Lichen planus pigmentosus inversus (LIPPI) is a rare variant of lichen planus, predominantly affecting intertriginous skin areas. Existing knowledge is mostly based on case reports, with few systematic reviews available. A systematic literature search identified 105 cases from case reports and series. Extracted data included demographics, clinical features, comorbidities, treatments, and outcomes. Statistical analysis included descriptive methods and uni-/multivariate logistic regression. The cohort was 64.8% female, with a median age of 58 years. Most lesions affected multiple inverse areas (

Oral lichen planus (OLP) is a chronic inflammatory mucocutaneous autoimmune disease characterized by diverse clinical manifestations in the oral cavity, with a pathogenesis that remains incompletely understood. Aim of this study was to evaluate antinuclear antibodies (ANA) as well as vitamin status in blood samples collected from OLP patients. Fifty OLP patients and 25 healthy controls were included in the study. This study explores the correlation between OLP, elevated ANA titers and elevated Vitamin B6 levels, hypothesizing that a shared autoimmune mechanism or metabolic dysregulation may li

BACKGROUND: Type 1-mediated chronic inflammatory skin diseases affect skin, hair, nails, and mucosa and dramatically impact patients' quality of life. The 2 most prominent examples are lichen planus (LP) and cutaneous lupus erythematosus (CLE). Various cell death pathways are activated in both diseases, including apoptosis and necroptosis. RIPK1 is a key regulator of programmed cell death and thus represents a potential new target for treatment of these diseases.
OBJECTIVE: We sought to determine the impact of RIPK1 on cell death and inflammation in LP and CLE.
METHODS: RNA sequencing of inf

OBJECTIVES: This study provides a descriptive, multi-institutional comparison of dental students' recognition accuracy and management decisions of oral soft tissue pathological entities across four US dental schools. While prior single-institution studies have evaluated diagnostic ability, this work provides a multi-institutional comparison to identify recognition and decision-making variability as well as potential educational implications.
MATERIALS AND METHODS: A voluntary, anonymous online survey with twenty image-based multiple-choice questions was administered to 160 students. Each ques

INTRODUCTION: Oral lichen planus (OLP) is a chronic inflammatory condition with malignant potential for oral squamous cell carcinoma (OSCC). Differential risks of pharmacological treatments, particularly long-term use, remain unclear. We aimed to quantify OSCC risk across treatment modalities and assess potential benefit of combining immunosuppressive and anti-inflammatory agents to inform safer strategies.
METHODS: We conducted a large, retrospective cohort study with propensity score matching to balance demographic and clinical covariates. Patients with OLP treated with either systemic or t

Inflammatory and immune-mediated skin diseases are increasingly recognized as disorders in which genetic susceptibility is shaped and sustained by environmentally responsive regulatory programs. Psoriasis, atopic dermatitis (AD), vitiligo, systemic sclerosis (SSc), lupus erythematosus (LE), and lichen planus (LP) are clinically distinct, yet they share chronic or relapsing inflammation, tissue remodeling, and limited durability of many current therapies. Because genetic variation alone cannot fully explain disease onset, flare dynamics, heterogeneity in severity, or lesion recurrence, epigenet

Background: Oxidative stress (OS) plays an important role in oral lichen planus (OLP) development; however, the precise functions of the genes associated with OS (OSRGs) remain unclear. This study aimed to identify and characterize OS-linked molecular markers in OLP. Methods: Data were obtained from the GSE38616 and GSE211630 datasets, along with 467 OSRGs. Candidate genes were identified by cross-referencing differentially expressed genes with OSRGs. Biomarkers were then selected through a protein-protein interaction network analysis using Cytoscape. Functional enrichment analysis, regulatory

Background: Second primary tumors (SPTs) are a major survivorship challenge in oral squamous cell carcinoma (OSCC), yet their biological phenotypes may differ according to exposure status and immune background. Methods: In this retrospective cohort (2011-2020), 242 surgically treated primary OSCC patients were classified as non-smoking, non-drinking (NSND; never smokers/never drinkers) or smoking and/or drinking (SD; any history of smoking and/or alcohol consumption). SPTs were categorized as extra-oral SPTs (eoSPTs) or multifocal oral SCC (mOSCC), with mOSCC (≥3) denoting ≥3 oral primaries. I

Patient-derived organoids (PDOs) have emerged as advanced bioengineered platforms for modeling oral mucosal tissues, offering unprecedented opportunities to interrogate disease mechanisms and evaluate therapeutic responses in a clinically relevant context. This article provides a critical and integrative assessment of recent advances in organoid technologies applied to oral mucosal disorders, with particular emphasis on oral squamous cell carcinoma (OSCC), oral lichen planus, and oral mucositis. Despite significant progress in therapeutic development, the clinical management of OSCC remains ch

Poikilodermatous skin changes represent a diagnostic challenge due to their non-specific clinical features and broad differential diagnoses. This report presents the case of a 29-year-old female with symmetrical, variably pigmented macules and patches over sun-exposed areas, including the neck and upper limbs. Histopathological examination revealed epidermal atrophy, pigment incontinence, basal layer pigmentation, dermal telangiectasia and a mild lymphocytic infiltrate - findings consistent with poikilodermatous change. No features suggestive of interface dermatitis, atypical lymphoid cells or

BACKGROUND: Oral lichen planus (OLP) is a chronic T-cell-mediated inflammatory disorder of unknown etiology. Accumulating evidence has demonstrated elevated cholesterol levels in OLP, and its oxidation products --oxysterols have been implicated in T cell dysfunction. However, whether the oxysterol is involved in OLP pathogenesis remains to be fully elucidated.
METHODS: Metabolomics was performed to profile oxysterols in the plasma of OLP patients, followed by functional enrichment analysis. Single-cell RNA sequencing was utilized to characterize gene expression dysregulation in tissue-residen

Mitochondrial metabolism and innate immune signaling are increasingly recognized as intersecting pathways in chronic inflammatory disease. Here, we identify a metabolically driven mechanism linking the TCA cycle imbalance to mucosal inflammation in oral lichen planus (OLP). Multi-omics analysis revealed that fumarate hydratase (FH) is upregulated in OLP tissues and cells, leading to significant fumarate depletion. This metabolic shift induces mitochondrial dysfunction, characterized by enhanced oxidative phosphorylation, proton leak, and TFAM downregulation. These changes destabilize the mitoc

Observational studies have consistently reported associations between oral lichen planus (OLP) and psychological distress, yet whether these relationships are causal remains uncertain. We performed a two-sample Mendelian randomization (MR) analysis to investigate the potential causal effects of depressive traits and anxiety-stress-related disorders (ASRD) on OLP. Genetic instruments for depressive traits (n = 500,199) and ASRD (n = 31,885) were obtained from the largest available genome-wide association study (GWAS) meta-analyses of European ancestry. Summary-level data for OLP were derived fr
