Rice spikelet rot disease (RSRD) threatens food safety by contaminating grains with diverse mycotoxins. This study revealed a climate-driven shift in pathogen dominance from Alternaria spp. to Fusarium spp., determining associated toxin chemotypes. Untargeted LC-MS metabolomics of wheat infected by Fusarium graminearum (wild-type vs ΔTri5) identified host-derived metabolites, 5-hydroxyindole-3-acetic acid (5-HIAA) and rutin, as key toxin-suppressing candidates. Both compounds strongly inhibited mycotoxin biosynthesis (>95%) with minimal antifungal activity. Mechanistic analyses showed that rut

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a prevalent disorder characterized by excessive hepatic lipid accumulation, frequently progressing to steatohepatitis, fibrosis, and cirrhosis. Transcription factor EB (TFEB) coordinates lysosomal biogenesis, autophagy, and lipid catabolism, and therefore represents a potential therapeutic target; however, safe and direct strategies for selectively enhancing hepatic TFEB activity remain limited. Here, liver-targeted lipid nanoparticles (LNPs) were developed to deliver mRNAs encoding wild-type TFEB or the constitutively active

INTRODUCTION: Cognitive impairment is a common non-motor symptom of Parkinson's disease (PD) that markedly reduces patients' quality of life. Given the clinical heterogeneity of PD-related cognitive impairment (PDCI), further clarification of clinical and circulating biological factors associated with cognitive status may help improve our understanding of cognitive dysfunction in PD.
METHODS: In this cross-sectional study, 243 patients with PD and 90 age- and sex-matched healthy controls (HCs) were enrolled. Cognitive function was evaluated using the Montreal Cognitive Assessment (MoCA), and

BACKGROUND: Accurate preoperative characterization of intraarticular abnormalities is important when planning hip-preserving surgery in young, active patients. Lesions of the ligamentum teres and the associated acetabular fossa and fovea capitis (forming the ligamentous-fossa-foveolar complex [LFFC]) are frequently observed intraoperatively, although their relationship with hip pain in patients without osteoarthritis remains uncertain. These lesions have received limited attention in preoperative imaging.
QUESTIONS/PURPOSES: (1) What is the intraobserver and interobserver reliability of tract

BACKGROUND: Sjögren's disease is a complex, systemic autoimmune disease with no approved targeted therapies. Ianalumab is an IgG1 monoclonal antibody with a dual mechanism of action, depleting B cells through antibody-dependent cellular cytotoxicity and inhibiting B-cell activation and survival through blockade of B-cell activating factor receptor.
METHODS: We conducted two 52-week, double-blind, randomized, placebo-controlled trials involving patients with active Sjögren's disease. Patients in the NEPTUNUS-1 trial were assigned in a 1:1 ratio to receive subcutaneous ianalumab (300 mg monthly

Psoriasis is a chronic autoimmune skin disease characterized by persistent oxidative stress and sustained inflammation, affecting approximately 1-8% of the global population, with no curative therapy has been well-established to date. To address this, we developed a bioactive hybrid nanocomposite (Exo-Ce) by chemically conjugating M2 macrophage-derived exosomes with cerium oxide nanoparticles (CeNP), which was applied for synergistic and immunomodulatory therapy of psoriasis. This nanohybrid system effectively integrates the natural immunomodulatory properties of exosomes with the reliable ant

Ferroptosis is an iron-dependent, non-apoptotic form of programmed cell death closely associated with various diseases. It is characterized by intracellular lipid peroxidation (LPO) and glutathione depletion. The process of lipid peroxidation is regulated by reactive oxygen species (ROS) and reactive sulfur species (RSS). Detecting ROS and RSS species during ferroptosis is particularly crucial for related research. However, the continuous temporal monitoring of these substances during ferroptosis remains an area that has not received sufficient attention. In this study, we constructed fluoresc

BACKGROUND AND AIMS: Maple syrup urine disease (MSUD) is an inherited disorder of branched-chain amino acid metabolism caused by deficiency of the branched-chain α-keto acid dehydrogenase complex. Neurological manifestations predominantly involve the central nervous system, whereas peripheral nervous system involvement has been rarely reported.
RESULTS: We report a genetically confirmed case of intermittent MSUD caused by compound heterozygous pathogenic variants in DBT in a female patient who developed recurrent ataxic episodes beginning in early childhood, followed by acute ascending weakne

Psoriasis is an inflammatory skin disorder characterized by epithelial hyperplasia and infiltration of inflammatory cells. Several immunological therapies have been developed for its treatment, including IL-17 inhibitors such as secukinumab, ixekizumab and bimekizumab. The safety profile of IL-17 inhibitors includes infectious diseases, gastrointestinal disorders, headaches, and cutaneous conditions such as eczema. Here, we report the first case of vitiligo, a depigmenting skin disorder characterized by the selective loss of melanocytes, occurring during treatment with bimekizumab. This case i

BACKGROUND: Prior studies have linked physical activity to cardiovascular-kidney-metabolic (CKM) prevalence or mortality and sedentary behavior to CKM severity, and both behaviors are established mortality risk factors in the general population; however, prospective evidence on their joint associations with cause-specific mortality across CKM stages 0-3 remains limited.
METHODS: We analyzed 8,556 NHANES 2007-2018 participants aged ≥20 years with CKM stages 0-3, complete prespecified covariates, and a positive fasting-subsample weight. Leisure-time physical activity (LTPA) was categorized as i

Maternal caregiving requires sensory vigilance to ensure offspring survival, particularly during sleep. Infant cries are salient sensory cues that reliably trigger maternal arousal and caregiving responses in both humans and dams, but the underlying neural circuitry remains poorly defined. Here we identify a corticohypothalamic circuit in mice that mediates maternal awakening in response to pup calls. We show that layer 6 (L6) neurons in the temporal association cortex and ectorhinal cortex (TeA/Ect) projecting to the preoptic area (POA) exhibit enhanced responsiveness to pup calls after the t

Here we present DynaVelo, a generative neural ordinary differential equation model that learns the joint dynamics of gene expression and transcription factor (TF) motif activity from single-cell multiome data. DynaVelo learns a functional form of the vector field, such that the model can be perturbed in silico and not only used to draw trajectories. We show that DynaVelo recovers the bifurcating in vivo dynamics of wild-type murine germinal center (GC) B cells and reveals how these dynamics change under heterozygous loss of epigenetic regulators Arid1a and Ctcf. In silico perturbation yields d

BACKGROUND: Cerebral small vessel disease (cSVD) encompasses white matter hyperintensities (WMH), enlarged perivascular spaces (PVS), intracerebral hemorrhage (ICH), small vessel stroke (SVS), and cognitive impairment-phenotypes that frequently co-occur yet have been studied as independent genetic endpoints. Whether a shared genetic architecture underlies these co-occurring cSVD-related phenotypes remains unclear.
METHODS: We applied Genomic SEM to GWAS summary statistics from six phenotypes (WMH, PVS, ICH, SVS, cognitive performance, systolic blood pressure), followed by multivariate associa

BACKGROUND: Large-for-gestational-age (LGA) infants are a common complication of gestational diabetes mellitus (GDM), increasing the risk of obstetric trauma and long-term metabolic disorders. Pre-pregnancy body mass index (ppBMI) and gestational weight gain (GWG) are modifiable maternal factors, but their joint impact on LGA in sub-high-altitude, multiethnic populations remain unclear.
OBJECTIVE: To examine the independent and combined associations of ppBMI and GWG with LGA risk among women with GDM in a sub-high-altitude, multiethnic region of southwestern China.
METHODS: In this retrospec
