Scoping review: biomarkers in chronic inducible urticaria.

OBJECTIVE: Chronic inducible urticaria (CIndU) affects approximately 0.5% of the population and is characterized by wheals triggered by specific stimuli, such as cold or heat exposure, physical exertion, mechanical friction, vibration, or delayed pressure, distinguishing it from chronic spontaneous urticaria. Biomarkers may improve diagnosis, define endotypes, predict disease course, monitor treatment responses, and subsequently guide personalized treatment strategies. This scoping review aimed to map current evidence on diagnostic, prognostic, predictive, and monitoring or treatment-response

Toxicological evaluation in vitro of novel photosensitizers intended for photodynamic therapy.

Three selected photosensitizers (PS) with potential applications in PDT (ZnPcS2, TMPyP, PPIX) were evaluated for skin irritation, phototoxicity and sensitization using exclusively non-animal test methods. The experimental approach was based on OECD TG with protocol modifications to accommodate the specific requirements of photosensitizers testing. Reconstructed human epidermis (RhE) based assays were performed using adapted protocols derived from OECD TG 439 and OECD TG 498, while skin sensitization was assessed using methods based on OECD TG 442C and OECD TG 442D. TMPyP and PPIX showed no evi

Seasonal variations in erythropoietic protoporphyria patients' responses about sun behaviour, symptoms, and treatment effect.

Patients with erythropoietic protoporphyria (EPP) have highly variable, increased levels of erythrocyte protoporphyrin IX (PpIX). Upon exposure to visible light, the PpIX is released from the erythrocytes to the skin, producing phototoxic reactions. Severe photosensitivity heavily impacts the patients' daily behavior regarding sun exposure and outdoor life. As patients with EPP in Northern European countries may be free of symptoms during winter, their individual evaluation of sun behavior in summer, the severity of summer symptoms, and effect of oral treatment with Cimetidine and/or zinc sulp

Novel cutaneous adverse effects of the increasingly prevalent glucagon-like peptide 1 agonists.

Prescribing of the glucagon-like peptide 1 receptor agonists (GLP-1RA) is rapidly increasing as their efficacy for multiple indications becomes apparent. A better understanding of their cutaneous adverse effect profile is crucial, particularly as they are suggested for use in inflammatory dermatoses. We describe two patients who developed novel cutaneous reactions associated with initiation of a GLP-1RA. Patient 1 developed multiple tender subcutaneous nodules on the trunk and limbs 4 weeks after the initiation of semaglutide. Histology demonstrated a noncaseating granulomatous dermatitis, and

Enhanced Extracellular Matrix Synthesis-Related Responses Induced by Ubiquinol-10 Derivatives in Human Dermal Fibroblasts.

Skin aging is characterized by impaired extracellular matrix (ECM) synthesis and increased inflammatory signaling in dermal fibroblasts. Notably, ubiquinol-10 (UqH-10), the reduced form of coenzyme Q10 (ubiquinone-10, Uq-10), supports mitochondrial function and reduces oxidative stress, but its dermal levels decrease with aging. The topical efficacy of UqH-10 is limited by its physicochemical properties, including oxidative instability, photo instability, and phototoxicity. This study aimed to evaluate the anti-aging potential of esterified UqH-10 derivatives-UqH 1,4-bis-hemi-succinate (UqH-SU

Reprogramming Photodynamic Therapy with BODIPY Photosensitizers: From Local Phototoxicity Toward Precision Photodynamic Oncology.

Photodynamic therapy (PDT) offers spatially confined, repeatable and mechanistically orthogonal tumor control, yet its broader clinical impact remains limited by inadequate light access, oxygen dependence and incomplete biological selectivity. Boron-dipyrromethene (BODIPY) dyes provide an unusually programmable platform for addressing these constraints. Although native BODIPYs are fluorescence-biased and inefficient triplet photosensitizers, their absorption, intersystem crossing, reactive oxygen species identity, aggregation, targeting and activation can be independently engineered through mo

Overcoming phototoxicity of sonosensitizers: Curcumin-coordinated iridium(III) complexes for sonodynamic therapy.

Sonodynamic therapy (SDT) holds promise for deep-seated tumors, but most sonosensitizers suffer from severe phototoxicity, requiring patients to avoid light exposure after treatment. Here, we introduce a strategy using natural product curcumin as an auxiliary ligand to construct iridium(III) complexes (Ir1-Ir3). Ir3, featuring dual natural-product-derived ligands (curcumin and coumarin 6), exhibits a well-separated HOMO-LUMO distribution and a small S1-T1 energy gap, enabling highly efficient ultrasound (US)-triggered singlet oxygen (1O2) generation while showing negligible phototoxicity under

Physiological and performance effects of a simulated prolonged service game in competitive male tennis players.

BACKGROUND: It is well established that ultraviolet radiation is related to non-melanoma skin cancer (NMSC) in Caucasians. Considering that patients with vitiligo have often no protective pigment in sun-exposed depigmented/white skin together with severe oxidative stress due to accumulation of millimolar epidermal hydrogen peroxide (H(2)O(2)), it would be expected that these patients develop a higher risk for early photodamage and NMSC. However, scattered reports on low patient numbers documented no increased risk for sun-induced skin cancers in this disease. OBJECTIVE: The aim of this study

940-nm Photobiomodulation Enhances ADSC-Mediated Tracheal Cartilage Repair through TRPV1-Associated Calcium Signaling and Mitochondrial Redox Remodeling.

BACKGROUND: : Azathioprine and mercaptopurine are commonly used in chronic active Crohn's disease. They share the disadvantage of a delayed onset of action and potentially serious side-effects, and are metabolized to thioguanine nucleotides which are thought to be the active metabolites. The direct use of 6-thioguanine may offer a more rapid and safer alternative. We conducted an open prospective study to investigate the efficacy and safety of 6-thioguanine in chronic active Crohn's disease. METHODS: : Thirty-seven patients with chronic active Crohn's disease and a Crohn's disease activity in

PEGylated isothiocyanate-functionalized zinc(II) phthalocyanine exhibits cell-type dependent photodynamic activity in 2D and 3D tumor models.

This study reports the synthesis and characterization of an asymmetric zinc(II) phthalocyanine (5) containing three tetraethyleneglycol monomethyl ether groups and one isothiocyanatophenoxy group at its periphery. The isothiocyanate unit was selected to ensure selective bioconjugation under mild reaction conditions and to reduce side product formation, while tetraethyleneglycol monomethyl ether groups were incorporated to increase solubility and tailor photophysical and photochemical properties relevant to photodynamic therapy applications. The compound showed a singlet oxygen quantum yield (Φ

A GSH-responsive Ru(II) complex: Mitochondria-targeted cell imaging and photodynamic therapy.

Glutathione (GSH) is a critical intracellular antioxidant. In cancer cells, the overexpression of GSH scavenges reactive oxygen species (ROS) produced during photodynamic therapy (PDT), thereby compromising therapeutic efficacy. To address this limitation, we designed and synthesized a GSH-responsive Ru(II) complex (Ru(phen)2(bpy-nap)(PF6)2) for simultaneous GSH imaging and enhanced PDT. This probe exhibits excellent GSH-sensing performance, including a significant phosphorescence turn-on response and a low detection limit (0.05 μM). It was successfully employed for visualizing both endogenous

Nano Enabled Dual-Responsive Drug Carrier Originated from Acetalated Dextran/Carboxylated Nanocellulose-Based Core-Shell Microspheres.

Magneto-photoresponsive polymeric microspheres represent a promising platform for targeted, externally triggered drug delivery. However, achieving precise control while minimizing phototoxicity remains a major challenge. In this study, we developed biodegradable core-shell microspheres composed of acetalated dextran (AcD) and cellulose modified by citric acid (CMC), co-loaded with a photoacid generator (PAG), Fe3O4 nanoparticles, and zerovalent iron (ZVI). These components enable dual-stimuli responsiveness, in which short UV exposure (365 nm, 10 min on/off cycles) and an alternating magnetic

Milk-derived exosomes (MDE) for photosensitizer delivery in photodynamic therapy: A review.

Photodynamic therapy (PDT) is a novel light-based modality that relies on photoactivatable photosensitizers (PSs) to generate cytotoxic reactive oxygen species (ROS), but its clinical prospects are impeded by issues related to poor tumour specificity, penetration and off-target phototoxicity. Milk-derived exosomes (MDE), naturally occurring extracellular vesicles (30-150 nm in diameter) with excellent biocompatibility and low immunogenicity, have recently emerged as promising vehicles in drug delivery. MDE enhance intercellular communication by transporting proteins, lipids, and nucleic acids

Hydroxyl radical from iridium(III)-based photosensitizer triggers PANoptosis and ferroptosis for hypoxia-tolerant photodynamic immunotherapy.

Exploring photosensitizers that efficiently generate oxygen-independent hydroxyl radical (•OH) and activate highly immunogenic PANoptosis and ferroptosis shows promise in addressing the limited efficacy of photodynamic immunotherapy (PDI) in the hypoxic and immunosuppressive tumor microenvironment (TME). However, challenges still exist due to the unsatisfactory •OH production efficiency in most organic photosensitizers and the unresolved relationship between •OH and PANoptosis. Here, we designed and synthesized two novel iridium(III) complexes, Ir1 and Ir2. Notably, Ir1 produced more type I/II

An azoreductase-responsive "OFF-ON-OFF" reversible photosensitizer for highly selective antibacterial photodynamic therapy.

Antibacterial photodynamic therapy (aPDT) represents a highly promising approach for combating resistant pathogens without inducing drug resistance. Selective photo-inactivation of bacteria without damage to the mammalian cells is a prerequisite for the clinical application of aPDT. In this work, based on the different azoreductase levels between mammalian cells and Staphylococcus aureus (S. aureus), an azoreductase-responsive "OFF-ON-OFF" photosensitizer, namely Ru-AzoCF3, was designed and synthesized as a proof-of-concept. The aPDT activity of Ru-AzoCF3 is initially suppressed by the presenc

Light-activated targeted degradation of VHL drug for selective tumor killing: A novel strategy for precision therapy of bladder cancer.

Photodegradation-targeting chimeras (PDTACs) addresses the off-target toxicity issues of proteolysis-targeting chimeras (PROTACs) in terms of technical principle; Difficulty in defining tumor boundaries and controlling light exposure increases the risk of damaging peritumoral tissues if light exposure oversteps the tumor. This study discovered that compared to SV-HUC-1 cells (normal cells), Von Hippel-Lindau (VHL) exhibits more active regulation of CDK2/4 in T24 cells (bladder cancer cells); Based on this, a series of PDTACs linking VHL ligands to photosensitizers were synthesized, and ZnPc-PE

Pulmonary delivery of GSH-responsive 131I-labeled albumin-based nanosystem for lung cancer with enhanced cerenkov-induced photodynamic therapy.

Cerenkov radiation-induced photodynamic therapy (CR-PDT) addresses the tissue penetration limitations of traditional photodynamic therapy for deep-seated tumors. However, continuous phototoxicity in normal tissues and inefficient tumor accumulation of co-delivery of radionuclides and photosensitizer remain the main challenges to the therapy efficacy of lung cancer. Here, we developed a novel pulmonary delivered 131I-labeled GSH-responsive chlorin e6 (Ce6)-conjugated Bovine Serum Albumin (BSA) nanoassembly (131I-CBA ) for efficient and selective tumor therapy in orthotopic lung cancer.
