Monkeypox virus (MPXV), a zoonotic viral disease first identified in 1958, has re-emerged as a major global health concern. The first human case was reported in 1970 in the Democratic Republic of Congo (DRC), and since then, MPXV has demonstrated a clinical resemblance to smallpox, with transmission occurring through both animal-to-human and human-to-human contact. In 2022, the largest recorded outbreak resulted in over 30,000 cases across the Western Pacific, Asia, Europe, and the Americas. Subsequently, between 2022 and 2024, more than 100,000 confirmed cases were documented worldwide, leadi

Orthopoxviruses remain a persistent global health concern due to the ongoing circulation of mpox, the possibility of the re-emergence of smallpox, and the threats posed by many poxviruses that infect animals and/or humans. The limited availability of antiviral drugs, the unproven efficacy in humans, and the emergence of resistant mutants underscore the need for new and better therapeutics. In this study, we identify and characterize ZW-2038, a hydroxypyridone-3-carboxamide analog, as an antiviral compound against vaccinia virus (VACV), monkeypox virus (MPXV), and cowpox virus (CPXV). BACKGROUND: The 2022 global outbreak of monkeypox virus (MPXV) clade IIb prompted widespread use of the modified vaccinia Ankara-Bavarian Nordic (MVA-BN) vaccine which conferred partial protection and contributed to outbreak control. In 2024, the emergence and regional expansion of MPXV clade Ib in central Africa raised new public health concerns and prompted questions about the extent to which immunity elicited by previous clade IIb infection or vaccination confers cross-neutralising immunity against clade Ib. We aimed to assess neutralising and binding antibody responses to MPXV clades Ia, I

OBJECTIVES: Mpox emerged as a significant public health threat beyond endemic regions during 2022-2023, with sustained transmission among men who have sex with men (MSM) and transgender populations. Community-based surveillance may improve case detection in high-risk groups.
METHODS: A cross-sectional sentinel surveillance study was conducted from October 2023 to February 2024 across seven community-based organizations in six geographic zones of India. Clinic attendees (n = 1796) were screened for Mpox-like disease (MPLD) using WHO/Centers for Disease Control and Prevention (CDC) case definit

BACKGROUND: Emerging zoonotic poxviruses such as lumpy skin disease virus (LSDV) and monkeypox pose significant threats to animal and human health, yet validated antiviral targets remain scarce. Variola virus, the causative agent of smallpox, also belongs to the Poxviridae family, emphasizing the importance of preparedness against potential re-emergence.
METHODS: We developed an AI-Enabled structural bioinformatics pipeline integrating sequence conservation, phylogenetic analysis, AlphaFold2/ESMFold structure prediction, structural comparison, active site mapping, molecular docking, molecular

The development of the smallpox vaccine two centuries ago laid the foundation for modern vaccinology. Smallpox, once a major killer in human history, was eradicated globally in 1980. This breakthrough encouraged the development of contemporary vaccine technologies, including inactivated, recombinant, viral vector, and mRNA platforms. Owing to developments in genetics, bioinformatics, and molecular biology, personalised vaccines have been made possible. Advances in the field of immunology facilitated an accelerated response to the COVID-19 pandemic. This perspective synthesises key developments

BACKGROUND: Mpox, caused by the monkeypox virus, emerged as a global public health concern following the 2022 outbreaks in non-endemic regions. Although classical presentations include a febrile prodrome and mucocutaneous lesions, atypical symptoms and stigma have complicated clinical recognition and containment. Smallpox-derived vaccines, such as ACAM2000 and modified vaccinia Ankara, have been deployed for prophylaxis; however, access, policy implementation, and public acceptance remain inconsistent across regions.
SUMMARY: Global mpox vaccination strategies demonstrate pronounced dispariti

Monkeypox (Mpox) remains a significant public health concern due to the emergence of rapidly evolving strains and the suboptimal efficacy of existing smallpox vaccines. Recombinant subunit vaccines offer a promising and safe alternative. The A29L protein of the Mpox virus (MPXV) is essential for viral attachment and entry. The N-terminal residues 1-34 of A29L, which harbor the heparin-binding domain, are critical targets for neutralizing antibodies. Based on this, we hypothesized that this truncated 34-residue peptide of A29L could serve as a potent subunit vaccine candidate. To enhance immuno

INTRODUCTION: This article describes the protocol of the PregInPoxVac vaccine trial, evaluating the safety and immunogenicity of two MVA-BN vaccination regimens (full-dose and half- dose) administered subcutaneously with a 28-day interval in infants aged 4 to <24 months old. Non-inferiority of the full dose and half dose regimens will be compared to adults receiving a full dose regimen as part of the POX-MVA-045 study (ClinicalTrials.govIdentifier:NCT06549530). The PregInPoxVac study is being conducted in Boende, Tshuapa Province, Democratic Republic of the Congo (DRC), a mpox-endemic area; th

The eradication of smallpox remains one of the most significant achievements in public health, symbolizing the power of international cooperation, scientific coordination, and collective responsibility. However, the continued retention of eradicated pathogens, particularly live variola virus, reflects a broader and unresolved challenge within global biosecurity governance. While debates surrounding smallpox have often been framed narrowly in terms of risk-benefit considerations specific to a single pathogen, they increasingly expose deeper structural limitations in the systems designed to prev

In settler colonial contexts, health interventions are often sites for contesting authority, belonging, and protection. On Lingít Aaní, colonial responses to infectious disease illustrate how vaccination and public health practices produced racial hierarchies and secured white futurity by exposing Tlingit people to biomedical extraction, risk, and surveillance. I conceptualize this dynamic as inoculating whiteness, a situated analytic for understanding how immunological interventions shielded settler bodies from biological and political threat while exposing Indigenous bodies to experimentatio

OBJECTIVE: The increasing global threat posed by viral infections that are transmitted to humans from animals has intensified interest in emerging pathogens such as monkeypox virus (MPV). This study investigated the potential of drug repurposing as an efficient therapeutic strategy against MPV, focusing on its close genetic relationships with vaccinia and variola viruses.
METHODS: Due to the structural and evolutionary similarities among these orthopoxviruses, the Food and Drug Administration (FDA)-approved smallpox drug tecovirimat was selected as a reference drug. A descriptive-analytical a

Vaccination is one of the most effective public health interventions in history, yet resistance to vaccines has persisted across centuries. This narrative historical review examines the evolution of vaccine hesitancy and anti-vaccine sentiment from the smallpox era to the COVID-19 pandemic. Sources were identified through PubMed and major public health organizations, including the World Health Organization and the U.S. Centers for Disease Control and Prevention, spanning the early 18th century through 2025. Across distinct historical periods, including early smallpox vaccination, mid-20th-cent

Monkeypox (Mpox) is a zoonotic disease caused by MPXV and remains a threat to global public health. MPXV Clade Ib has higher mortality, whereas Clade IIb exhibits widespread international transmission. WHO has twice designated mpox epidemics as a Public Health Emergency of International Concern (PHEIC). This highlights that the virus is not only pathogenic but also has the potential to cause a global pandemic, posing a challenge to the international public health system. The rapid transmission of MPXV between and within hosts is related to two key virus particles: IMV and EEV. Antigenic protei

Human Monkeypox is a smallpox-like zoonotic infection transmitted through animals, contaminated objects, or human contact, posing growing public health risks. Despite its declaration as a global health emergency in 2022, community awareness remains limited in Ethiopia's Oromia region, where ecological and socioeconomic factors heighten vulnerability. Thus, this community-based cross-sectional study, conducted from July 6 to August 30, 2025, among 842 adults in Oromia, assessed knowledge, attitudes, and associated factors regarding Human Monkeypox. Data were analyzed using descriptive statistic

The impact of smallpox (variola) on Aboriginal communities in Australia beginning in 1789 was catastrophic and continues to cause intergenerational trauma. Historically biased perspectives and contemporary misinformation of the disease's introduction and spread impede modern-day truth-telling and efforts towards reconciliation and national healing. Understanding whether the disease entered and spread from pre-colonial Makassan (Indonesian) trade along the northern coast, or from the British First Fleet's arrival in southeastern Australia in 1788, is necessary to estimate the demographic impact

To expand the supply of the modified vaccinia Ankara (MVA) smallpox/mpox vaccine, fractional doses administered intradermally (ID) have been considered as alternatives to the standard dose given subcutaneously (SC). In a recent clinical trial, participants received the standard 108 infectious units of vaccine SC or a one-fifth (IDH) or a one-tenth (IDL) dose ID twice 28-days apart. Two weeks after the second dose, sera were analyzed using a vaccinia virus (VACV) strain Western Reserve plaque-reduction test (PRNT) with the conclusion that IDH but not IDL was non-inferior to SC. The present stud
