NIR pH-Responsive PEGylated PLGA Nanoparticles as Effective Phototoxic Agents in Resistant PDAC Cells.

Pancreatic ductal adenocarcinoma (PDAC) is one of the deadliest cancers worldwide due to its resistance to conventional therapies that is attributed to its dense and acidic tumor microenvironment. Chemotherapy based on gemcitabine usually lacks efficacy due to poor drug penetration and the metabolic characteristics of the cells adapted to grow at a more acidic pHe, thus presenting a more aggressive phenotype. In this context, photodynamic therapy (PDT) offers a promising alternative since it generally does not suffer from the same patterns of cross-resistance observed with chemotherapy drugs.

Pyrazolo[1,5-a]pyrimidine-Based Type-I Photosensitizer as an Efficient Pyroptosis Inducer for Tumor Ablation.

Pyroptosis is a proinflammatory and lytic programmed cell death form, which can promote cytotoxic T lymphocyte (CTL) maturation and tumor infiltration through the release of damage-associated molecular patterns (DAMPs). Therefore, the induction of pyroptosis by small molecules is a promising strategy to activate antitumor immunity. In this work, we report the design of a new class of pyrazolo[1,5-a]pyrimidine-based type-I photosensitizers (PSs) as efficient pyroptosis inducers for cancer photodynamic therapy (PDT). Among the compounds, ZS-3 exhibited the most excellent reactive oxygen species

Assessing phototoxic drug properties of hydrochlorothiazide using human skin biopsies.

The diuretic drug hydrochlorothiazide (HCT) is associated with an increased risk of non-melanoma skin cancer upon UV exposure. The underlying cellular and molecular mechanisms behind this association remain elusive. Herein, a human skin model to assess the photocarcinogenic effects of HCT is established. Skin biopsies collected from human body donors are treated with HCT and irradiated with 300 mJ/cm2 low dose UVA or UVB or with 5 J/cm2 high dose UVA. In HCT-treated biopsies but not in control, low dose UVA irradiation results in activation and nuclear translocation of the tumor-suppressor pro

Novel Molecular Weight Gradient Hyaluronate Dissolving Microneedles for Sustained Intralesional Delivery and Photodynamic Activation of Hematoporphyrin in Port-Wine Stain Therapy.

Port-wine stain (PWS), a progressive congenital vascular malformation characterized by ectatic dermal capillaries, demonstrates age-dependent lesion expansion and chromatic intensification, resulting in significant psychosocial comorbidity. While systemic hematoporphyrin (HP) administration remains the clinical paradigm for photodynamic therapy (PDT), its therapeutic utility is severely constrained by non-targeted biodistribution. Pharmacokinetic analyses reveal prolonged dermal retention and suboptimal lesion accumulation, predisposing 42% of patients to phototoxic reactions. To address these

Photo-responsive antibacterial metal organic frameworks.

The misuse and overuse of antibiotics have caused the emergence of antibiotic-resistant bacteria, making bacterial infections more challenging. The increasing prevalence of multidrug-resistant pathogens has driven researchers to explore novel therapeutic strategies. Phototherapy strategies that utilize photo-responsive biomaterials for their antibacterial properties have gained widespread attention due to their capability of precisely controlling bacterial inactivation with minimal side effects. Despite their potential, photodynamic therapies suffer from phototoxicity and low efficiency of pho

Photo-controlled co-delivery of verteporfin and acriflavine via platelets achieves potentiated glioblastoma-targeted photodynamic therapy.

The potential of glioblastoma (GBM) photodynamic therapy (PDT) is limited by inadequate GBM drug delivery, and the development of resistance to PDT as a result of cellular damage response that critically involves the hypoxia-inducible factor-1α (HIF-1α) and yes-associated protein (YAP). Herein, addressing these challenges, we demonstrated a strategy of photo-controlled, targeted co-delivery of verteporfin (Vp), a photosensitizer and YAP inhibitor as well, and acriflavine (Af), a HIF-1α inhibitor via platelets for enhanced GBM PDT. Mouse platelets were separately loaded with Vp (Vp@Plt) and Af

Greenness assessment and phototoxicity of rose bengal and methylene blue on immature aquatic stages of malaria vector Anopheles pharoensis.

This study systematically evaluated the efficiency of rose bengal and methylene blue as photosensitizers against the immature aquatic stages of Anopheles pharoensis. Genetic identification using the COI partial sequence confirmed the species, and the obtained sequence was submitted to GenBank (Accession No. PQ346929). Both photosensitizers exhibited 100% mortality in larvae I within 24 h at their highest concentrations, demonstrating strong biocidal activity. LC50 values for rose bengal increased from 1.50 ppm (24 h) and 1.34 ppm (48 h) in larvae I to 3.83 ppm (24 h) and 3.12 ppm (48 h) in pup

A Reductively Activatable Phototheranostic Agent Based on a Water-Soluble Nickel Naphthalocyanine Precursor.

Metal naphthalocyanine (MNc) is one of the most promising near-infrared (NIR) dyes as a photosensitizer for phototheranostics, combining photoacoustic imaging (PAI) and photothermal therapy (PTT). However, the extremely low solubility of MNc has limited its practical application as a theranostic agent. In this research, we synthesized NiNc precursors (ox-NiNc) having two alkoxy groups on the α-carbons of the diagonal pyrroles with significantly improved solubility. We also prepared an amphiphilic derivative, ox-NiNc-PEG, and investigated its conversion to NiNc through reductive aromatization.

Investigation of oxidative-stress-induced axon beading using photo-oxidation.

Oxidative stress caused by excessive reactive oxygen species adversely affects cellular function. Here, we investigate axonal beading induced by the photo-oxidation of a membrane-based fluorescent dye. Our experiments show that, apart from using oxygen scavengers, chelation of free calcium, or stabilization of microtubules or actin filaments by pharmacological agents can mitigate oxidative-stress-induced beading. Furthermore, we take advantage of this light-induced oxidative stress to explore axonal responses to spatially confined perturbations. We demonstrate that low excitation causes locali

Fibrin-targeted photosensitizer aggregates significantly enhance the delivery of chemotherapeutic drugs to tumor vasculature.

Current drug carriers, while effective in mitigating therapeutic side effects, frequently elicit non-drug-related adverse reactions, including hepatorenal toxicity, oxidative stress and allergic responses. To address these issues and enhance drug efficacy, we propose a "drug-carrying-drug" strategy that integrates photodynamic therapy (PDT) with chemotherapy for potential tumor eradication. Traditional photosensitizers often confront a paradoxical dilemma between aggregation-induced photodynamic deactivation and dispersion-mediated premature photobleaching. To overcome these limitations, we de

A mono-substituted aluminum phthalocyanine photosensitizer with high phototoxicity index for effective bladder cancer treatment.

Photosensitizers are central to the efficacy of photodynamic therapy (PDT) in cancer treatment. The phototoxic index (PI) defines the therapeutic window of a photosensitizer, with the clinically used Photosens® exhibiting a PI of 725. In our study, we observed that Zn(II) phthalocyanine mono-β-substituted with 4-carboxyphenoxy (ZnPc-COOH) displayed a phototoxic IC50 in bladder cancer T24 cells at approximately 11.2 nM, yet its dark toxicity was notably high, yielding a PI of only 199. We hypothesize that the elevated dark toxicity is linked to mitochondrial localization. Rather than employing

Boosting PDT with DPA-NI-Bu: high photocytotoxicity through redox homeostasis perturbation.

Photodynamic therapy (PDT) is a promising cancer treatment that relies on reactive oxygen species (ROS) to disrupt cellular redox homeostasis, ultimately leading to cell death. The thioredoxin (Trx) system is a pivotal regulatory system for antioxidant defence, which plays a key role in immune response and cell death. Thus, perturbating the Trx system could enhance the efficacy of PDT. Naphthalimide skeletons are research hotspots in photosensitizers due to their tunable photophysical properties and high ROS yield. A series of novel photosensitizers based on naphthalimide skeletons were design

Tumor microenvironment responsive smart nanoplatform for synergistic tumor therapy through co-enhancement of GSH depletion and hypoxia relief.

As innovative therapeutic strategy, chemodynamic therapy (CDT) and photodynamic therapy (PDT) show great promise for tumor therapy. However, their therapeutic efficacy is seriously limited by the hypoxia and insufficient H2O2 supply of the tumor microenvironment. Juglone (JUG), a naturally-occurring naphthoquinone, can increase the intracellular H2O2 concentration and serve as an inhibitor of peptidyl-prolyl cis-trans isomerase NIMA-interacting 1. Herein, a nanoplatform composed of Fe-based metal-organic framework core, MnO2 layer, JUG payload, and hyaluronic acid (HA) decoration (Fe-TCPP@MnO2

Advances in Stimuli-Responsive Release Strategies for Sonosensitizers in Synergistic Sonodynamic Immunotherapy against Tumors.

Despite intensive efforts to develop diagnostic and therapeutic tools, the successful treatment of cancer is still hampered by uncontrolled tumor proliferation and metastasis. Sonodynamic therapy (SDT) is a cutting-edge, noninvasive treatment modality that activates sonosensitizers via low-intensity ultrasound (US) to generate reactive oxygen species (ROS), offering deep tissue penetration, low phototoxicity, and minimal side effects. Beyond direct cytotoxicity, it can trigger immunogenic cell death (ICD), thereby enhancing systemic antitumor immune responses. However, its efficacy is constrai

Acousto-optogenetics bandpass stabilizer: A programmable platform for mapping single-cell phenotypic life trajectories.

Photobleaching-induced signal decay undermines the accuracy of fluorescence-based drug screening across multiple disease models (e.g., HER2/EGFR-targeted therapies). Traditional phototoxicity mitigation strategies, primarily through reduced illumination, inevitably compromise imaging resolution and further providing false positive/false negative. Here, we introduce an innovative active photobleaching intervention strategy to solve these problems with the potential for broad applicability across a wide range of cell species. We employ the ultra-low-frequency (500-2000 Hz) acoustic modulation to

Facile synthesis and biological evaluation of [a]-benzannulated BODIPY derivatives as novel heavy-atom-free photosensitizers for photodynamic anticancer therapy.

Boron dipyrromethenes (BODIPYs) have garnered significant attention for their broad applications in photodynamic anticancer therapy. However, the development of these materials remains a challenging task, partially due to their low singlet oxygen (1O2) quantum yield usually require incorporating heavy atoms and high laser power density to produce thermal energy and synergistically enhance photodynamic therapy (PDT). Concerns about potential dark toxicity of heavy atoms and the strong photothermal radiation causing acute pain significantly reduces patient compliance, hinders the broader clinica

Metabolism of Oxybenzone and Dioxybenzone to Phototoxins in Human Skin Cells.

Recent research indicated that exposure of sea anemones in full-spectrum sunlight to the UV filter, oxybenzone, induced increased mortality attributable to the production of phototoxic glucoside metabolites. This study evaluated whether exposure of oxybenzone and dioxybenzone to human skin cell models formed analogous glucuronide conjugates and induced a reduction in cell viability under full-spectrum sunlight. In dose-response evaluations with two human skin cell models, increasing doses of oxybenzone or dioxybenzone (up to 0.1%) or increasing exposure times (up to 24 h) were associated with

Label-Free Longitudinal Imaging of Single Cell Drug Response with a 3D-Printed Cell Culture Platform.

Image-based phenotypic screening has emerged as a powerful tool for revealing single-cell heterogeneity and dynamic phenotypic responses in preclinical drug discovery. Compared to traditional static end-point assays, live-cell longitudinal imaging captures the temporal trajectories of individual cells, including transient morphological adaptations, motility shifts, and divergent subpopulation behaviors, enabling high content features and more robust early prediction of treatment outcomes. Fluorescence-based screening, while highly specific, is constrained in live-cell contexts by broad spectra

Photosensitization and phototoxicity mechanism of diclofenac sodium (NSAID) under ambient UVR exposure.

Diclofenac sodium (DS) is used for pain, inflammation and musculoskeletal disorders. Despite its absorption of UV radiation, the phototoxic potential and molecular mechanisms in keratinocytes under UVA and UVB exposures are largely unexplored. In this study, we have employed a comprehensive array of in-silico, in-chemico, and in-vitro approaches to assess DS's photostability, phototoxic, and photogenotoxic properties under UV irradiation. Our findings indicate that DS undergoes photodegradation, forming 3 photoproducts. Further, docking studies revealed DS's affinity for DNA and Bax, implicati
