Derivation of a Point of Departure using NAMs for application in Quantitative Risk Assessment of fragrance materials.

Skin sensitization is a key endpoint for the safety assessment of topical consumer products. Ingredients with the potential to act as skin sensitizers differ markedly in their threshold for induction but can be used safely if their potency is characterized and exposure remains within an appropriate margin of safety. To this end, the fragrance industry co-developed Quantitative Risk Assessment (QRA) which starts with the No-Expected-Sensitization-Induction-Level (NESIL). Historically, QRA relies on a weight of evidence approach based on animal data, human confirmatory tests and read across. To

Lymphotoxin Beta Receptor, but Not Its Lymphotoxin Alpha-Containing Ligands, Is Essential for the Development of Experimental Dermatitis.

Allergic contact dermatitis (ACD) is a chronic inflammatory skin disorder the development of which is driven by allergen sensitization in peripheral lymphoid organs and local cutaneous inflammation. Lymphotoxin (LT) and its receptor LTβR are critical for lymphoid organogenesis and immune regulation in barrier tissues, but their role in ACD pathogenesis remains incompletely defined. This study aimed to delineate differential contribution of the LTβR-dependent signaling in oxazolone-induced dermatitis. We examined Lta knockout (Lta KO) mice, which lack both soluble LTα3 and membrane-bound isofor

Applicability of the U-SENS™ assay for assessing skin sensitization potential of agrochemical active ingredients and formulations.

Skin sensitization testing is essential for the regulatory classification of agrochemicals, including active ingredients (AIs) and formulations. Although several alternative methods have been developed, their application to agrochemicals remains limited. In particular, the performance of the U-SENS™ assay, which addresses key event three (dendritic cell activation) of the adverse outcome pathway, has not yet been evaluated for these products. This study assessed the U-SENS™ applicability to predict the sensitization potential of agrochemical formulations and their AIs. Twelve formulations and

Contact Allergy to Alkyl Glucosides: A Review-Part 1.

This is Part 1 of a 2-part narrative review of contact allergy and allergic contact dermatitis (ACD) to alkyl glucosides. It discusses the nature of alkyl glucosides (mainly decyl, lauryl, cetearyl, and coco-glucoside), their properties and applications, results of patch testing in routine testing and in groups of selected patients, and presents all reported case series and case reports of ACD to decyl and lauryl glucoside. The alkyl glucosides are nonionic surfactants that are produced by the reaction of glucose with fatty alcohols and that are increasingly being used in cosmetics. This class

Occupational Allergic Contact Dermatitis From Over-Ear Headphones: Chemical Analysis Reveals Octylisothiazolinone and Other Potential Skin Sensitizers.

BACKGROUND: Over-ear headphones are a growing source of allergic contact dermatitis (ACD). Identifying well-known and new culprit sensitizers in such products is challenging because of a lack of labelling and limited cooperation from manufacturers. OBJECTIVES: To report potential skin sensitizers in occupationally-used headphones from patients attending an occupational allergy clinic. PATIENTS AND METHODS: The over-ear headphones from two octylisothiazolinone (OIT)-allergic patients with ear ACD were chemically analysed using ultra performance liquid chromatography-tandem mass spectrometry (

Contact allergies from clothing textiles - is there a cause for growing concern?

Allergic contact dermatitis (ACD) due to chemicals in textiles and other clothing has recently been declared to be of growing concern in Europe. An Annex XV dossier proposing a generic ban of skin sensitisers in these products under the European Chemicals legislation REACH estimated the prevalence of clothing-related ACD in the general population to be 0.8-1.0%. The present study took a critical look at the data basis, the methodological approach and the assumptions underlying the calculation of the burden of disease in the Annex XV report. The reported incidence and prevalence estimates were

BACKGROUND: Maudsley Family Therapy (MFT), and its manualised version, Family-Based Therapy (FBT), are the only well-established treatment interventions for adolescent anorexia nervosa (AN), with treatment efficacy primarily measured by improvements in eating behaviours and weight restoration. A crucial component of this therapy is an intensive home-based refeeding intervention that requires a substantial commitment from parents for up to one year. While this treatment works to restore weight in a proportion of adolescents, very little is known about its impacts on family distress, relationshi

Iturin A disrupts Cryptococcus neoformans membrane integrity, attenuates virulence, and synergizes with amphotericin B.

BACKGROUND: Use of the ergot-derived dopamine receptor agonists (cabergoline and pergolide) is associated with an increased risk of cardiac valvulopathy. Pergolide was withdrawn from the US market in 2007 because of the risk of valvular heart disease, while the European Medicines Agency (EMA) required a reduction in the maximum daily dosage of cabergoline and pergolide from 6 mg/day to 3 mg/day in 2008. In Japan, the package inserts of both drugs were revised in April 2007 to request that physicians conduct periodic ultrasonic cardiography (UCG) examinations for patients taking cabergoline or

Hematological phenotypes in GATA2 deficiency syndrome arise from aging, maladaptation to proliferation, and somatic events.

BACKGROUND: As self-applied manicures have gained popularity, it is important for physicians to remain informed about potential associated side effects. Traditional polish remains most popular among nail enthusiasts, but the pursuit of durability and convenience accelerated development of other nail cosmetic products, including gel polish, acrylic nails, and press-on nails. Despite documented adverse effects among beauty professionals and salon customers, individuals practicing at-home nail care routines may face similar, if not more, frequent complications due to misinformation, inadequate tr

Extended fragrance ingredients surveillance study (EFISS)-protocol for a clinical surveillance study on contact allergy to 7 fragrance materials in widespread use but hitherto not systematically patch tested.

Contact allergy (CA) is not uncommon in the population, including to various fragrance allergens. If not diagnosed correctly, allergic contact dermatitis (ACD) may ensue, because targeted allergen avoidance is not possible. The primary objective of the study is to estimate the prevalence of CA to seven fragrance materials in patients with suspected ACD across Europe. Based on the outcome, a conclusion will be drawn as to whether present risk management regarding maximum recommended concentrations of each of these, based on quantitative risk assessment (QRA2), is adequate. The planned study is

TRPV1 Blocker, Peptide HCRG21 from Sea Anemone Heteractis magnifica, Exhibits Effectiveness in Psoriasis and Dermatitis in In Vivo Models.

Psoriasis and allergic contact dermatitis (ACD) are the most common chronic inflammatory diseases, which are accompanied by epithelial alterations and a T cell-mediated immunopathology. In this study, we investigated the anti-ACD and anti-psoriasis effects of sea anemone Heteractis magnifica peptide HCRG21, a blocker of the TRPV1 channel, in 2,4-dinitrofluorobenzene (DNFB)- and imiquimod (IMQ)-induced mouse models, respectively. We found that topical application of 0.005-0.1% HCRG21 gels normalized hematological and immunological blood parameters in mice, significantly reduced the severity of

Contact Allergy to Ingredients of Hair Cosmetics Associated with Occupational and Non-Occupational Exposure-Trends from 1995 to 2020 in Central Europe, with or without Regulation.

BACKGROUND: Hair cosmetic products contain various chemicals which are partly potent contact sensitizers. These may cause occupational hand eczema in hairdressers, but also scalp and face dermatitis in consumers. OBJECTIVE: To (i) highlight differences between the spectrum of occupational and non-occupational exposure reflected in sensitization, and (ii) correlate trends in the last decades with regulatory interventions (or the lack thereof). METHODS: Patch test and clinical data collected by the IVDK (https://www.ivdk.org) between 01/1995 and 12/2020 were subjected to a pooled re-analysis.

Use of human predictive patch test (HPPT) data for the classification of skin sensitization hazard and potency.

Since the 1940s, patch tests in healthy volunteers (Human Predictive Patch Tests, HPPTs) have been used to identify chemicals that cause skin sensitization in humans. Recently, we reported the results of a major curation effort to support the development of OECD Guideline 497 on Defined Approaches (DAs) for skin sensitization (OECD in Guideline No. 497: Defined Approaches on Skin Sensitisation, 2021a. https://doi.org/10.1787/b92879a4-en ). In the course of this work, we compiled and published a database of 2277 HPPT results for 1366 unique test substances (Strickland et al. in Arch Toxicol 97:

Hair Product Allergy: A Review of Epidemiology and Management.

Allergy to hair products is an increasingly common issue among people given the exposure to these products on a daily basis. Allergic reactions could be in the form of delayed-type contact dermatitis or the form of immediate-type hypersensitivity reactions. Hair products contain many ingredients and chemicals that patients may have allergies to, but common allergens are hair dyes, fragrances, persulfate salts, ammonium thioglycolate, coconut fatty acid derivatives, and acrylates. Allergy to hair dye is the most common followed by other allergens such as fragrances and persulfate salts. We disc

Fragrance Contact Allergy - A Review Focusing on Patch Testing.

Fragrance materials are widely used in various types of products in daily life and many of them can be contact sensitizers. Contact allergy to fragrances has been reported to be common worldwide. Unlike other groups of contact allergens such as metals and preservatives, fragrance materials in consumer products can be present as single fragrance chemicals or in the form of mixtures known as natural complex substances. Due to the complexity of the fragrance materials and the high number of fragrance substances known to cause contact sensitization, selecting suitable materials for patch testing i
