Approval of New Patch Tests for the Diagnosis of Allergic Contact Dermatitis: An FDA Public Workshop.

Contact dermatitis (CD) is the 5th most common skin disease1 and affects 15-20% of the general population over their lifetimes.2 Currently, the T.R.U.E. TEST® is the only FDA-approved diagnostic product to aid in the diagnosis of Allergic Contact Dermatitis. To address that diagnostic gap, the US Food and Drug Administration held a public workshop in October 2025. These proceedings summarize the workshop presentations and discussions.

Reproducibility of Perfusion-Weighted and Diffusion-Weighted Magnetic Resonance Imaging in Patients with Ocular Tumours.

PURPOSE: Allergic diseases are frequently underdiagnosed in older adults due to overlapping symptoms with other chronic conditions and age-related changes in the immune system. As the elderly population continues to grow, understanding the prevalence and clinical impact of allergic diseases in this group is essential. This study aimed to assess the prevalence of common allergic conditions in individuals aged 65 and older and to examine their associated comorbidities. PATIENTS AND METHODS: We conducted a retrospective chart review of approximately 17,000 patients aged 65 and older who were dia

Application of a next generation risk assessment framework, integrated testing strategy defined approaches and read-across for evaluating the skin sensitization potency: Case studies with 5-hydroxymethylfurfural and 5,5'-oxydimethylenebis (2-furfural).

Evaluating skin sensitization is crucial for the safety of cosmetics and pharmaceuticals. Regulatory bans on animal testing have spurred the development of New Approach Methodologies (NAMs), Integrated Testing Strategies (ITS), and Next Generation Risk Assessment (NGRA). Defined Approaches (DAs) that combine various NAMs have been developed to assess skin sensitization hazards and potency, as recognized by the OECD. However, deriving a quantitative point of departure (PoD) through NAMs in accordance with NGRA remains challenging. This study evaluated the skin sensitization hazard and potency o

A nationwide cross-sectional survey of major allergic diseases in China during 2010-2015 involving 120,000 participants.

Although the prevalence of allergic diseases has been rising in China since the late twentieth century, comprehensive nationwide epidemiological data covering all age groups and multiple allergic conditions remain scarce. To investigate the epidemiological patterns of eight major allergic diseases in China, including their national and regional prevalence, demographic distribution, and comorbidity profiles, the National Epidemiology Study of Asthma and Allergies in China (NESAAC) was a nationwide cross-sectional survey conducted from September 2010 to December 2015. A multistage stratified clu

Occupational Contact Urticaria, Protein Contact Dermatitis and Concomitant Airway Diseases in the Finnish Register of Occupational Diseases in 2005-2020: Main Causes and Occupations at Risk.

BACKGROUND: Occupational contact urticaria (OCU) and protein contact dermatitis (PCD) may co-exist with occupational asthma (OA) and occupational rhinitis (OR) and be caused by the same allergen. These diseases are commonly caused by immediate (immunoglobulin E (IgE)-mediated) allergic sensitisation. Limited data exist on the prevalence and causes of concomitant occupational skin and airway diseases (CSAD). OBJECTIVES: To analyse data from the Finnish Register of Occupational Diseases (FROD) for prevalence, main causes, and at-risk occupations for concomitant occupational skin and airway dise

Reframing Eczema: Th2-Skewed Contact Sensitization, Atopy Patch Testing, and Systemic Contact Dermatitis.

PURPOSE OF REVIEW: We review allergic contact dermatitis (ACD) with predominant Th2 type cytokine expression in the context of chronic cutaneous inflammation. While more has been written about Th1 skewed ACD due to potent allergens in the setting of healthy skin, this review highlights recognition of Th2 skewed ACD in both atopic dermatitis and systemic contact dermatitis and the role of allergen avoidance as an alternative to systemic therapies. RECENT FINDINGS: Th2 skewed ACD rarely occurs to potent allergens. It more commonly occurs in response to allergens considered “non-sensitizers” in t

Investigation of the potential of bisphenol A substitutes to induce allergic contact sensitization using OECD defined approaches.

Bisphenol A is a high production volume chemical used extensively in the manufacture of polycarbonate plastics, epoxy resins, and thermal printer paper with a high potential for occupational and post-production dermal exposure. Bisphenol A-containing plastics were commonly used in food packaging, resulting in significant public exposure through leaching into foodstuff. The public is also at risk of dermal exposure due to environmental contamination. Due to public health concerns regarding the potential for endocrine disrupting effects, efforts have been applied to replace bisphenol A with sa

Occupational Primary Red Meat Allergy With Sensitization to Serum Albumins, Sus s 1, and Bos d 6.

To date, occupational primary red meat allergies have not been reported. A 42-year-old man visited our clinic with allergic symptoms after ingesting red meat. For the past 5 years, he has developed a tingling in the throat and itchy erythema over the whole body immediately after ingesting rare pork and beef, and fresh cream. He had been working at a butcher's shop since he was 22, mainly handling pork and beef. At the age of 26, he noticed wheals appearing on the parts of his body which had been in direct contact with raw meat. At the same time, his atopic dermatitis was worsening. Specific Ig

Hypersensitivity reactions associated with nutraceuticals and dietary supplements: A narrative review.

The global use of nutraceuticals and dietary supplements-including vitamins, minerals, probiotics, and botanical extracts-has increased substantially, driven largely by consumer perceptions of safety and health benefits. However, their potential to cause hypersensitivity reactions (HSRs) is underrecognized. This review synthesizes published case reports, case series, systematic reviews, and pharmacovigilance data to characterize HSRs linked to these products, identify risk factors, and examine diagnostic and regulatory challenges. We classified reactions as immediate (< 1-6 h after administrat

Intraosseous Schwannoma of the Spine: A Report of Three Cases.

BACKGROUND: Asparagus (Asparagus officinalis) is an extensively grown and consumed vegetable. To a lesser extent than other Liliaceae vegetables, allergic contact dermatitis (ACD) due to asparagus has been reported. However, only a few case reports of asparagus IgE-mediated allergy have been published. In a previous study, we demonstrated that two lipid transfer proteins (LTPs) (Aspa o 1.01 and Aspa o 1.02) were relevant allergens of asparagus. OBJECTIVE: We retrospectively analysed the 27 patients diagnosed with asparagus allergy during the last 5 years. All of them reported adverse symptoms

Plasma fractionation in Latin America: a path towards sovereignty and sustainability in the supply of plasma-derived medicines.

PURPOSE: Increased reactive oxygen species (ROS) released by NADPH oxidase and inflammation are associated with arterial hypertension and eye diseases associated with high blood pressure, including glaucoma, retinopathies (e.g., age-related macular degeneration), and choroidopathies affecting ocular function; however, the mechanisms underlying these adverse outcomes remain undefined. The present study was designed to highlight the importance of oxidative stress in severe hypertension-related eye damage. METHODS: Male Wistar rats (n = 7, unless otherwise specified for specific experiments) wer

Benzoyl Peroxide's Sensitisation Potential and Potency in Experimental Methods and Review of Contact Allergy and Allergic Contact Dermatitis.

Positive patch test responses to benzoyl peroxide (BPO) have been reported from patients without and with known exposure. Up to 6.5% were found in the United States and 7.8% in a study including patients from Germany, Austria and Switzerland. We provide an overview of the skin sensitisation potential and potency of BPO based on animal experiments and non-animal methods. BPO tested positive in Guinea Pig Tests and the Local Lymph Node Assay. Application of the current OECD guideline to identify a skin sensitizer by combining non-animal method results gave differing outcomes. Moreover, patch tes

Allergens in Atopic Dermatitis.

Atopic dermatitis (AD) is a chronic inflammatory skin disease with a complex relationship to allergens. While AD itself is not an allergic reaction and does not necessarily involve allergen sensitization, AD patients show higher rates of sensitization to food and inhalant allergens compared to the general population. Recent evidence refining the "dual allergen exposure hypothesis" demonstrates that early oral exposure to allergens through an intact gastrointestinal barrier typically promotes tolerance, while exposure through compromised skin or respiratory barriers often leads to sensitization

Just scratching the surface: A review of pediatric skin allergies.

The skin is a large and sophisticated organ populated by innate and adaptive immune effector cells. These immune cells provide a critical first line of defense against pathogens, but genetic and environmental factors can lead to inappropriate signaling that may manifest as hypersensitivity. The most common cutaneous allergic disorders in children include atopic dermatitis, urticaria/angioedema, and contact dermatitis. In this review, we will briefly review these conditions, with a focus on recent developments in our understanding of the diagnosis and management of these disorders.

Using new approach methodologies for the identification of a sensitizing co-formulant in a plant protection product.

According to Regulation (EC) No 1272/2008, plant protection products (PPPs) can be classified for skin sensitisation by either considering the full formulation or the individual components. For a fraction of PPPs, an application of both assessment strategies results in discrepant classification results. We here aimed to resolve this discrepancy for PPP 1, an exemplary product, which was classified as sensitizing by a positive local lymph node assay but scored negative by the component-based method. We collected further data, as suggested by the regulation, based on a combination of new approac
