Single-Cell Transcriptomic Analysis of Tumor Heterogeneity and the Microenvironment in Pseudomyxoma Peritonei.

Pseudomyxoma peritonei (PMP) is characterized by progressive mucus accumulation, extensive stromal fibrosis, rare extraperitoneal metastasis, limited therapeutic options, and frequent recurrence. However, the microenvironmental ecosystem of PMP, particularly in metastatic lesions, remains poorly understood. Here, we integrated single-cell RNA sequencing, whole-exome sequencing, bulk RNA sequencing, and histopathologic validation to construct a high-resolution atlas of primary and paired metastatic tumors. Epithelial cells showed distinct functional states, including a TFF3+ mucus secretion-ass

Fusion Gene Detection Enhances Clinical Outcome Prediction of Hepatocellular Carcinoma Through Machine Learning Analyses.

Hepatocellular carcinoma (HCC) is one of the most lethal malignancies for humans. Assessing the clinical outcomes of HCC remains challenging. In this study, a panel of 20 fusion genes in 200 HCC samples was analyzed to predict the recurrence and survival rates of patients with HCC undergoing surgical interventions using machine learning models. The results showed that fusion genes, Milan criteria, serum α-fetoprotein (AFP), and pathology grade had moderate predictive accuracy for HCC recurrence. However, the combination of selected fusion genes with these clinical parameters significantly enha

Mechanosensors Collaborate to Regulate DNA Damage Repair in Hepatocellular Carcinoma.

UNLABELLED: Increased matrix stiffness is a biomechanical feature of hepatocellular carcinoma (HCC) that functions as an important promoting factor driving tumor progression. Characterization of the mechanosensors and the underlying downstream mechanisms driving HCC could help identify potential treatment strategies. In this study, we developed a standard procedure for screening and identifying mechanosensors, revealing two HCC mechanosensors, SDC4 and PKD2. These two mechanosensory pathways collaboratively mediated matrix stiffness-promoted DNA damage repair (DDR). Specifically, SDC4 and PKD2

KRAS Mutations Reprogram RNA m6A Modifications to Drive CD73-Dependent Immune Evasion in Colorectal Cancer.

UNLABELLED: KRAS mutations, present in about 40% of colorectal cancers, are strongly associated with an immunosuppressive tumor microenvironment characterized by restricted immune infiltration and poor responses to immunotherapy. In this study, we found that KRAS mutations remodel the N6-methyladenosine (m6A) epitranscriptome to promote immune evasion. Methylated RNA immunoprecipitation sequencing revealed that KRAS-mutant cells exhibit increased m6A deposition on CD73 mRNA, enhancing its stability through IGF2BP3. TEAD4-dependent recruitment of the METTL3 methyltransferase complex induced the

Windows of opportunity for genotoxic E. coli in colorectal cancer initiation.

Polyketide synthase-positive Escherichia coli (pks+ E. coli) has attracted significant attention following the discovery that its genotoxic metabolite, colibactin, imprints distinct mutational signatures in intestinal epithelial cells that are detectable in ∼12% of colorectal cancer (CRC) cases. Although these signatures are present in established tumors, growing evidence suggests that colibactin acts early in tumorigenesis, positioning pks+ E. coli as a potential contributor to CRC initiation. Mechanistic studies show that colibactin genotoxicity is highly context dependent and requires direc

Age- and sex-dependent associations of metabolic syndrome with colorectal cancer in the UK Biobank.

BACKGROUND: The concurrent rise in metabolic syndrome (MetS) and the changing age profile of colorectal cancer (CRC) necessitates clarifying the heterogeneity in their association across demographic groups. This study aimed to define the precise age- and sex-specific associations between MetS and CRC risk, addressing a key gap to inform targeted prevention paradigms. METHODS: In this cohort study, we analysed data from UK Biobank participants aged 40-69 years (recruited 2006-2010). Participants were stratified by age (<60 or ≥60 years) and sex into four groups. Multivariable Cox proportional

Robotic Versus Laparoscopic Gastrectomy for Advanced Gastric Cancer: Prognostic Implications of Surgical Recovery and Oncologic Control in a Propensity Score-Matched Study.

BACKGROUND: Robotic gastrectomy may improve precision during radical lymphadenectomy, but its prognostic relevance in advanced gastric cancer remains uncertain. The authors hypothesized that robotic surgery may influence outcomes through improved recovery and oncologic control. METHODS: A prospectively maintained registry included 426 curative-intent minimally invasive gastrectomies from 2017 to 2025 (laparoscopic, n = 176; robotic, n = 250). Propensity score-matching yielded 154 pairs. Endpoints included severe morbidity (Clavien-Dindo grade ≥ III), postoperative inflammation, adjuvant chemo

Spatial proteomics deciphers the immune niche landscape of non-ampullary duodenal adenocarcinoma.

BACKGROUND: Non-ampullary duodenal adenocarcinoma (NADA) is a rare malignancy with an incompletely characterised tumour immune microenvironment (TME). We aimed to elucidate its immune niche landscape and validate a clinically applicable transcriptomic surrogate. METHODS: In 38 resected NADA cases, multiplex immunofluorescence spatial proteomics was performed on tissue microarrays, validated by cohort-wide whole-slide imaging. A 10-gene qRT-PCR panel capturing adaptive immune resistance mechanisms was integrated with spatial data. Molecular microsatellite instability (MSI) testing confirmed mi

Benzovindiflupyr at estimated drinking water concentrations induces hepatic injury via the NF-κB signaling pathway: In vivo, in vitro, and in silico studies.

While the extensive application of benzovindiflupyr (BZF) contributes to crop protection, it may pose potential threats to non-target species. However, limited research has investigated the effects of environmentally relevant concentrations of BZF on non-target organisms. By integrating in vivo, in vitro, and in silico approaches, we revealed that BZF induces hepatic injury in zebrafish via the NF-κB signaling pathway activation at estimated drinking water concentrations. Specifically, following a 28-day exposure to gradient BZF concentrations (0, 5 and 50 μg/L), adult zebrafish exhibited mark

Assessing Graded Histological Regression as Basis for Treatment Modification after Neoadjuvant Therapy in LAGC: An Exploratory Phase II Study.

BACKGROUND: Whether to change adjuvant regimen after ineffective neoadjuvant chemotherapy in locally advanced gastric cancer (LAGC) remains unclear. We conducted an exploratory study on whether graded histologic regression (GHR) < 50% was a potential indicator for regimen modification. PATIENTS AND METHODS: The study was a prospective, single-center, open-label, randomized, phase II trial. Patients were randomly assigned to group A (paclitaxel-modified regimen) and group B (original regimen) for adjuvant chemotherapy. The primary endpoint was disease-free survival (DFS), with secondary endpoi

Rare Molecular Variants of Gastrointestinal Stromal Tumors (GISTs): Clinical Implications and Overview of Current Evidence.

Gastrointestinal stromal tumors are the most common sarcomas of the gastrointestinal tract. They are characterized by a distinct molecular profile, most frequently involving activating mutations in the KIT or PDGFRA genes, the identification of which has enabled the development of effective targeted therapies and has significantly improved patient outcomes. Despite significant therapeutic advances, treatment with tyrosine kinase inhibitors remains associated with the risk of primary or secondary resistance in a subset of patients. This phenomenon has highlighted the considerable biological het

Cilia-Structured Lipiodol Droplets Enable Long-Term Transarterial Embolization Therapy by Nano-Interlocking Adhesion.

Transarterial embolization (TAE) offers an effective and minimally invasive therapy for unresectable hepatocellular carcinoma (HCC), one of the most lethal malignancies globally, but being limited by short-term embolization failure for example recanalization due to weak interfacial adhesion. Herein, we report cilia-structured lipiodol (CS-lipiodol) droplets by self-assembling dual-asymmetric microparticles with hydrophilic cilia-structured hemispheres and hydrophobic plain hemispheres at the lipiodol-water interface, enabling long-term TAE therapy by the cilia-mediated nano-interlocking effect
