The first pyrido[1,2-a]benzimidazole-fused porphyrazine photosensitizers with submicromolar photodynamic activity against cancer cells.

Novel pyrido[1,2-a]benzimidazole-fused tetrapyrrolic photosensitizers with an intense near-infrared absorption (711-747 nm) in the region of high transparency of biological tissues were prepared. Target photosensitizers demonstrated high ΦΔ values (0.49-0.60) resulted from the ability to form triplet states under irradiation. A test with nitroblue tetrazolium showed that in addition to singlet oxygen, they can generate type I reactive oxygen species such as the superoxide anion radical. Water-soluble quaternized forms of target photosensitizers demonstrated low cytotoxicity in the dark and sub

Bystander effects of a photoactivated ruthenium-based complex in 2D and 3D cell culture models.

Photodynamic therapy (PDT) using ruthenium-based photosensitizers (PSs) has attracted increasing interest due to their favorable photophysical properties and their ability to generate reactive oxygen species (ROS) upon irradiation. Although PDT is generally considered a localized treatment, indirect effects on neighboring non-irradiated cells, known as bystander effects, may occur. In this study, we investigated, for the first time, bystander responses induced by the photoactivatable ruthenium nitrosyl complex {TPyP[Ru(NO₂)(bpy)₂]₄}(PF₆)₄ (RuNO₂TPyP) using two-dimensional (2D) and three-dimens

Targeted photodynamic therapy for MCF-7 breast cancer using antibody-conjugated, green-synthesized gold nanoparticles loaded with hypericin: a proof of concept.

The treatment of breast cancer remains a significant clinical challenge, with limited responsiveness to conventional therapies. Photodynamic therapy, using the natural photosensitizer Hypericin, offers a promising modality, but its efficacy is limited by poor solubility, poor selectivity, and non-specific binding. To address these limitations, we developed a targeted nanoplatform leveraging the biocompatibility of gold nanoparticles and targeting capabilities of a monoclonal antibody. This study aimed to synthesize and characterize a novel nanoplatform comprising eco-friendly, green-synthesize

Quantifying single-cell responses to irradiation in 3D.

INTRODUCTION: Understanding how cells respond to internal and external inputs requires investigating cells within three-dimensional (3D) environments, which better mimic physiological conditions. Compared to two-dimensional (2D) systems, 3D cultures more accurately simulate tissue architecture, including cell-cell and cell-extracellular matrix interactions, as well as gradients of oxygen and nutrients. Despite these advantages, quantifying signaling dynamics in 3D remains difficult due to limitations in imaging depth, phototoxicity, and computational analysis. METHODS: We developed experiment

Designing an Osmium(II) Complex to Inhibit the Growth and Recurrence of Tumors by Integrating Photodynamic Therapy, Chemotherapy, and Immunotherapy.

To effectively inhibit the growth and recurrence of tumors, we proposed to design a new osmium (Os) complex integrating photodynamic therapy (PDT), chemotherapy, and immunotherapy. To this end, we optimized a series of Os(II) complexes derived from differently modified thiosemicarbazone ligands to obtain an Os(II) 2-quinoxalinecarbaldehyde thiosemicarbazone complex (Os3) with remarkable cytotoxicity and phototoxicity to cancer cells. The establishment of an MC38 colon cancer xenograft model demonstrated that Os3 not only significantly inhibited the growth and recurrence of tumors but also exhi

Synthesis and evaluation of mitochondrion-targeted tetraphenylethylene-based photosensitizer.

As typical compounds with aggregation induced emission (AIE) property, tetraphenylethylene-based photosensitizers with unique molecular structures and excellent photophysical properties have been widely applied in the bio-specific imaging, diagnosis, photodynamic therapy (PDT) and other related fields. In this work, we have developed a concise and efficient synthetic route to modify the structure of the AIE photosensitizer PS2, incorporating an aminomethyl side chain at the thiophene terminus. This modification offers a versatile platform for subsequent structural diversification. Following th

Photodynamic therapy with zinc phthalocyanine suppresses immune checkpoints in human melanoma cells.

Melanoma remains a major public health concern due to rising incidence and frequent resistance to immune checkpoint inhibitors. Photodynamic therapy (PDT) has emerged as a promising complementary strategy that exerts direct cytotoxic effects while simultaneously reshaping the tumor microenvironment to enhance immune recognition. This study aimed to examine the effects of PDT on immune checkpoint expression and immunogenic cell death markers in melanoma cells. We performed in silico analysis using TCGA-SKCM and GTEx databases via GEPIA platform to identify dysregulated immune checkpoint molecul

Biophysical and Raman Spectroscopy assessment of drug-induced photosensitivity: Insights from a randomised controlled pilot study.

BACKGROUND: Drug-induced photosensitivity is a recognised adverse reaction, yet systematic quantification remains limited. This study investigates the quantitative impact of commonly prescribed drugs on human skin physiology following oral intake and subsequent UV exposure. METHODS: Ten drugs with known photosensitising potential were selected: acetylsalicylic acid, amiodarone, diclofenac, doxycycline, enalapril, furosemide, hydrochlorothiazide, pantoprazole, verapamil, and voriconazole. Twelve healthy volunteers received five allocated substances, separated by washout periods. Defined skin a

Hypericin-loaded small extracellular vesicles increase photodynamic therapy and remodel miRNA-mRNA axes in AGS gastric cancer cells.

Gastric cancer remains a cause of cancer mortality, and there is a need for locally controllable therapies that limit systemic toxicity and overcome heterogeneous responses. Photodynamic therapy (PDT) is attractive, yet efficacy is constrained by limited intracellular availability of photosensitizers. We tested whether small extracellular vesicles (sEVs) enhance hypericin (HYP) delivery and PDT responses in AGS cells, and whether cytotoxicity aligns with transcript and epigenetic remodeling. Human sEVs were loaded with HYP by probe sonication and characterized by DLS/ELS, TEM, and CD9/CD63/CD8

The clinical ionizable lipid SM-102 enhances photochemical immunogenic cell death by verteporfin-conjugated liposomes and improves survival in vivo.

Light activation of photosensitizers, including verteporfin-conjugates entrapped in liposomes, can induce photochemical immunogenic cell death (ICD) in cancer cells by exposing damage-associated molecular patterns (DAMPs) under specific conditions. In this study, we investigated how incorporation of the clinical ionizable lipid SM-102 into light-activated liposomes containing the verteporfin-conjugate 20:0 BPD-PC effects key photochemical and photobiological attributes we previously identified to be strongly associated with photochemical ICD. We show that SM-102 incorporation increases the rat

Safety evaluation of glutaric acid as a novel pharmaceutical salt for first-in-human dosing.

Salt selection is critical in pharmaceutical development. Glutaric acid, an endogenous dicarboxylic acid, offers potential advantages as a counterion but is underexplored in pharmaceutical applications. The nonclinical safety of glutaric acid as a pharmaceutical counterion was assessed using LY3972406 hemi-glutarate hemi-hydrate (HG) as a model, with direct comparison to the LY3972406-hydrochloride (HCl) salt. Comprehensive safety assessments included genetic toxicology (Ames, in vitro and in vivo micronucleus tests), hazard evaluation (phototoxicity, and dermal and corneal irritation), and re

Cupuaçu (Theobroma grandiflorum) butter nanoparticles associated with oligosaccharides as cosmetic carriers for prebiotic effect: development, physicochemical characterization, and preclinical evaluation.

The skin microbiota is essential for cutaneous homeostasis, and prebiotics may support it. Nanostructured systems, such as cupuaçu-derived nanoparticles, enhance the delivery and stability of cosmetic active substances, but their role in modulating the skin microbiota remains unclear and requires targeted evaluation. In this study, lipid-polymeric hybrid nanoparticles based on cupuaçu (Theobroma grandiflorum) butter were developed as potential carriers for oligosaccharides. Three formulations were developed: cupuaçu butter (CB-NP), α-glucan oligosaccharide (AGO-CB-NP), and polydextrose nanopar

Structural, Compositional, and Dielectric State Profiling in Label-Free Single-Cell Monitoring.

Individual cells sense and transition between functional states, and the distribution of these states over time determines drug response, disease progression, and cell manufacturing outcomes. However, repeated measurement is difficult with label-based acquisition, as photobleaching, phototoxicity, and probe-induced perturbation accumulate. Label-free monitoring that leverages intrinsic physical signals circumvents these constraints, shifting the analytical burden from label chemistry to instrumental drift. In this review, we organize this field into three measurement modalities, including imag

Environmental Concentrations of Herbicide Prometryn Increase the Susceptibility of Reef-Building Corals to Nitrate Stress.

Herbicide and nitrate pollution frequently co-occur in coastal waters, while their combined effect on corals has been a critical knowledge gap. Although corals can exhibit acclimation to nitrate enrichment, it is unclear whether this acclimation persists under coexposure to PSII herbicides. Here, we investigated the combined effects of nitrate and the PSII herbicide prometryn on the reef-building coralAcropora hyacinthusby integrating physiology, ultrastructure, and host-symbiont multiomics. The results demonstrate that prometryn acts as the primary driver of photodamage and photosynthetic tox

Smart cyclometalated iridium photosensitizers: stimuli-responsive and theranostic systems for precision phototherapy.

Cyclometalated Iridium(III) complexes have emerged as one of the most versatile classes of photosensitizers for photodynamic therapy (PDT), owing to their tunable photophysics, high phosphorescence quantum yields, and intrinsic capacity for real-time optical imaging. However, the clinical translation of conventional PDT remains limited by nonspecific activation, poor selectivity, and off-target phototoxicity. The dynamic and heterogeneous tumour microenvironment (TME)- defined by anomalous pH, elevated Glutathione (GSH) levels, malfunctioned redox homeostasis, enzyme overexpression, and irregu

A Molecular Engineering Strategy to Fine-Tune Phototoxicity of AIE Probes for Super-Resolution Imaging of Mitochondrial Cristae Dynamics.

Understanding mitochondrial morphology and function is critical for investigating mitochondrial diseases, yet fluorescent probes that label the inner mitochondrial membrane (IMM) and report dysfunction at high spatiotemporal resolution remain limited. Here, we rationally designed aggregation-induced emission-active probes with tunable alkyl chain lengths and carboxyl groups, allowing us to fine‑tune phototoxicity and achieve a controlled, mild level of reactive oxygen species (ROS) generation. Two probes, named OTS-7C and OTS-12C, enabled super‑resolution imaging of mitochondrial cristae using

Discovery of ALA-hydroxypyridinone hybrids: novel strategies for photodynamic therapy against superficial tumours.

Superficial tumours can be effectively treated with photodynamic therapy (PDT), which is characterised by its targeted approach and enhanced safety profile. 5-aminolevulinic acid (ALA), a precursor to the endogenous photosensitiser PpIX, has been widely utilised in PDT. However, its high hydrophilicity, poor permeability and stability, necessitates structural modifications to improve its drug-like properties and increase the PpIX yield. Hydroxypyridinone (HPO) iron chelators can synergistically enhance PpIX production when administered in conjunction with ALA, as well as inducing tumour cell d

Dual-masked cathepsin B-activated 5-aminolevulinic acid prodrugs with enhanced photodynamic cytotoxicity.

5-Aminolevulinic acid-mediated photodynamic therapy (ALA-PDT) exploits tumor-selective accumulation of protoporphyrin IX (PpIX). However, its clinical utility is constrained by poor physicochemical properties and suboptimal tumor selectivity. To address these limitations, we rationally designed and synthesized 17 N-terminal dipeptide-modified 5-ALA prodrugs using a dual-masked strategy. This approach integrates lipophilic shielding at the carboxyl terminus with a cathepsin B (Cath B)-cleavable dipeptide at the amino terminus, enabling tumor-specific release of active 5-ALA. Formulation development and optimization of sparfloxacin-loaded solid lipid nanoparticles integrated with microneedles for the treatment of biofilm-infected wounds.

The efficient management of infected skin wounds is severely limited by the thick bacterial biofilm within the lesion, the intractable stratum corneum barrier of the perilesional skin, and the high toxicities of potent antibiotics. Sparfloxacin (SPFX) was withdrawn from the market due to its severe systemic side effects and UV-induced phototoxicity, while its severe hydrophobicity causes localized crystalline cytotoxicity upon topical application. To safely rejuvenate this potent drug, an integrated transdermal system combining photoshielding, SPFX-loaded solid lipid nanoparticles (SPFX-SLNs)
