Epidemiological investigation of Blastocystis in dairy cattle in Xinjiang, China: Unusual detection of avian-adapted subtype ST7.

Blastocystis is a common anaerobic intestinal protist with a broad host range, including humans and various animals. In this study, a total of 907 fecal samples were collected from dairy cattle across six large-scale farms in Xinjiang and screened for the presence of Blastocystis by PCR targeting the small-subunit ribosomal RNA (SSU rRNA) gene. The overall infection rate was 18.1% (164/907), with significant variation among farms (1.4%-29.6%) and age groups (12.1%-20.8%). Ten subtypes of Blastocystis were identified: ST1 (n = 6), ST5 (n = 14), ST7 (n = 13), ST10 (n = 18), ST14 (n = 7), ST21 (n

Infection-Triggered Disease Flare With Extraintestinal Manifestations in Trichohepatoenteric Syndrome: A Case Report.

Trichohepatoenteric syndrome (THES) is a rare, autosomal recessive disorder characterized by early-onset diarrhea, woolly hair, and facial dysmorphism and variable multisystem involvement. Herein, we report a 17-year-old male with a genetically confirmed THES who presented with worsening diarrhea, fatigability, weight loss, and epigastric pain following intestinal amebiasis. His disease course was notable for extraintestinal and inflammatory manifestations, including chronic recurrent multifocal osteomyelitis, syringomyelia, and inflammatory bowel disease-such as enteropathy, highlighting a br

Contributions of intestinal protists on the human gut landscape through the lens of Entamoeba spp.

Intestinal protists represent an underappreciated yet functionally significant component of the human gut microbiome. Historically dismissed as parasites or transient contaminants, many of these microbial eukaryotes, particularly Entamoeba spp., are now recognized as integral to gut ecosystem function and host immune homeostasis. This review examines the complex roles of Entamoeba spp. in the mammalian gut, positioning them as dynamic microbiome "landscapers" that influence host-pathogen interactions, immune tone, and microbial diversity. We explore the evolutionary adaptation of Entamoeba to

Comparative gut microbial enzyme functions in human amoebiasis: Implications for host-microbe interactions and veterinary parasitic models.

BACKGROUND: The intestinal microbiome is a critical component of host defense and metabolic regulation in both humans and animals. Parasitic infections such as amebiasis disrupt microbial enzyme activities, potentially influencing host physiology. AIM: To investigate functional differences in gut microbial enzymes between humans infected with Entameba histolytica and healthy controls and to provide insights relevant to comparative parasitic pathobiology. METHODS: Stool samples collected from patients diagnosed with amebiasis, as well as from healthy control individuals, were analyzed using 1

Gastrointestinal parasites circulating in wild boars from central Italy and molecular characterization of Balantioides coli, Blastocystis sp. and Enterocytozoon bieneusi.

The growing abundance and geographic spread of wild boar (Sus scrofa) populations in Europe have heightened concerns about their contribution to the transmission of parasites of zoonotic and veterinary importance. This study investigated the occurrence and zoonotic potential of gastrointestinal parasites in 46 wild boars sampled during the 2023 hunting season in Italy. Copromicroscopic analyses revealed that 47.8% of animals were positive for at least one parasite taxon, with frequent coinfections. Thirteen taxa were identified by microscopy, including protozoans (Entamoeba coli, Iodamoeba but

Entamoeba histolytica Gal/GalNAc lectin intermediate subunit as a potential driver of inflammation and epithelial damage in intestinal amebiasis.

Entamoeba histolytica is a common enteric protozoan that sometimes transitions from a symbiont to a pathogen, causing intestinal amebiasis and extraintestinal abscesses in the host. The early-to-intermediate stages of intestinal amebiasis are characterized by an intense inflammatory response. Here, using the cecal infection model of C3H/HeNCrl mice, we performed single-cell RNA sequencing of cecal tissues and identified the prominent pro-inflammatory function of host macrophages in intestinal amebiasis. On the amebic cell surface, Igl is the intermediate subunit of galactose- and N-acetyl-D-ga

Blastocystis, an Enigmatic Parasite.

Blastocystis spp. is the most prevalent human enteric protist, yet its clinical significance remains controversial. Currently recognized as a genetically diverse stramenopile with zoonotic potential, it comprises at least 42 subtypes, of which ST1-ST4 account for most human infections. Despite more than a century of research, its pathogenic role and ecological interactions are still debated. We performed a comprehensive bibliometric analysis of global scientific production on Blastocystis spp. from 1917 to 2025 using the Scopus database, retrieving 3710 publications. Temporal trends, geographi

Modulatory role of Blastocystis-derived solubilized antigens in colorectal carcinogenesis: A systematic review.

Colorectal cancer develops through complex interactions among genetic alterations, immune responses, and the intestinal microenvironment. Increasing evidence suggests that intestinal parasites may also contribute to colorectal carcinogenesis (CRC). Blastocystis sp. (Blastocystis) is one of the most common intestinal protozoans worldwide. Although its role in disease remains controversial, it has been associated with chronic inflammation and disruption of normal gut homoeostasis. Therefore, this systematic review aimed to summarise the current experimental evidence on the immunomodulatory effec

Host-parasite interactions during Entamoeba histolytica infection: an ex vivo human intestinal model.

Amebiasis, caused by the protozoan parasite Entamoeba histolytica, remains a significant public health problem in specific endemic regions. Current experimental models have limitations in accurately replicating human pathogenesis, highlighting the need for alternative platforms that preserve tissue complexity and physiological context. This study aimed to evaluate E. histolytica infection using an ex vivo human precision-cut intestinal slice (PCIS) model by analyzing histological and molecular changes over time (1, 3, 12, 24, and 48 h post-infection). Tissue integrity and parasite invasion wer

Evaluation of intestinal immune responses to Entamoeba dispar using the first domestic clinical isolate in Japan.

BACKGROUND: Entamoeba dispar is a non-pathogenic sibling species of Entamoeba histolytica, yet the mechanisms that enable E. dispar to persist in the intestine without causing disease remain poorly understood. Experimental models for dissecting intestinal immune responses to E. dispar have been limited. In Japan and other East Asian countries, E. histolytica infection is relatively common among men who have sex with men, whereas E. dispar infection has rarely been identified. METHODOLOGY/PRINCIPAL FINDINGS: We report the first domestically acquired and culture-established E. dispar infection

Amebic liver abscess: A case report.

Amebic liver abscess (ALA), caused by Entamoeba histolytica, is the most common extraintestinal manifestation of amebiasis. However, in contemporary clinical practice, diagnosis increasingly relies on serology and polymerase chain reaction (PCR), whereas direct microscopic identification of motile trophozoites in aspirated specimens has become rarely documented. Furthermore, optimal management necessitates complete anti-amebic therapy covering both invasive tissue stages and luminal intestinal colonization to prevent relapse. Here, we report a case of a 25-year-old male with a history of splen

Concomitant Borreliosis and Invasive Amoebiasis: A Rare Case with Suspected Sexual Acquisition.

BACKGROUND: The movement of endemic diseases from tropical and disadvantaged areas is gradually spreading to developed countries as well. Amoebiasis, in particular, represents a significant threat to public health, amplified by globalization and increasing contact between humans and animals or vectors. In this way, atypical diseases may emerge in our country, also through an unusual mode of transmission-likely sexual, as "sexually transmitted enteric (STE) diseases". OBJECTIVE: We report and discuss the clinic approach to a rare case of dual human infestations by ecto- and endo-parasites with

Exploring glycocalyx components powering the contact between Entamoeba histolytica and bacteria during non-opsonic phagocytosis.

Phagocytosis is a cornerstone of the immune defence, allowing cells to eliminate pathogens through both opsonic and non-opsonic pathways. In protozoa such as Entamoeba histolytica, non-opsonic phagocytosis is not only central to survival but also to its ability to cause disease, with its life cycle closely intertwined with the intestinal microbiota, which supplies bacteria and their metabolic byproducts to support trophozoite growth. Yet, despite its significance, the molecular basis of how E. histolytica recognises and engulfs bacteria remains largely unresolved. This review examines the cell

Diagnosis of Intestinal Entamoeba histolytica Infection by Rectal Swab: An International Multisite Cross-Sectional Study Between Taiwan and Japan.

BACKGROUND: Stool samples are the standard specimen for molecular diagnosis of intestinal Entamoeba histolytica infection. Rectal swabs, an easier sample collection method, are increasingly used for diagnosing many enteric pathogens; however, their diagnostic utility for E. histolytica remains insufficiently evaluated. METHODS: Stool and rectal swab specimens were collected simultaneously from participants tested for intestinal E. histolytica infection. The detection capability of rectal swabs using a modified DNA extraction protocol was compared with that of stool specimens using E. histolyt
