A comparison of two- and three-dose MVA-BN mpox vaccination series on anti-monkeypox virus immunity.

The 2022 global outbreak of clade IIb mpox represented a turning point in public health's handling of poxviruses. The primary vaccine available for the prevention of mpox is modified vaccinia Ankara from Bavarian-Nordic (MVA-BN). We previously reported a nondurable and low-avidity antibody response against mpox elicited by MVA-BN. In this study, we expanded upon this knowledge by employing a microneutralization assay to measure monkeypox virus (MPXV) neutralizing titers and a multiplexed immunoassay to assess IgG titers and avidity against eight MPXV antigens and two vaccinia antigens. Through

An improved and high-throughput mpox virus microneutralization assay.

Since the 2022 global mpox outbreak, the lack of specific antibodies for mpox virus (MPXV) detection has hindered precise immunoassays due to cross-reactivity among Orthopoxviruses (OPXVs). This study developed and characterized two monoclonal antibodies (mAbs), CML01 and CML02, targeting the MPXV A35 protein, a conserved surface antigen of extracellular virions. Cross-reactivity assessments via enzyme-linked immunosorbent assay (ELISA), western blot, and indirect immunofluorescence assays (IFA) confirmed that both mAbs bound exclusively to MPXV A35, showing no reactivity with homologous prote

[Mpox virus (MPXV) DNA replication machinery: Structural mechanisms and antiviral targets]: A review.

Current control of mpox virus (MPXV) still relies mainly on smallpox-derived vaccines and antiviral agents, but their clinical use remains limited by uncertain efficacy, potential toxicity, and increasing resistance. Meanwhile, the key mechanisms underlying MPXV replication and its potential druggability remain insufficiently understood. Because viral DNA replication is directly linked to viral proliferation, the core proteins of the replication machinery are not only essential for maintaining the viral life cycle but may also represent more selective therapeutic targets. Therefore, systematic

[John Glasgow Kerr and the Compilation of Zhong Dou Jie Fa and Niu Dou Qian Shuo].

Zhong Dou Jie Fa and Niu Dou Qian Shuo were written by John Glasgow Kerr, a missionary from America, trained local smallpox practitioners in Guangzhou in the second half of the 19th century. The two books Zhong Dou Jie Fa and Niu Dou Qian Shuo introduced simple and easy ways for practical inoculation. The books suggested that it was not necessary to consider acupoints and seasons for vaccination. They introduced the method of storing vaccine in capillary tubes and made use of microscopes and vaseline, popularly used in Europe at the time, promoting the development of vaccination technology obj

Phylogenetic and genome-wide analysis of codon usage reveals geographic and host-specific adaptation in Vaccinia virus.

Vaccinia virus (VACV) is a member of the Poxviridae family and served as the vaccine strain used to eradicate smallpox. As a large double-stranded DNA virus, VACV exhibits a broad host range and remains a model organism for studying viral evolution and gene expression. Codon usage bias (CUB) is a fundamental feature of genome evolution that reflects the interplay between mutational pressure and natural selection. In this study, we systematically analyzed codon usage patterns in 107 strains of Vaccinia virus (VACV) to uncover the driving forces behind its genomic evolution and host adaptation.

Proteomic Applications in Vaccine Development.

Controversy surrounding vaccines is not new; the first vaccine proven to be effective in preventing disease was subjected to wild rumors ( https://www.who.int/news-room/spotlight/history-of-vaccination/history-of-smallpox-vaccination ). In 1796 Dr. Edward Jenner inoculated James Phipps with pus from a cowpox sore taken from the hand of Sarah Nelmes (see Fig. 10.1). While 8-year-old James fell ill with a mild case of cowpox, he eventually recovered. Poor James was not yet free from his life as a "lab rat," as Dr. Jenner later inoculated him with pus taken from an individual suffering with small

Digitization of Historical Data from Somalia's Last Smallpox Outbreaks 1976-1977.

Over more than three thousand years, smallpox caused millions of deaths worldwide. With smallpox declared eradicated in 1980, potential use of the variola virus as a bioterrorist threat remains a concern. While modern disease outbreaks are modelled using current data, understanding how to control an eradicated disease such as smallpox relies on the analysis of historical data. Handwritten smallpox outbreak data from Somalia (1976-1977) were obtained from the World Health Organization by a Public Health England study team (now UK Health Security Agency). The data were manually transcribed into

Mpox knowledge, attitudes toward Mpox vaccine and hesitancy for Mpox vaccination among teachers in Kenya.

BACKGROUND: Sixteen African countries, including Kenya, reported Mpox cases over a two-month period, prompting the Africa Centers for Disease Control and Prevention to declare the outbreak a Public Health Emergency of Continental Security on 13th August 2024. The World Health Organization issued guidance for Mpox prevention in schools, where teachers are expected to support infection prevention. This study assessed teachers' knowledge of Mpox and attitudes toward Mpox vaccination, given these factors that may influence teachers' confidence in promoting preventive behaviours and vaccination awa

Perturbation of RNA homeostasis impairs mitochondrial respiration during poxvirus infection through excess RNA accumulation.

Induction of RNA degradation in infected cells is a strategy used by many viruses to promote efficient replication. Vaccinia virus, the prototype poxvirus and the vaccine platform for smallpox and mpox, encodes two decapping enzymes to accelerate mRNA and double-stranded RNA (dsRNA) degradation during infection, through functional coordination with host cell RNA exonuclease. Previous studies have largely focused on RNA degradation as a mechanism for regulating viral gene expression and evading innate immune sensing. Here, we show that impaired RNA degradation in vaccinia virus-infected cells,

Circular RNA vaccines encoding fusion proteins of Mpox Virus A35R-M1R and B6R-A29L induce robust and durable protective immunity in mice.

Developing a safe and effective vaccine is crucial to control the recent worldwide outbreaks of mpox. Here, building upon our previously established lipid nanoparticle (LNP)-encapsulated circRNA vaccine platform, we constructed two bivalent mpox virus (MPXV) circRNA vaccines: cirBA encoding a B6R-A29L tandem antigen and cirAM encoding an A35R-M1R fusion antigen. Both bivalent MPXV circRNA vaccines, whether administered alone or in combination (designated cirMix), could induce robust and durable MPXV antigen-specific humoral and cellular immune responses in mice, conferring complete protection

Partners, Pride, and Prevention: Scaling Mpox Vaccination Access Across Minnesota.

Mpox is a rare but potentially serious vaccine-preventable disease. The 2022 United States outbreak disproportionately impacted gay, bisexual, and other men who have sex with men, people living with HIV, and people of transgender experience. Early vaccination efforts revealed substantial racial and geographic inequities, with lower uptake among Black and Hispanic cisgender men, transgender women, and residents of rural areas. To address these challenges, Fairview's Minnesota Immunization Networking Initiative (MINI), a 20-year-old mobile health collaborative, partnered with state and local pub

CircRNA based bi-antigen vaccines against mpox virus induce potent and durable cross-protection in mice.

The ongoing epidemic of mpox highlights the urgency of developing novel and effective vaccines against mpox virus (MPXV). Circular RNA (circRNA) has emerged as a promising novel vaccine platform due to its high stability, resistance to exonuclease-mediated degradation, and potential to support prolonged antigen expression. Based on our lipid nanoparticle (LNP)-encapsulated circRNA vaccine platform, we developed two novel bi-antigen circRNA vaccines cirEV and cirMV, which respectively encode the combination of MPXV extracellular enveloped virion (EEV) antigens (A35R and B6R) or intracellular ma

[Responses and resistance in the fight against smallpox, Chile, eighteenth to twentieth centuries].

This article explores how political and health authorities in Chile responded to smallpox epidemics. It focuses on the methods employed (isolation, variolation, and vaccination) and the institutional strengthening that led to mandatory smallpox vaccination. We argue that these responses reflect the centralization of state power in public health and a process of institutional consolidation. However, the implementation of these policies met with passive resistance, as evidenced by low vaccination rates in the first half of the twentieth century. This reveals tensions between state health objecti

[Monkeypox virus (mpox) reemergence and One Health : from concept to global reality].

The Monkeypox virus (mpox) is a double-stranded DNA orthopoxvirus responsible for a zoonotic disease historically confined to Central and West Africa. First identified in humans in 1970, it was long regarded as an infection characterized by limited human-to-human transmission, primarily driven by repeated zoonotic spillover events from animal reservoirs, particularly rodents. The global outbreak of 2022 fundamentally reshaped this understanding by demonstrating the virus's capacity to sustain prolonged human-to-human transmission across multiple continents. This epidemiological shift has unfol

Mapping Mpox vaccine hesitancy using an integrated machine learning and structural equation modeling approach.

This study employs an integrated computational approach to investigate Mpox vaccine intention in Bangladesh as Mpox immunisation strategies require a thorough understanding of behavioural determinants. Data from 405 participants were analysed using a structured framework that included frequentist statistics to examine demographic relationships, t-distributed Stochastic Neighbour Embedding (t-SNE) for unsupervised grouping, and structural equation modelling (SEM) to confirm underlying behavioural trends. About 77% of people were willing to get vaccinated, and this was mainly influenced by their
