Study on transdermal delivery system of sulforaphane chitosan nanoparticles based on methacryloylated chitosan microneedles: application in the treatment of irritant contact dermatitis.

Treatment of irritant contact dermatitis (ICD) is limited by the stratum corneum barrier, which hinders drug delivery and bioavailability. Sulforaphane (SFN) offers anti-inflammatory potential but suffers from poor stability and a short half-life. To address this, we developed a chitosan methacrylate (CSMA) hydrogel microneedle (MN) patch loaded with SFN-chitosan nanoparticles (SFN-CSNPs) for enhanced transdermal delivery.Network pharmacology identified 33 potential ICD-related targets of SFN, including SRC, EGFR, and JAK2, implicating pathways such as AGE-RAGE and Relaxin signaling. SFN-CSNPs

Reframing Eczema: Th2-Skewed Contact Sensitization, Atopy Patch Testing, and Systemic Contact Dermatitis.

PURPOSE OF REVIEW: We review allergic contact dermatitis (ACD) with predominant Th2 type cytokine expression in the context of chronic cutaneous inflammation. While more has been written about Th1 skewed ACD due to potent allergens in the setting of healthy skin, this review highlights recognition of Th2 skewed ACD in both atopic dermatitis and systemic contact dermatitis and the role of allergen avoidance as an alternative to systemic therapies. RECENT FINDINGS: Th2 skewed ACD rarely occurs to potent allergens. It more commonly occurs in response to allergens considered “non-sensitizers” in t

Double-blind randomised controlled trial of an emollient cream with and without 1 % supercritical CO2 extract of Calendula officinalis in contact dermatitis.

This double-blind randomised controlled trial aimed to evaluate the efficacy of an emollient cream with and without 1 % supercritical CO2 extract of Calendula officinalis in contact dermatitis. Twenty healthy volunteers without pre-existing dermatological conditions participated in this single-centre study. Each participant's forearm was divided into three test sites: control (no treatment), placebo (base emollient), and intervention (calendula cream). The study employed random allocation of test sites using Microsoft Excel software. Both investigators and participants were blinded to the trea

Requirements for Alternative In Vitro and In Silico Skin Models of Irritant and Allergic Contact Dermatitis.

Contact dermatitis is an inflammatory disease characterised by skin redness and swelling, oozing of fluid or pus and pain, caused by direct skin contact with an irritant or allergen. Its prevalence has skyrocketed worldwide, probably because human skin is in daily contact with an increasing gamut of irritants and allergens, which should require systematic surveillance. Large-scale testing is feasible by using organotypic and in silico skin models, which are becoming commonplace in industry and academic labs. In this work, we summarise the literature on models of contact dermatitis and provide

A 21-Year Perspective on Occupational Skin and Respiratory Diseases Among Food Handlers.

BACKGROUND: Food handlers may have an increased risk of developing occupational skin and respiratory diseases. METHODS: This retrospective study was based on examinations, skin prick testing, and patch testing performed at the Unit of Occupational Medicine at the University of Trieste (N-E Italy) between 2002 and 2022 in food-handler workers referred to the unit for suspected occupational allergic diseases. RESULTS: More than half of the population (58.1%) experienced occupational skin diseases, with a higher prevalence among women (OR 3.3, 95% CI 1.5-7.6). Irritant contact dermatitis was th

Skin Barrier Biomarkers in Patch-Induced and Clinical Allergic and Irritant Contact Dermatitis.

BACKGROUND: Skin barrier impairment is central to irritant (ICD) and allergic contact dermatitis (ACD). Stratum corneum (SC) components cholesterol sulphate (CholSulph), glucosylcholesterol (CholGlc) and natural moisturising factor (NMF) are critical for barrier function, but their changes in ICD and ACD remain underexplored. OBJECTIVES: To measure CholSulph, CholGlc, NMF and IL-1α in patch-induced ICD and ACD and in hand dermatitis (HD) diagnosed as ICD or ACD. METHODS: SC samples were collected from HD patients undergoing patch testing. Biomarkers were analysed in positive reactions to sod

Hypersensitivity Reaction to Detergents - A Myth or Reality?

Dear Editor, Contact allergic dermatitis (CAD) and urticaria are among the most common allergic diseases in dermatovenerology (1,2). In order to diagnose CAD, it is necessary to demonstrate contact sensitization through patch testing, the presence of dermatitis, and a clinically relevant exposure to a positive allergen (1). In comparison with CAD, acute urticaria most often do not require extensive diagnostic workup, unless food or drug hypersensitivity is suspected (2). Patients presenting at a hospital with eczema and/or hives often report that they have stopped using and or have experimente

Blockade of interleukin-13 signalling improves skin barrier function and biology in patients with moderate-to-severe atopic dermatitis.

BACKGROUND: Interleukin (IL)-13 is a key driver of inflammation and barrier dysfunction in atopic dermatitis (AD). While there is robust evidence that tralokinumab - a monoclonal antibody that neutralizes IL-13 - reduces inflammation and clinical disease activity, less is known about its effects on barrier function. OBJECTIVES: To characterize the effects of tralokinumab treatment on skin barrier function. METHODS: Transepidermal water loss (TEWL), stratum corneum hydration (SCH), natural moisturizing factor content, histopathological characteristics, biomarker expression and microbiome comp

Development of Sensitive In Vitro Protocols for the Biocompatibility Testing of Medical Devices and Pharmaceuticals Intended for Contact with the Eyes: Acute Irritation and Phototoxicity Assessment.

This study introduces a novel in vitro methodology that employs the 3-D reconstructed tissue model, EpiOcular, to assess the irritation and phototoxicity potential of medical devices and drugs in contact with the eye. Our study evaluated diverse test materials, including medical devices, ophthalmological solutions and an experimental drug (cemtirestat), for their potential to cause eye irritation and phototoxicity. The protocols used in this study with the EpiOcular tissue model were akin to those used in the ultra-mildness testing of cosmetic formulations, which is challenging to predict with

Phototoxic reaction to oral terbinafine due to Tinea capitis in a child.

We report the case of an 18-month-old boy who developed a phototoxic skin reaction to terbinafine on his scalp, ears, and face in the form of disseminated erythematous plaques, which resembled subacute lupus erythematosus (SCLE) in their clinical presentation. Skin changes appeared a short time after the boy was exposed to sunlight during the period of time when he was treated with oral terbinafine due to Microsporum canis fungal scalp infection. Tinea capitis is a common dermatophyte infection primarily affecting prepubertal children (1). Microsporum canis remains the predominant causative or

Nitric Oxide Signal Transduction and Its Role in Skin Sensitization.

Nitric oxide (NO) is a signaling molecule that plays a crucial role in numerous cellular physiological processes. In the skin, NO is produced by keratinocytes, fibroblasts, endothelial cells, and immune cells and is involved in skin functions such as vasodilation, pigmentation, hair growth, wound healing, and immune responses. NO modulates both innate and adaptive immune responses. As a signaling molecule and cytotoxic effector, NO influences the function of immune cells and production of cytokines. NO is a key mediator that protects against or contributes to skin inflammation. Moreover, NO ha

Humulus japonicus ameliorates irritant contact dermatitis by suppressing NF‑κB p65‑dependent inflammatory responses in mice.

As a type of contact dermatitis (CD), irritant CD (ICD) is an acute skin inflammation caused by external irritants, such as soap, water and chemicals. Humulus japonicus (HJ) is a herbal medicine widely distributed in Asian countries and has anti-inflammatory, antimicrobial and antioxidant effects. The current study aimed to investigate the anti-dermatitis effect of HJ on ICD and determine the molecular basis of this effect using 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced dermatitis mice models and lipopolysaccharide (LPS)-stimulated RAW264.7 cells. Mice were orally administered HJ and

Properties and safety of topical dihydroxyacetone in sunless tanning products: A review.

Sunless tanning products have risen in popularity as the desire for a tanned appearance continues alongside growing concerns about the deleterious effects of ultraviolet radiation exposure from the sun. Dihydroxyacetone (DHA) is a simple carbohydrate found nearly universally in sunless tanning products that serves to impart color to the skin. The Food and Drug Administration (FDA), which regulates sunless tanning products as cosmetics, allows DHA for external use while maintaining that its ingestion, inhalation, or contact with mucosal surfaces should be avoided. Given its widespread use and a

Vitamin D supplementation in patients with atopic dermatitis, chronic urticaria and contact irritant and allergic dermatitis - possible improvement without risk.

Introduction: There has been a lot of talk lately about the importance of reduced serum vitamin D levels and their supplementation for patients with inflammatory skin diseases such as atopic dermatitis (AD) and other allergic diseases. Serum vitamin D values are associated with a number of factors such as limited sunlight exposure (modern lifestyle, extended indoor stay, enhanced sun protection, etc.) which can affect different diseases. Aim: To evaluate serum vitamin D values in patients with inflammatory skin diseases, comparing them on the basis of other parameters (age, gender/sex, residen
