Chronic ultraviolet B radiation-induced biochemical changes in the skin of hairless mice.

Skh:HR-1 hairless mice were irradiated chronically with sub-erythemal doses of UVB radiation, and a number of biochemical parameters in the skin were determined after 6, 12, 18, and 24 wk of exposure. The parameters measured were water, collagen, elastin, and glycosaminoglycan content; collagenase and elastase levels; and Bz-Tyr-OEt (N-benzoyl-L-tyrosine ethyl ester) and BAPNA (alpha-N-benzoyl-DL-arginine-p-nitroanilide) hydrolyzing activities. Data for UVB radiation-exposed and chronological age-matched control mice were compared with respect to unit area and to unit mass of skin. On a unit a

Common cutaneous disorders in athletes.

Athletic activity may cause or aggravate skin disorders, which in turn may diminish athletic performance. Since many sporting activities necessitate prolonged exposure to the sun, athletes must avoid painful sunburn which will adversely affect their performance. Drugs and chemicals also may cause photoallergic and/or phototoxic reactions, including polymorphous light eruption and athletes should thus avoid photosensitising drugs and chemicals. The effects of chronic ultraviolet exposure include ageing, pigmentation and skin cancers. The most effective protection against excessive exposure to s

Mechanisms of phototoxicity in porphyria cutanea tarda and erythropoietic protoporphyria.

The porphyrias are the only group of diseases caused by endogenous phototoxic agents. While patients with erythropoietic protoporphyria and those with porphyria cutanea tarda both have skin lesions on sun-exposed areas, there are differences in their cutaneous manifestations. Based on information discussed in this chapter, the following pathophysiologic mechanisms can be proposed. In porphyria cutanea tarda, photoactivation of the complement system in the presence of uroporphyrin results in activation of dermal mast cells, which release their proteases. This results in dermal-epidermal separat

Actinic DNA damage and the pathogenesis of cutaneous malignant melanoma.

The near epidemic of melanoma and non-melanoma skin cancer in the United States and certain other industrialized nations is attributable to cutaneous exposure to sunlight more than to any other factor. Chronic exposure to UV irradiation and a high total cumulative dose may be less deleterious than are periodic bursts of large amounts of sun exposure leading to severe sunburn. Such an exposure pattern is characteristic of individuals such as office workers whose outdoor activities are irregular rather than daily, as with farmers or fisherman. Although UV irradiation is injurious to many cellula

Collagen metabolism in ultraviolet irradiated hairless mouse skin and its correlation to histochemical observations.

Early biochemical studies of ultraviolet (UV) irradiated human skin reported a loss of insoluble collagen with a concomitant increase in the soluble fraction. Recent work has described an early increase in type III collagen during chronic irradiation of hairless mice as determined by cyanogen bromide digests of whole skin. In order to understand the correlation of these events and those seen with histochemistry, in the present study we irradiated hairless mice for up to 24 weeks with approximately 4 minimal erythema doses (MEDs) of UVB thrice weekly with Westinghouse FS-40 bulbs. Skin samples

[Effect of directed infrared radiation on the skin and eye of rabbits after acute and chronic exposure].

The establish a threshold dose, an acute experiment was carried out by 30 min. radiation of the rabbit's shaved head with all ranges of infrared, 200 W/m2, 350 W/m2 and 500 W/m2 intensities. Irradiation was prolonged up to 2 hrs. Having established the threshold dose, a chronic (5 months') experiment was carried out, irradiating 9 rabbits' skin (in 3 groups) with 350 W/m2 intensity in the range of all three bands. Similarly, tests on the eye transparent part were performed, irradiating only one eye, the other being handled as the control. The results show that the 350 W/m2 intensity of IR-A an

Nalidixic acid-induced photosensitivity in mice: a model for pseudoporphyria.

Porphyria cutanea tarda (PCT)-like photosensitivity eruptions have occurred in patients treated with nalidixic acid, furosemide (Lasix), or tetracycline. An animal model for nalidixic acid photosensitivity was developed in young CF-1 female mice. The hair on the back was plucked from groups of animals that were injected i.p. each day with nalidixic acid or saline. The animals were exposed to black-lamp radiation for 12 h daily for 6 weeks, followed by a 2-week rest period, and then 4 more weeks of UV radiation exposure. The nalidixic acid-treated animals developed far greater gross and chronic

Cellular hypersensitivity to UV-A: a clue to the aetiology of actinic reticuloid?

Fibroblasts cultured from six patients with actinic reticuloid (AR) showed striking cytopathic changes and inhibition of RNA synthesis after exposure to near-ultraviolet radiation that had no effect on normal and other photosensitive cell strains, such as those of xeroderma pigmentosum and Bloom syndrome. An abnormal pattern of DNA fragmentation was observed after doses insufficient to cause cytopathic effects. These results suggest a cellular defect in the prevention or repair of some damage caused by free radicals and other photoproducts. In order to explain the pathogenesis of AR, it is pro

Solar radiation and malignant melanoma of the skin.

Several observations suggest that a majority of cases of malignant melanoma of the skin are linked to sun exposure. Evidence includes higher occurrence of melanoma on anatomic areas heavily exposed during recreation, development of melanoma more frequently in lightly pigmented persons, and correlation of melanoma incidence and mortality with proximity to the equator. The role of the sun exposure in the pathogenesis of melanoma remains unclear, however. Many cases of melanoma may be related to heavy doses of solar radiation received during recreation. Chronic sun exposure is not so clearly link
