Cardiovascular, Metabolic and Endocrine, Cancer, Mortality, Derma, and Other Outcomes of Olive Oil and Oleic Acid: An Umbrella Review.

The research on health effects of olive oil and oleic acid remains variable. This umbrella review aimed to synthesize evidence from systematic reviews (with or without meta-analyses) regarding the health outcomes of oleic acid and olive oil, clarify the validity and strength of the evidence, and finally help to make dietary recommendations for different people. We conducted a comprehensive search of the Cochrane, PubMed, Web of Science, Medline, and Embase databases. These studies were assessed using AMSTAR 2 and GRADE tools. 19 out of 741 eligible studies were included, and the majority were

Topical β-hydroxybutyrate suppresses allergic dermatitis via HCAR2-CaMKK-AMPK-mediated barrier protection.

Ketone bodies, particularly β-hydroxybutyrate (BHB), have attracted attention for their anti-inflammatory and immunomodulatory properties. We previously demonstrated that nutritional interventions elevating circulating BHB levels suppress immediate-type allergic responses in dinitrofluorobenzene (DNFB)-induced allergic contact dermatitis (ACD). However, whether this effect is mediated by systemic BHB or via direct local activity in the skin remains unclear. Here, we investigated the effect of topical BHB application on mast cell degranulation and skin barrier function during an allergen challe

Development of a skin sensitization Site-of-Contact Exposure Threshold (SoCET) for botanicals and natural substances.

The concept of identifying threshold exposures to chemicals below which the risk to human health is negligible was originally proposed in the late 1960's. The induction of skin sensitization has been shown to demonstrate a dose response, and thus, a threshold concentration, or exposure, required to induce sensitization can be derived. The idea of a Dermal Sensitization Thresholds (DST) for chemicals has been investigated since 2008. However, DST developments have so far focused on individual sensitizing chemicals, rather than complex mixtures or botanical or naturally derived substances (BNS).

Nickel; a metal with threats to human health, focusing on its intoxication mechanisms.

Background and ObjectiveNi is an important metal found in the human environment, including air, water, soil, food, kitchenware, and jewelry. It is widely used in electrical engineering, medicine, and industry. Ni compounds, whether insoluble (such as oxides and sulfides) or soluble (such as sulfates, chlorides, and acetates), pose serious risks to the environment, ecosystems, and human health. Such contamination can lead to harmful effects, especially on the respiratory, digestive, and skin systems. This study aims to emphasize the importance of early detection and diagnosis of Ni poisoning, a

Requirements for Alternative In Vitro and In Silico Skin Models of Irritant and Allergic Contact Dermatitis.

Contact dermatitis is an inflammatory disease characterised by skin redness and swelling, oozing of fluid or pus and pain, caused by direct skin contact with an irritant or allergen. Its prevalence has skyrocketed worldwide, probably because human skin is in daily contact with an increasing gamut of irritants and allergens, which should require systematic surveillance. Large-scale testing is feasible by using organotypic and in silico skin models, which are becoming commonplace in industry and academic labs. In this work, we summarise the literature on models of contact dermatitis and provide

A 21-Year Perspective on Occupational Skin and Respiratory Diseases Among Food Handlers.

BACKGROUND: Food handlers may have an increased risk of developing occupational skin and respiratory diseases. METHODS: This retrospective study was based on examinations, skin prick testing, and patch testing performed at the Unit of Occupational Medicine at the University of Trieste (N-E Italy) between 2002 and 2022 in food-handler workers referred to the unit for suspected occupational allergic diseases. RESULTS: More than half of the population (58.1%) experienced occupational skin diseases, with a higher prevalence among women (OR 3.3, 95% CI 1.5-7.6). Irritant contact dermatitis was th

Skin Barrier Biomarkers in Patch-Induced and Clinical Allergic and Irritant Contact Dermatitis.

BACKGROUND: Skin barrier impairment is central to irritant (ICD) and allergic contact dermatitis (ACD). Stratum corneum (SC) components cholesterol sulphate (CholSulph), glucosylcholesterol (CholGlc) and natural moisturising factor (NMF) are critical for barrier function, but their changes in ICD and ACD remain underexplored. OBJECTIVES: To measure CholSulph, CholGlc, NMF and IL-1α in patch-induced ICD and ACD and in hand dermatitis (HD) diagnosed as ICD or ACD. METHODS: SC samples were collected from HD patients undergoing patch testing. Biomarkers were analysed in positive reactions to sod

Mechanistic in vivo studies of secondary skin-to-skin peanut allergen transfer to determine the impact of hand washing in reducing the risk of cutaneous sensitization.

BACKGROUND: In Europe, 1-2% of children are diagnosed with a peanut allergy. Atopic dermatitis (AD) is a significant risk factor for food allergy development, with cutaneous allergen exposure playing a causative role in allergic sensitization, particularly in early childhood. OBJECTIVES: To investigate the primary and secondary skin-to-skin transfer of peanut proteins and the impact of hand washing in reducing allergen transfer. METHODS: Primary transfer and secondary skin-to-skin transfer of fluorescently labelled peanut proteins was determined with and without hand washing, using two commo

Ultraflexible lipid vesicles loaded with cyanocobalamin or cyclosporin A downregulate acute and chronic immuno-inflammatory skin states.

Inflammatory skin conditions affect chronically a notable number of patients worldwide. Despite their prevalence, effective long-term treatments with minimal side effects remain limited. In this piece of work, three lipid-based nanocarriers were developed and applied topically to enhance the dermal delivery of two drugs, cyanocobalamin (vitamin B12) and cyclosporine A (CsA), with different physicochemical properties and negligible skin absorption (>500 Da). Previous reports show potential activity of both drugs to tackle inflammatory states. However, these studies provide only partial insights

Hypersensitivity Reaction to Detergents - A Myth or Reality?

Dear Editor, Contact allergic dermatitis (CAD) and urticaria are among the most common allergic diseases in dermatovenerology (1,2). In order to diagnose CAD, it is necessary to demonstrate contact sensitization through patch testing, the presence of dermatitis, and a clinically relevant exposure to a positive allergen (1). In comparison with CAD, acute urticaria most often do not require extensive diagnostic workup, unless food or drug hypersensitivity is suspected (2). Patients presenting at a hospital with eczema and/or hives often report that they have stopped using and or have experimente

Contact Urticaria and Related Conditions: Clinical Review.

Contact urticaria (CoU) is an immediate contact reaction occurring within minutes to an hour after exposure to specific proteins or chemicals. CoU is categorised into non-immunologic (NI-CoU) and immunologic (I-CoU) types, with I-CoU potentially leading to anaphylaxis. Both forms of CoU can be associated with protein contact dermatitis and the CoU syndrome. Patients with I-CoU may also have other type I (immediate) allergic diseases, such as allergic conjunctivitis, rhinitis, asthma or food allergy. This review provides a detailed overview of CoU and related conditions, focusing on triggers, d

Is Food-Triggered Atopic Dermatitis a Form of Systemic Contact Dermatitis?

Food allergy in atopic dermatitis is mediated by complex immune interactions between genetics, diet, environment, and the microbiome. When contact between inflamed skin and food antigens occurs, contact hypersensitivity can develop. Consequently, systemic contact dermatitis (SCD) can occur after ingestion of allergenic foods or food additives in the setting of a Th2 response with CLA-positive T cells, triggering dermatitis where skin resident memory lymphocytes reside. This phenomenon explains food-triggered dermatitis. Atopy patch tests (APTs) detect sensitization to food proteins responsible

Revitalizing Photoaging Skin through Eugenol in UVB-Exposed Hairless Mice: Mechanistic Insights from Integrated Multi-Omics.

Chronic ultraviolet (UV) exposure causes photoaging, which is primarily responsible for skin damage. Nutritional intervention is a viable strategy for preventing and treating skin photoaging. Eugenol (EU) presents anti-inflammatory and antioxidant properties, promotes wound healing, and provides contact dermatitis relief. This study explored the ability of EU to mitigate skin photoaging caused by UVB exposure in vitro and in vivo. EU alleviated UVB-induced skin photodamage in skin cells, including oxidative stress damage and extracellular matrix (ECM) decline. Dietary EU alleviated skin photoa

Sunscreens part 2: Regulation and safety.

The second part of this CME article discusses sunscreen regulation and safety considerations for humans and the environment. First, we provide an overview of the history of the US Food and Drug Administration's regulation of sunscreen. Recent Food and Drug Administration studies clearly demonstrate that organic ultraviolet filters are systemically absorbed during routine sunscreen use, but to date there is no evidence of associated negative health effects. We also review the current evidence of sunscreen's association with vitamin D levels and frontal fibrosing alopecia, and recent concerns re
