Regulation of MRGPRX2-mediated mast cell function by competence-stimulating peptide 1 and pro-adrenomedullin peptide.

INTRODUCTION: Mas-related G protein-coupled receptor (GPCR)-X2 (MRGPRX2, mouse ortholog MrgprB2) is predominantly expressed in mast cells (MCs) and is activated by a wide range of cationic ligands such as bacterial quorum sensing molecules (QSMs) and pro-adrenomedullin peptide 9-20 (PAMP-12). Activation of MrgprB2 by competence-stimulating peptide-1 (CSP-1), a QSM produced by Gram-positive bacteria, has recently been shown to promote antibacterial immunity, whereas its activation by PAMP-12 has been implicated in allergic contact dermatitis (ACD) and itch. Mechanisms via which the activation o

Skin Sensitisation and Dermatitis Among Epoxy-Exposed Workers in a Wind Turbine Plant: Influence of Occupational Factors.

BACKGROUND: Epoxy polymers, widely used in industries such as wind turbine production, are common causes of allergic contact dermatitis (ACD). However, gaps remain mostly regarding the type of occupational exposure (specific tasks) and protective strategies. OBJECTIVES: To evaluate epoxy resin sensitisation in a wind turbine blade plant and assess its associations with tasks, medical history, clinical patterns, and personal protective equipment (PPE) use. METHODS: A cross-sectional study was performed in a large wind turbine blade facility. A group of volunteer workers completed a questionna

Comprehensive Patch Testing: An Essential Tool for Care of Allergic Contact Dermatitis.

Patch testing (PT) is the gold standard diagnostic test for allergic contact dermatitis (ACD) and is essential for guiding allergen avoidance. Comprehensive PT refers to the completion of PT for all potentially relevant and testable allergens for a given patient. It is necessary for most patients with contact allergy and can illustrate allergen trends and identify allergens that otherwise would be missed using a limited screening panel. Early identification of relevant allergens leads to clinical improvement, improved quality-of-life, and reduced health care utilization. This article illustrat

Skin inflammation and itch response are independently regulated by distinct nociceptor subsets.

Common dermatological disorders are characterized by inflammation and severe itching, which represent a significant clinical challenge. The mechanisms by which nociceptive sensory neurons (nociceptors) integrate peripheral signals to modulate inflammation and itch remain unclear. Here, we showed that two subsets of nociceptors had distinct and nonredundant biological functions in a model of allergic contact dermatitis (ACD). Retrograde axonal tracing of skin-projecting neurons combined with single-cell profiling identified a subset of MrgprD+ nonpeptidergic neurons that adopted a transitory re

Genetically predicted anti‑rubella virus IgG levels and dermatitis risk: A two‑sample Mendelian randomization study.

BACKGROUND: Dermatitis is a common inflammatory skin condition with multifactorial origins. Although immune responses following vaccination or natural infection have been linked to dermatological outcomes, the causal significance of virus-specific immune markers remains unclear. This study used Mendelian randomization (MR) to investigate whether genetically predicted virus-specific IgG levels are associated with dermatitis risk. METHODS: A two‑sample MR analysis was performed using genome‑wide association study (GWAS) summary statistics for IgG levels against eight common viral antigens from

Contact sensitisers activate keratinocytes and induce cytotoxicity via Transient Receptor Potential Ankyrin 1 in allergic contact dermatitis.

BACKGROUND AND PURPOSE: Allergic contact dermatitis (ACD) is a frequent inflammatory skin disease with limited therapeutic options. While neuronal Transient Receptor Potential Ankyrin 1 (TRPA1) has been implicated in ACD, the role of keratinocyte TRPA1 remains unclear. We investigated whether allergen binding to keratinocyte TRPA1 drives cytotoxicity, cytokine release and inflammatory amplification. EXPERIMENTAL APPROACH: We combined in vivo oxazolone-induced hypersensitivity in wild-type and Trpa1 knockout mice with pharmacological inhibition by HC-030031, in vitro assays using TRPA1-express

Influence of Atopic Dermatitis on the Dermoscopic Phenotype of Positive Patch Test Reactions: A Prospective Comparative Study.

Atopic dermatitis (AD) may alter the morphology of clinically positive allergic patch test reactions due to epidermal barrier dysfunction and a chronic inflammatory background, thereby complicating routine clinical interpretation. This prospective observational study aimed to evaluate the dermoscopic features of clinically positive allergic patch test reactions and compare their phenotypic expression in patients with and without concomitant AD. Consecutive patients undergoing routine patch testing were screened, and clinically positive reactions (1+, 2+, or 3+ according to ICDRG criteria) were

Cannabinoids for Dermatological Applications: Mechanistic Insights, Clinical Evidence, and Emerging Nanotechnology-Enabled Delivery Strategies.

Cannabinoids (CBs) derived from Cannabis sativa have attracted growing interest for dermatological applications due to their anti-inflammatory, antiproliferative, antimicrobial, antifibrotic, and antipruritic properties. However, their clinical translation is significantly limited by physicochemical and pharmacokinetic challenges, including poor aqueous solubility, lipophilicity, instability, variable skin penetration, and inconsistent bioavailability. At the molecular level, CBs modulate keratinocyte proliferation, sebocyte activity, fibroblast function, melanocyte balance, and immune signall
