Efficacy and Safety of Topical and Systemic Therapies for Atopic Dermatitis in Patients With Skin of Colour: A Systematic Review.

Atopic dermatitis (AD) is more common and often more severe in people with skin of colour, yet the trials that shaped modern treatment enrolled mostly White participants. The disease can look different in darker skin, and erythema-based severity scores may miss active inflammation. Whether current topical and systemic drugs work as well, and are as safe, across racial, ethnic, and Fitzpatrick skin-type groups, is still uncertain. This review brought together the evidence on AD treatment outcomes reported by skin colour. Preferred Reporting Items for Systematic reviews and Meta-Analyses (PRISMA

Atopy is associated with osteoarthritis severity: results from the KHOALA cohort.

OBJECTIVE: Atopy has been identified as a risk factor for osteoarthritis, but without specifying its impact on disease severity. We sought to investigate the association between atopy and the symptomatic and structural severity of knee and hip osteoarthritis. METHODS: We used data from KHOALA, a French multicentre cohort of patients with knee and/or hip osteoarthritis (OA) investigating "atopic" subjects, defined as those with history of asthma and/or atopic dermatitis (AD) compared to "non-atopic" subjects. We compared: WOMAC pain, stiffness and function scores; and the proportion of having

Efficacy of Dupilumab in Treating Trachyonychia and Other Nail Dystrophies: A Systematic Review of Case Reports.

Trachyonychia and related nail dystrophies are often associated with inflammatory skin diseases such as atopic dermatitis, psoriasis, and alopecia areata. Management remains difficult, as standard therapies frequently yield limited benefit. Dupilumab, an IL-4Rα antagonist, is effective for Th2-driven conditions, but its role in nail dystrophies has not been systematically assessed. This review evaluates the existing evidence on dupilumab for trachyonychia and related nail disorders. A systematic search of PubMed, Wiley, Web of Science, MEDLINE, Google Scholar, and EBSCO was performed from ince

Convergence in Pruning and Circadian Regulation in Obsessive-Compulsive Disorder and Allergic Diseases: Evidence From a Secondary Genome-Wide Analysis.

BACKGROUND: Obsessive-compulsive disorder (OCD) and allergic diseases co-occur far more often than expected, yet genome-wide genetic correlations are typically modest or negative. The objective of this study was to test whether OCD and allergic diseases show convergence at the level of pre-specified biological pathways, especially synaptic pruning and circadian regulation, rather than at the level of broad genome-wide correlation. METHODS: Using the Multi-marker Analysis of GenoMic Annotation (MAGMA) pipeline, we re-analysed summary statistics from a large allergic-diseases genome-wide associ

Severe Cutaneous Adverse Reactions Associated With Newer-Generation Antiseizure Medications: A Real-World Pharmacovigilance Study Based on FAERS and JADER.

BACKGROUND: Severe cutaneous adverse reactions (SCARs) are rare but potentially fatal immune-mediated toxicities associated with antiseizure medications (ASMs). Phenotype-resolved post-marketing safety profiles for newer-generation ASMs have not been comprehensively characterized across pharmacovigilance systems. METHODS: A cross-database disproportionality analysis was conducted using FAERS and JADER (January 2004-September 2025). Twenty-two newer-generation ASMs coded as primary-suspect drugs were included; outcomes were the four SCAR phenotypes (Stevens-Johnson syndrome, SJS; toxic epiderm

Reporting of concomitant and rescue topical therapies in randomized controlled trials evaluating a systemic treatment in atopic dermatitis: a scoping review.

BACKGROUND: Randomized controlled trials (RCTs) evaluating new systemic treatments for atopic dermatitis (AD) have increased dramatically over the last decade. These trials often incorporate topical therapies either as permitted concomitant or rescue treatments. Differential use of these topicals postrandomization introduces potential bias as they may nullify or exaggerate treatment responses. OBJECTIVES: To determine the proportion of RCTs that clearly report the allowance or prohibition of concomitant and rescue topical treatments; and to examine the reporting of specific key parameters for

Systematic review of the use of the Dermatology Life Quality Index in routine clinical practice: evidence from 287 articles across 56 countries.

BACKGROUND: Although quality of life instruments are widely used in research, it is challenging to find evidence of their use in routine clinical use. The most widely used measure for skin disease burden is the Dermatology Life Quality Index (DLQI), and its scores have validated clinical meaning. OBJECTIVES: To identify evidence of the use of the DLQI in routine clinical practice and explore the nature of its use. METHODS: The study followed PRISMA guidelines, and the protocol was registered with PROSPERO. MEDLINE (Ovid), Embase, Scopus and CINAHL (EBSCO) databases were systematically search

Ensuring the Safe Use of Bee Products: A Review of Allergic Risks and Management.

Honeybee products (HBPs), including honey, bee pollen, bee bread, royal jelly, propolis, beeswax, and bee brood, are increasingly used in food, nutraceutical, and cosmetic contexts. Because of their natural origin, HBPs can provoke allergic reactions ranging from localised dermatitis to life-threatening, systemic anaphylaxis. As the use of bee products for health purposes grows in apitherapy (a branch of alternative medicine), raising public awareness of their potential risks is essential. This narrative review synthesises the clinical manifestations of HBP allergy, culprit allergens present i

Multicentre, prospective, single-arm, non-controlled, open-label trial to evaluate the safety and efficacy of live attenuated influenza vaccine in paediatric patients with atopic dermatitis undergoing dupilumab therapy: a protocol.

INTRODUCTION: Atopic dermatitis (AD) is a chronic inflammatory skin condition that impairs the quality of life of affected paediatric patients and their families. Dupilumab, an antagonist of the shared alpha chain subunit of the cytokines interleukin-4 and interleukin-13, has revolutionised the management of moderate-to-severe AD by effectively targeting type 2 inflammation. However, live attenuated vaccines, including live attenuated influenza vaccines (LAIVs), are contraindicated during dupilumab therapy owing to limited safety data. This restriction poses challenges to immunisation strategi

Skin care by washing with water is not inferior to washing with a cleanser in children with atopic dermatitis in remission in summer: WASH study.

BACKGROUND: Washing with water is not inferior to washing with soaps and detergents in children with atopic dermatitis (AD) in remission during the fall-winter seasons. We investigated whether this finding varies during summer based on the type of cleanser (soaps and detergents). METHODS: This evaluator-blinded, pragmatic, randomized, and non-inferiority study enrolled patients with AD whose eczema was controlled following regular steroid ointment application 2 days/week. For 8 ± 4 weeks, participants washed their upper and lower limbs with a cleanser on one side and with water alone on the o

Blood Cells Count Derived Inflammation Indexes as Predictors of Early Treatment Response to Dupilumab in Patients with Moderate-to-Severe Atopic Dermatitis.

Derived inflammatory indexes from routine hematological parameters might be useful for predicting early-response vs. late/non-response to dupilumab, the first biological agent approved for moderate-to-severe atopic dermatitis (AD). We tested this hypothesis by retrospectively investigating the association between pre-specified baseline inflammatory indexes and dupilumab response (≥50% reduction in the Eczema Area and Severity Index, EASI 50) at 4 and 16 weeks in a consecutive series of 66 AD patients (38 males and 28 females). Forty-six patients (69.7%) were early-responders at 4 weeks, wherea

Feasibility and Tolerability of Adjuvant Capecitabine-Based Chemoradiation in Patients With Breast Cancer and Residual Disease After Neoadjuvant Chemotherapy: A Prospective Clinical Trial.

PURPOSE: Addition of adjuvant capecitabine improves overall survival for patients with breast cancer lacking pathologic complete response to standard-of-care neoadjuvant chemotherapy. Combining radiosensitizing capecitabine concurrent with radiation may further improve disease control, although the feasibility and tolerability of chemoradiation in this setting is unknown. This study aimed to determine the feasibility of this combination. Secondary objectives included the effect of chemoradiation on physician-reported toxicity, patient-reported skin dermatitis, and patient-reported quality of l
