AA in Rheumatoid Arthritis and Lupus.

Inflammation and immune dysregulation are at the center of various diseases, including autoimmune diseases, especially rheumatoid arthritis (RA) and systemic lupus erythematosus (lupus). In general, corticosteroids, disease-modifying drugs, and monoclonal antibodies against IL-6 and TNF-α, JAK inhibitors, and various other immunomodulators are extensively used in the treatment of RA and lupus. It is known that EFA (essential fatty acid) metabolism, especially that of AA, is altered in RA and lupus. It is proposed that metabolism of AA and of other PUFAs may play a role in predicting disease pr

Quantification of Chikungunya Virus RNA by Real-Time RT-PCR Assay.

RT-qPCR is characterized by‌ high sensitivity, strong specificity, excellent reproducibility, and operational simplicity, ‌enabling‌ rapid quantification of viral RNA within a short timeframe. The real-time PCR system relies upon the measurement of a fluorescent reporter during PCR, in which the amount of emitted fluorescence is directly proportional to the amount of the PCR product in a reaction. Here, we describe the use of TaqMan® probe-based one-tube Real-time reverse transcription quantitative polymerase chain reaction (RT-qPCR) for the detection and quantification of the Chikungunya viru

Hip range of motion is differentially associated with pain sensitivity in individuals with and without knee osteoarthritis.

BACKGROUND: Knee osteoarthritis (KOA) is a multifactorial condition influenced by both mechanical and neurophysiological factors. Hip joint mobility may be associated to knee joint loading and pain processing; however, the relationships between hip range of motion (ROM), pain sensitivity, and muscle torque in individuals with KOA remain unclear. This study aimed to investigate the association between knee pain sensitivity and hip rotator ROM in individuals with and without KOA. Additionally, it examined the association between hip rotation ROM and muscle torque and compared hip ROM between gro

Discordance in Treatment Response Assessment Between Clinicians and Patients With Skin Chronic Graft-vs-Host Disease.

IMPORTANCE: Clinician-reported and patient-reported outcomes are critical measures of therapeutic efficacy in cutaneous chronic graft-vs-host disease (cGVHD) but are not always correlated. Discordance in treatment response between clinicians and patients hinders interpretation of outcomes in clinical trials and complicates therapeutic decision-making in clinical practice. OBJECTIVE: To identify factors associated with discordance in clinician-reported and patient-reported treatment response assessments and to evaluate the association of clinician-reported and patient-reported responses with s

Association between depressive symptoms and thoracic aortic calcification, and their independent and joint prognostic value for incident atherosclerotic cardiovascular disease in the MESA cohort.

Depression is highly prevalent; however, its association with atherosclerotic cardiovascular disease (ASCVD) events remains incompletely understood. This study aimed to evaluate the association between depressive symptoms and thoracic aortic calcification (TAC) and assess their joint prognostic value for future ASCVD events. We analyzed 2,600 participants from the Multi-Ethnic Study of Atherosclerosis (MESA) without baseline ASCVD. TAC was quantified using non-contrast chest CT as a marker of subclinical atherosclerosis. Depressive symptoms were assessed using the Center for Epidemiologic Stud

Effects of an aromatherapy-integrated exercise program on knee pain and physical function in community-dwelling older adults with osteoarthritis.

Osteoarthritis (OA) is a major cause of chronic pain in older adults and negatively affects physical function and daily life. Aromatherapy is a complementary therapy with potential pain-relieving effects. However, evidence supporting its use alongside exercise in older adults is limited. In this single-blind randomized controlled trial, participants with knee pain ≥ 40 mm on the visual analog scale (VAS) were assigned to either an aromatherapy combined with exercise group (ACG) or an exercise control group (ECG). Both groups participated in 60-minute rehabilitation sessions twice weekly for 8

Preoperative coronal plane alignment of the knee phenotype does not predict patient-reported outcomes after medial opening-wedge high tibial osteotomy.

INTRODUCTION: Phenotype-based classification systems have been proposed to characterize coronal alignment and joint-line orientation and to support individualized correction strategies. Although developed primarily for arthroplasty, they are increasingly evaluated in osteotomy populations. This study assessed whether preoperative coronal plane alignment of the knee (CPAK) phenotype predicts patient-reported outcome measures (PROMs) after medial opening-wedge high tibial osteotomy (MOWHTO). METHODS: This retrospective cohort study included 130 knees in 111 patients undergoing valgus-producing

The crossing-sign angle and trochlear dysplasia length: New radiographic parameters to measure the dysplastic trochlea.

INTRODUCTION: Trochlear dysplasia is one of the most relevant anatomic factors predisposing patients to patellofemoral instability and its recurrence. This study introduces two novel radiographic measurements: the crossing-sign angle and the trochlear dysplasia length. METHODS: A concordance study was conducted in patients aged 10-40 years with at least one episode of patellofemoral dislocation and radiographic evidence of trochlear dysplasia on a true lateral knee radiograph. Three observers independently measured the crossing-sign angle, the trochlear dysplasia length, and bump height, as w

Single Versus Double Pin Fixation of the Calcaneocuboid Joint During Calcaneal Lengthening Osteotomy for Pediatric Idiopathic Flatfoot.

BACKGROUND: Calcaneocuboid (CC) joint subluxation is a recognized complication after calcaneal lengthening osteotomy for pediatric idiopathic flexible flatfoot, and temporary pin fixation is often performed to prevent it. This study compared postoperative radiologic outcomes at 2 years of single- and double-pin fixation of the CC joint. METHODS: We retrospectively reviewed 50 feet from 30 patients aged 9 to 15 years who underwent calcaneal lengthening osteotomy for idiopathic flexible flatfoot with a follow-up of at least 2 years. Fourteen feet (28%) underwent single-pin fixation, and 36 (72%

Exploring the Role of Hyperfocus in the Psychophysiology of Probable Bruxism and Painful Temporomandibular Disorders.

BACKGROUND: Bruxism and temporomandibular disorders (TMD) are multifactorial conditions in which cognitive and behavioural traits may play an underrecognised role. Hyperfocus, defined as intense and sustained attentional engagement, has been proposed as a potential driver of heightened sensorimotor activity and pain vulnerability, yet its relevance to bruxism and painful TMD remains poorly understood. OBJECTIVES: To examine whether hyperfocus is associated with probable bruxism and painful TMD (pTMD), and to determine whether probable bruxism mediates the relationship between hyperfocus and p
