According to Regulation (EC) No 1272/2008, plant protection products (PPPs) can be classified for skin sensitisation by either considering the full formulation or the individual components. For a fraction of PPPs, an application of both assessment strategies results in discrepant classification results. We here aimed to resolve this discrepancy for PPP 1, an exemplary product, which was classified as sensitizing by a positive local lymph node assay but scored negative by the component-based method. We collected further data, as suggested by the regulation, based on a combination of new approac

Allergic contact dermatitis (ACD), also known as allergic eczema, is a common inflammatory skin disorder that affects millions of Americans and imposes significant physical, psychological, and economic burdens. Differentiating ACD from other forms of dermatitis remains a challenge, with patch testing as the gold standard. Despite its utility, patch testing can lack diagnostic accuracy, highlighting the importance of molecular biomarkers to refine diagnosis and treatment. Advances in transcriptomics and machine-learning have enabled the identification of biomarkers involved in ACD, such as lori

Type 2 (Th2) allergic diseases are chronic conditions characterized by a Th2-polarized immune response to allergens. These diseases can be categorized by affected barrier sites: skin (atopic dermatitis, allergic contact dermatitis), gut (food allergy), and respiratory tract (e.g., asthma, chronic rhinosinusitis). The global prevalence of Th2 allergic diseases has increased the need for a deeper understanding of their pathophysiology. Several associations have been identified between genetic variants in the genes encoding components of the complement system and allergic disease. Moreover, level

Assays to predict the skin sensitisation potential of substances commonly address the molecular initiating event (MIE) of the adverse outcome pathway (AOP), which is the covalent binding of chemicals to proteins. This represents the first key event in the pathway. In chemico approaches have emerged as validated alternatives to conventional animal testing methods to determine this process. This review explores the integration of biosensor-based approaches to supplement other in chemico methods, for use as alternatives to the 'gold standard' Local Lymph Node Assay (LLNA) in skin sensitisation te

Allergic contact dermatitis (ACD) is a common skin disorder caused by contact with allergens. ACD treatment is based on patient education to avoid contact with allergens. However, sometimes patients may not be able to avoid the allergen because of its close proximity to their living environment. Thus, a novel therapeutic strategy that does not involve the avoidance of allergens is required. We previously reported that methionine, an essential amino acid, significantly suppressed ACD development caused by the repeated application of 1-fluoro-2,4-dinitrobenzene (DNFB). However, the magnitude of

The Spectrophotometric Direct Peptide Reactivity Assay (Spectro-DPRA) is a high-throughput in chemico assay designed to evaluate Key Event 1 (KE1)-covalent binding to skin proteins-in the skin sensitization adverse outcome pathway (AOP). Although prior validation studies have demonstrated its predictive capacity, its classification thresholds differ from those established in the standard Direct Peptide Reactivity Assay (DPRA). In this study, we optimized the decision-making framework of Spectro-DPRA utilizing a recursive partitioning algorithm trained on 54 reference chemicals with established

The research on health effects of olive oil and oleic acid remains variable. This umbrella review aimed to synthesize evidence from systematic reviews (with or without meta-analyses) regarding the health outcomes of oleic acid and olive oil, clarify the validity and strength of the evidence, and finally help to make dietary recommendations for different people. We conducted a comprehensive search of the Cochrane, PubMed, Web of Science, Medline, and Embase databases. These studies were assessed using AMSTAR 2 and GRADE tools. 19 out of 741 eligible studies were included, and the majority were

Ketone bodies, particularly β-hydroxybutyrate (BHB), have attracted attention for their anti-inflammatory and immunomodulatory properties. We previously demonstrated that nutritional interventions elevating circulating BHB levels suppress immediate-type allergic responses in dinitrofluorobenzene (DNFB)-induced allergic contact dermatitis (ACD). However, whether this effect is mediated by systemic BHB or via direct local activity in the skin remains unclear. Here, we investigated the effect of topical BHB application on mast cell degranulation and skin barrier function during an allergen challe

The concept of identifying threshold exposures to chemicals below which the risk to human health is negligible was originally proposed in the late 1960's. The induction of skin sensitization has been shown to demonstrate a dose response, and thus, a threshold concentration, or exposure, required to induce sensitization can be derived. The idea of a Dermal Sensitization Thresholds (DST) for chemicals has been investigated since 2008. However, DST developments have so far focused on individual sensitizing chemicals, rather than complex mixtures or botanical or naturally derived substances (BNS).

Background and ObjectiveNi is an important metal found in the human environment, including air, water, soil, food, kitchenware, and jewelry. It is widely used in electrical engineering, medicine, and industry. Ni compounds, whether insoluble (such as oxides and sulfides) or soluble (such as sulfates, chlorides, and acetates), pose serious risks to the environment, ecosystems, and human health. Such contamination can lead to harmful effects, especially on the respiratory, digestive, and skin systems. This study aims to emphasize the importance of early detection and diagnosis of Ni poisoning, a

Contact dermatitis is an inflammatory disease characterised by skin redness and swelling, oozing of fluid or pus and pain, caused by direct skin contact with an irritant or allergen. Its prevalence has skyrocketed worldwide, probably because human skin is in daily contact with an increasing gamut of irritants and allergens, which should require systematic surveillance. Large-scale testing is feasible by using organotypic and in silico skin models, which are becoming commonplace in industry and academic labs. In this work, we summarise the literature on models of contact dermatitis and provide

BACKGROUND: Skin barrier impairment is central to irritant (ICD) and allergic contact dermatitis (ACD). Stratum corneum (SC) components cholesterol sulphate (CholSulph), glucosylcholesterol (CholGlc) and natural moisturising factor (NMF) are critical for barrier function, but their changes in ICD and ACD remain underexplored.
OBJECTIVES: To measure CholSulph, CholGlc, NMF and IL-1α in patch-induced ICD and ACD and in hand dermatitis (HD) diagnosed as ICD or ACD.
METHODS: SC samples were collected from HD patients undergoing patch testing. Biomarkers were analysed in positive reactions to sod

BACKGROUND: In Europe, 1-2% of children are diagnosed with a peanut allergy. Atopic dermatitis (AD) is a significant risk factor for food allergy development, with cutaneous allergen exposure playing a causative role in allergic sensitization, particularly in early childhood.
OBJECTIVES: To investigate the primary and secondary skin-to-skin transfer of peanut proteins and the impact of hand washing in reducing allergen transfer.
METHODS: Primary transfer and secondary skin-to-skin transfer of fluorescently labelled peanut proteins was determined with and without hand washing, using two commo

Inflammatory skin conditions affect chronically a notable number of patients worldwide. Despite their prevalence, effective long-term treatments with minimal side effects remain limited. In this piece of work, three lipid-based nanocarriers were developed and applied topically to enhance the dermal delivery of two drugs, cyanocobalamin (vitamin B12) and cyclosporine A (CsA), with different physicochemical properties and negligible skin absorption (>500 Da). Previous reports show potential activity of both drugs to tackle inflammatory states. However, these studies provide only partial insights

Dear Editor, Contact allergic dermatitis (CAD) and urticaria are among the most common allergic diseases in dermatovenerology (1,2). In order to diagnose CAD, it is necessary to demonstrate contact sensitization through patch testing, the presence of dermatitis, and a clinically relevant exposure to a positive allergen (1). In comparison with CAD, acute urticaria most often do not require extensive diagnostic workup, unless food or drug hypersensitivity is suspected (2). Patients presenting at a hospital with eczema and/or hives often report that they have stopped using and or have experimente

Skin adhesives containing 2-octyl cyanoacrylate are a common source of allergic contact dermatitis (ACD), a complication of postoperative wound healing. Few studies have described postoperative cutaneous reactions to skin adhesives and their clinical management. The aim of this study was to review the incidence, descriptions, and clinical management of surgical site ACD caused by 2-ocyl cyanoacrylate. A comprehensive systematic literature search was conducted in September 2023 of the PubMed/MEDLINE (National Library of Medicine, Bethesda, MD), Web of Science (Clarivate Analytics, Philadelphia,

Psoriasis and allergic contact dermatitis (ACD) are the most common chronic inflammatory diseases, which are accompanied by epithelial alterations and a T cell-mediated immunopathology. In this study, we investigated the anti-ACD and anti-psoriasis effects of sea anemone Heteractis magnifica peptide HCRG21, a blocker of the TRPV1 channel, in 2,4-dinitrofluorobenzene (DNFB)- and imiquimod (IMQ)-induced mouse models, respectively. We found that topical application of 0.005-0.1% HCRG21 gels normalized hematological and immunological blood parameters in mice, significantly reduced the severity of
