The Maintenance of Early Responses and Achievement of Delayed Responses to Deucravacitinib Treatment in Psoriais: A 104-Week Real-World Study in Japan.

The tyrosine kinase 2 inhibitor deucravacitinib is effective for psoriasis. However, it is unclear whether early responses to deucravacitinib in real-world practice are maintained for 2 years and whether patients without early responses can achieve delayed responses. This study is aimed to evaluate the sustainability of week 16 responses to deucravacitinib treatment for psoriasis and to evaluate delayed responses in week 16 poor responders. This prospective study included 117 Japanese patients with moderate-to-severe psoriasis treated with deucravacitinib. Patients who achieved psoriasis area

Relative Efficacy of Immunomodulatory Monotherapies for Psoriasis of the Scalp: A Network Meta-Analysis Study.

BACKGROUND: Recently, the literature has expanded with peer-reviewed studies on immunomodulatory agents' efficacy on scalp psoriasis-which, in turn, widened knowledge gaps regarding these agents' relative effectiveness. We determined the relative efficacy of immunomodulatory monotherapies for scalp psoriasis. METHODS: We ran Bayesian network meta-analyses (NMAs) using outcomes related to Psoriasis Scalp Severity Index (PSSI) and scalp-specific Physician's Global Assessment of clear (0) or almost clear (1) (Sc-PGA 0/1). RESULTS: We estimated the relative efficacy of 22 interventions (includin

Development and in vitro/in vivo evaluation of polymeric dissolving microneedle formulation for psoriasis treatment.

Psoriasis is an inflammatory autoimmune skin disease that significantly impacts quality of life. The chronic and recurrent nature of the disease can last a lifetime. Current conventional treatment options have several drawbacks, including inadequate or short-lived efficacy, issues with tolerability and adherence, practical limitations associated with topical applications and phototherapy, and significant safety and cost concerns related to systemic treatments. Generally, symptomatic treatments are applied, but complete recovery is rarely achieved. Researchers are exploring alternative treatmen

Role of real-world evidence from patient registries for psoriasis in decision-making: a systematic review.

INTRODUCTION: Treatment of moderate to severe psoriasis typically requires the use of multiple systemic therapies over a patient's lifetime. The efficacy and safety of systemic treatments are typically evaluated in clinical trials; however, patient registries are increasingly used to monitor long-term outcomes of systemic therapies for psoriasis in real-world settings. Psoriasis registries also generate important real-world evidence about psoriasis treatment that may facilitate a greater understanding of outcomes outside of a controlled clinical trial setting. This study thus characterises the

Hydrogen ameliorates psoriasis-like skin inflammation via inhibiting the cGAS-STING pathway.

Psoriasis is a chronic disease caused by abnormal immune system response, which is characterized by excessive keratinocyte proliferation and the activation of cytokine signaling pathways. In a previous study, we demonstrated in a psoriasis mouse model that hydrogen-rich water, an effective reactive oxygen species (ROS) scavenger, significantly improves disease severity. However, the precise molecular mechanism by which hydrogen helps in psoriasis treatment remains inadequately understood. This study assessed the role of hydrogen in suppressing keratinocyte hyperproliferation. We observed that

Reprogramming immunity at the metabolic-epidermal interface in obesity-associated psoriasis.

Obesity and psoriasis are chronic inflammatory disorders, now recognized to be interconnected, in which metabolic overload drives immune dysregulation and therapeutic resistance. Excess adiposity converts adipose tissue into an inflammatory organ that releases adipokines and cytokine-like mediators, reprogramming keratinocytes and immune cells to sustain cytokine-driven inflammatory circuits in the skin. Excess nutrients and lipotoxic stress impair mitochondrial function, enhance glycolysis, and induce epigenetic remodeling in myeloid and epithelial lineages, generating metabolic memory that p

Contribution of Phenotypic Age Acceleration and Body Mass Index to Psoriasis Risk in US Adults.

Psoriasis, a prevalent chronic inflammatory skin disease, substantially impairs patients' quality of life and is frequently linked to numerous systemic comorbidities. We aimed to investigate whether phenotypic age acceleration (PhenoAge-accel) and obesity synergistically affect the risk of psoriasis. This analysis utilised data from 11 344 participants in the 2003-2006 and 2009-2010 cycles of the National Health and Nutrition Examination Survey (NHANES). Weighted multivariable logistic regression, along with subgroup and sensitivity analyses, was conducted to assess the combined influence of P

Natural killer cells are required for imiquimod-induced psoriasis-like skin inflammation in mice.

BACKGROUND/PURPOSE(S): Psoriasis is a skin-specific autoimmune disease that causes scaling, erythema and thickening. The disease involves immune cell infiltration and keratinocyte proliferation in skin lesions. Previous studies have demonstrated that Th17 cells and IL-22 play prominent roles in the pathogenesis of psoriasis via IL-17, IL-22 and IL-23 signaling. However, the role of innate cells in this process remains unclear. METHODS: In this study, using the TLR7/8 agonist imiquimod (IMQ) in a well-established murine model, we demonstrated that natural killer (NK) cells play an important ro

Exploration of shared gene signatures and molecular mechanisms between psoriasis and COVID-19: evidence from transcriptome data.

Emerging evidence suggests that the onset or worsening of psoriasis can occur following COVID-19 infection or vaccination. Additionally, individuals with psoriasis may be more susceptible to COVID-19. However, the underlying mechanisms driving this phenomenon remain unclear. This study aims to explore the potential shared mechanisms and the complex interplay between psoriasis and COVID-19. Gene expression profiles for COVID-19 (GSE162183) and psoriasis (GSE54456) were obtained from the Gene Expression Omnibus (GEO) database. Common differentially expressed genes (DEGs) were identified, followe

Impact of Brodalumab on Serum Levels of IL-6, IL-17A, IFN-α, IFN-γ, and TNF-α in Patients with Psoriasis Who Failed Treatment with TNF-α Inhibitors.

Psoriasis is a chronic, immune-mediated inflammatory skin disorder that significantly impacts patients' quality of life. While TNF-α inhibitors are frequently used to treat moderate-to-severe cases, not all patients respond adequately. Brodalumab, a monoclonal antibody targeting the IL-17 receptor A, has emerged as an alternative for individuals unresponsive to prior therapies. This prospective study investigated the effects of brodalumab on serum cytokine levels-specifically IL-6, IL-17A, IFN-α, IFN-γ, and TNF-α-and their correlation with disease severity as assessed by Psoriasis Area and Sev

A review of current therapeutics for psoriasis: Systemic and topical use of non-biological drugs.

PURPOSE: Psoriasis is a chronic, immune-mediated inflammatory disease characterized by dysregulated cytokine signaling, keratinocyte hyperproliferation, and recurrent relapses requiring sustained management. Although biologic agents have transformed treatment by selectively targeting IL-17, IL-23, and TNF-α pathways, important limitations persist, including high costs, limited accessibility, treatment failures, and insufficient long-term safety data. Therefore, topical therapies and non-biologic small-molecule systemic agents remain essential, particularly for mild-to-moderate disease and for
