Photodegradation-targeting chimeras (PDTACs) addresses the off-target toxicity issues of proteolysis-targeting chimeras (PROTACs) in terms of technical principle; Difficulty in defining tumor boundaries and controlling light exposure increases the risk of damaging peritumoral tissues if light exposure oversteps the tumor. This study discovered that compared to SV-HUC-1 cells (normal cells), Von Hippel-Lindau (VHL) exhibits more active regulation of CDK2/4 in T24 cells (bladder cancer cells); Based on this, a series of PDTACs linking VHL ligands to photosensitizers were synthesized, and ZnPc-PE

Cerenkov radiation-induced photodynamic therapy (CR-PDT) addresses the tissue penetration limitations of traditional photodynamic therapy for deep-seated tumors. However, continuous phototoxicity in normal tissues and inefficient tumor accumulation of co-delivery of radionuclides and photosensitizer remain the main challenges to the therapy efficacy of lung cancer. Here, we developed a novel pulmonary delivered 131I-labeled GSH-responsive chlorin e6 (Ce6)-conjugated Bovine Serum Albumin (BSA) nanoassembly (131I-CBA ) for efficient and selective tumor therapy in orthotopic lung cancer. The 131I

Cancer treatment faces significant challenges, including high invasiveness, systemic toxicity, and recurrence risks. Minimally invasive therapies such as photodynamic therapy (PDT), sonodynamic therapy (SDT), and photothermal therapy (PTT) have gained attention for their selectivity and low toxicity. Porphyrin-based compounds, with excellent photo/sono-sensitivity, are ideal for these therapies but face limitations like poor water solubility, aggregation, rapid metabolism, insufficient tumor targeting, and potential phototoxicity. Nanotechnology provides solutions: (1) Enhancing porphyrin's so

Fluorescence imaging-guided photodynamic therapy (PDT) is a promising modality for precision cancer treatment, yet most organic photosensitizers emitting in the visible or first near-infrared (NIR-I) region suffer from limited penetration, strong photon scattering, and high tissue autofluorescence. Organic fluorophores with second near-infrared emission (NIR-II, >1000 nm) mitigate these constraints, enabling deeper imaging, improved spatial resolution, and higher signal-to-background ratios for accurate tumor delineation, real-time therapeutic guidance and longitudinal monitoring of PDT respon

INTRODUCTION: Photodynamic therapy (PDT) and photobiomodulation (PBM) are light-based modalities with differing applications but which share overlapping spectral and irradiance requirements. PDT, which uses an exogenous photosensitizer, can be used to impart a selective cytotoxic response in target malignant tissues, while PBM is typically used to stimulate biological responses that are beneficial in healthy tissues. The inherent compatibility in light delivery requirements suggests the potential for exploring interactions between these modalities which could enhance tumor response to PDT.

Urothelial carcinoma of the bladder is one of the most common malignancies of the urinary tract. It remains a therapeutic challenge due to its high recurrence rate and resistance to conventional treatments. Photodynamic therapy (PDT) is an attractive, minimally invasive alternative that enables spatially controlled tumor ablation by activating photosensitizers (PS) with visible light, generating reactive oxygen species (ROS), mainly singlet oxygen (1O2), and inducing the targeted death of tumor cancer cells. However, due to poor aqueous solubility and aggregation of porphyrin-based PS, their c

Photoactive ruthenium polypyridyl complexes are intensively studied as imaging probes and (photo)therapeutics because of their attractive photophysical properties, including long-lived, oxygen-responsive luminescence, and their ability to sensitize reactive oxygen species (ROS) or to drive photoinduced electron or proton-coupled electron transfer (PCET), which may be advantageous in treating hypoxic tumors where oxygen-dependent therapies often fail. Despite their potential potency, a key limitation is their poor membrane permeation. This study focused on Ru(II) complexes containing the biolog

Novel pyrido[1,2-a]benzimidazole-fused tetrapyrrolic photosensitizers with an intense near-infrared absorption (711-747 nm) in the region of high transparency of biological tissues were prepared. Target photosensitizers demonstrated high ΦΔ values (0.49-0.60) resulted from the ability to form triplet states under irradiation. A test with nitroblue tetrazolium showed that in addition to singlet oxygen, they can generate type I reactive oxygen species such as the superoxide anion radical. Water-soluble quaternized forms of target photosensitizers demonstrated low cytotoxicity in the dark and sub

Photodynamic therapy (PDT) using ruthenium-based photosensitizers (PSs) has attracted increasing interest due to their favorable photophysical properties and their ability to generate reactive oxygen species (ROS) upon irradiation. Although PDT is generally considered a localized treatment, indirect effects on neighboring non-irradiated cells, known as bystander effects, may occur. In this study, we investigated, for the first time, bystander responses induced by the photoactivatable ruthenium nitrosyl complex {TPyP[Ru(NO₂)(bpy)₂]₄}(PF₆)₄ (RuNO₂TPyP) using two-dimensional (2D) and three-dimens

The treatment of breast cancer remains a significant clinical challenge, with limited responsiveness to conventional therapies. Photodynamic therapy, using the natural photosensitizer Hypericin, offers a promising modality, but its efficacy is limited by poor solubility, poor selectivity, and non-specific binding. To address these limitations, we developed a targeted nanoplatform leveraging the biocompatibility of gold nanoparticles and targeting capabilities of a monoclonal antibody. This study aimed to synthesize and characterize a novel nanoplatform comprising eco-friendly, green-synthesize

INTRODUCTION: Understanding how cells respond to internal and external inputs requires investigating cells within three-dimensional (3D) environments, which better mimic physiological conditions. Compared to two-dimensional (2D) systems, 3D cultures more accurately simulate tissue architecture, including cell-cell and cell-extracellular matrix interactions, as well as gradients of oxygen and nutrients. Despite these advantages, quantifying signaling dynamics in 3D remains difficult due to limitations in imaging depth, phototoxicity, and computational analysis.

To effectively inhibit the growth and recurrence of tumors, we proposed to design a new osmium (Os) complex integrating photodynamic therapy (PDT), chemotherapy, and immunotherapy. To this end, we optimized a series of Os(II) complexes derived from differently modified thiosemicarbazone ligands to obtain an Os(II) 2-quinoxalinecarbaldehyde thiosemicarbazone complex (Os3) with remarkable cytotoxicity and phototoxicity to cancer cells. The establishment of an MC38 colon cancer xenograft model demonstrated that Os3 not only significantly inhibited the growth and recurrence of tumors but also exhi

Azobenzene derivatives constitute a prototypical class of photoresponsive molecular switches with broad utility in synthetic chemistry and biomedical research, owing to their distinctive physicochemical properties. Recent molecular engineering has enabled red-shifted photoisomerization into the visible biological window, thereby enhancing tissue penetration and reducing phototoxicity. This review systematically surveys contemporary advances in azobenzene-based photoswitchable systems with a specific focus on medicinal chemistry and photopharmacology. Emphasis is placed on rational design strat

As typical compounds with aggregation induced emission (AIE) property, tetraphenylethylene-based photosensitizers with unique molecular structures and excellent photophysical properties have been widely applied in the bio-specific imaging, diagnosis, photodynamic therapy (PDT) and other related fields. In this work, we have developed a concise and efficient synthetic route to modify the structure of the AIE photosensitizer PS2, incorporating an aminomethyl side chain at the thiophene terminus. This modification offers a versatile platform for subsequent structural diversification. Following th

Melanoma remains a major public health concern due to rising incidence and frequent resistance to immune checkpoint inhibitors. Photodynamic therapy (PDT) has emerged as a promising complementary strategy that exerts direct cytotoxic effects while simultaneously reshaping the tumor microenvironment to enhance immune recognition. This study aimed to examine the effects of PDT on immune checkpoint expression and immunogenic cell death markers in melanoma cells. We performed in silico analysis using TCGA-SKCM and GTEx databases via GEPIA platform to identify dysregulated immune checkpoint molecul

Gastric cancer remains a cause of cancer mortality, and there is a need for locally controllable therapies that limit systemic toxicity and overcome heterogeneous responses. Photodynamic therapy (PDT) is attractive, yet efficacy is constrained by limited intracellular availability of photosensitizers. We tested whether small extracellular vesicles (sEVs) enhance hypericin (HYP) delivery and PDT responses in AGS cells, and whether cytotoxicity aligns with transcript and epigenetic remodeling. Human sEVs were loaded with HYP by probe sonication and characterized by DLS/ELS, TEM, and CD9/CD63/CD8

Light activation of photosensitizers, including verteporfin-conjugates entrapped in liposomes, can induce photochemical immunogenic cell death (ICD) in cancer cells by exposing damage-associated molecular patterns (DAMPs) under specific conditions. In this study, we investigated how incorporation of the clinical ionizable lipid SM-102 into light-activated liposomes containing the verteporfin-conjugate 20:0 BPD-PC effects key photochemical and photobiological attributes we previously identified to be strongly associated with photochemical ICD. We show that SM-102 incorporation increases the rat

Salt selection is critical in pharmaceutical development. Glutaric acid, an endogenous dicarboxylic acid, offers potential advantages as a counterion but is underexplored in pharmaceutical applications. The nonclinical safety of glutaric acid as a pharmaceutical counterion was assessed using LY3972406 hemi-glutarate hemi-hydrate (HG) as a model, with direct comparison to the LY3972406-hydrochloride (HCl) salt. Comprehensive safety assessments included genetic toxicology (Ames, in vitro and in vivo micronucleus tests), hazard evaluation (phototoxicity, and dermal and corneal irritation), and re

The skin microbiota is essential for cutaneous homeostasis, and prebiotics may support it. Nanostructured systems, such as cupuaçu-derived nanoparticles, enhance the delivery and stability of cosmetic active substances, but their role in modulating the skin microbiota remains unclear and requires targeted evaluation. In this study, lipid-polymeric hybrid nanoparticles based on cupuaçu (Theobroma grandiflorum) butter were developed as potential carriers for oligosaccharides. Three formulations were developed: cupuaçu butter (CB-NP), α-glucan oligosaccharide (AGO-CB-NP), and polydextrose nanopar
