Dermatotoxicity is the skin's response to chemical substances that produce damage. Penetration of the chemical to the various skin layers is important and may result in irritation, allergic contact dermatitis, contact urticaria, one of various forms of photosensitization, or other cutaneous reactions. The testing of new products or industrial chemicals for potential dermatotoxicity is an important process in protecting the public's health.

A review of medical surveillance records was undertaken for 197 personnel who had been assigned to an ethyleneamines (EA) production unit for one or more months between 1947 and 1983. A topical application of a chalcone derivative, 4,2',4'-trihydroxychalcone (isoliquiritigenin) inhibited epidermal ornithine decarboxylase (ODC) induction and ear edema formation, i.e. inflammation, caused by a topical application of 12-O-tetradecanoylphorbol-13-acetate (TPA) in CD-1 mice. 3851 consecutive patients patch tested between January 1985 and March 1990 have been analysed for rubber allergies. The incidence of rubber allergy was 3.8% (n = 145). In 80/145 patients (55%), the source of rubber sensitization was occupational, 67 of whom (84%) had acquired allergy from wearing rubber gloves at work. Euxyl K 400 is a preservative for cosmetics and toiletries containing 2 active ingredients, 1,2-dibromo-2,4-dicyanobutan and 2-phenoxyethanol. 2057 consecutive patients with contact dermatitis were patch tested with Euxyl K 400 2.5% pet. and ethanol. We need to improve education of farm workers and their families to the potential hazards of exposure to the chemicals and other agents that they are in contact with on a daily basis. Of a total of 3731 patients investigated between 1974 and 1990, 1844 (49.4%) had an occupational skin disease. We found m-xylylene diamine, a widespread additive in plastics, to be a very potent sensitizer. Patch tests with other diamines revealed in all four patients allergic cross reactions to benzylamine. Two persons had also acquired an additional allergy to ethylenediamine. In a 59-year-old man, the allergic contact dermatitis provoked the first manifestation of an atopic dermatitis.
