues, MASLD is a systemic disease strongly associated with extrahepatic complications such as cardiovascular disease (CVD), chronic kidney disease (CKD), cancers, endocrine disorders, musculoskeletal problems, and sle

BACKGROUND: Rising antimicrobial resistance of Helicobacter pylori is a public health challenge. Genomic-based susceptibility testing allows for the identification of resistance-associated mutations, complementing conventional diagnostics and advancing towards pathogen-based personalised therapies. Our study aimed to identify genes and mutations involved in antimicrobial resistance in H pylori and evaluate the extent to which these markers can be used as predictors of phenotypic resistance against clarithromycin and levofloxacin.
METHODS: In this retrospective phenotypic and genotypic observa
