Laryngo-onycho-cutaneous syndrome (LOCS), or Shabbir's syndrome, linked to mutations in the LAMA3 gene, is predominantly observed in children from Punjabi families. We present the case of a 2.5-year-old male who was initially misdiagnosed with asthma due to progressive hoarseness and stridor. Further evaluation revealed biphasic stridor, paronychia, conjunctival inflammation, nail dystrophy and widespread mucocutaneous lesions. Laryngoscopy showed glottic granulation tissue and subglottic stenosis. While genetic testing was unavailable due to resource limitations, histopathological findings su

Dystrophic epidermolysis bullosa (DEB) is a clinically heterogeneous blistering disease caused by pathogenic variants in the COL7A1 gene. Phenotypes range from localized to generalized and are primarily determined by the variant type, though additional modifiers influence disease severity. To characterize the molecular basis of the disease phenotype in a diabetic patient presenting with localized skin blistering and nail dystrophy. Skin biopsies were taken from the index patient for immunofluorescence mapping and keratinocyte culture. Exome sequencing was used to screen for pathogenic variants

We reported a 15-year-old boy with extensive linear porokeratosis confirmed by clinical, histopathologic, and immunohistochemical assessment. Ixekizumab treatment lead to a near-complete clearance of lesions by 3 months, while nail dystrophy showed minimal response. The presence of IL-17A-positive infiltrates supported the immunologic basis for therapeutic efficacy. These findings suggest that ixekizumab is a promising targeted therapy for refractory linear porokeratosis.

Congenital malalignment of the great toenail (CMGT) is a rare nail disorder characterized by lateral deviation of the nail plate due to malrotation of the nail matrix. We describe the case of a female adolescent with progressive nail discoloration, abnormal growth, and recurrent shedding of the great toenails. Similar findings were also observed in other family members, suggesting a hereditary predisposition. Fungal studies were negative. The patient was managed conservatively with protective footwear and regular trimming, while surgical matricectomy was declined. This case underscores the imp

Background/Objectives: Keratitis-ichthyosis-deafness (KID) syndrome is an exceptionally rare congenital multisystem disorder, with an estimated prevalence below 1:1,000,000 and fewer than 100 reported cases worldwide. It is characterized by hyperkeratosis, alopecia, nail dystrophy, hearing loss, and ocular involvement. While dermatological management is well described, surgical experience with wound treatment in KID syndrome remains extremely limited. The objective of this report is to describe the surgical management and outcomes of a patient with chronic lower-limb wounds associated with KID

Poikiloderma with neutropenia is a genetic disorder characterized by skin abnormalities, nail dystrophy, bone anomalies, and neutropenia. USB1 encodes a phosphodiesterase essential for processing spliceosomal U6 RNA and some microRNAs, regulating their stability. This study describes a heterozygous de novo USB1 variant (p.P44L) identified in a patient with recurrent infections, hypogammaglobulinemia, and low neutrophil counts. Unlike previously reported mutations, p.P44L affects a conserved proline in the N-terminal domain, predicted to be critical for protein interactions and stability. Funct

In recent decades, cancer mortality rates have declined due to development of systemic antineoplastic agents targeting specific cancer pathways. Nail toxicities associated with these therapies are underrecognized by many and contribute significantly to patient morbidity. Nail changes from chemotherapy occur due to toxicity to continuously dividing nail matrix keratinocytes. Preventive measures, including avoiding repeated trauma from manicuring and artificial nails, regular nail trimming, and applying emollients to cuticles are essential in mitigating nail toxicities. Frozen gloves or socks ha

Onycholysis is a disorder of the nails, described as the separation of the nail bed from the nail plate. While onycholysis most commonly occurs due to trauma to the nail, such as after manicures or chemical exposure, it can also result from drug-induced photosensitivity. Many medications have been linked to onycholysis, including tetracyclines, psoralens, and retinoids, which are synthetic derivatives of vitamin A. Oral retinoid use has been associated with various nail changes, but there are no reports of onycholysis resulting from oral vitamin A intake. Thus, we present the case of an 85-yea

Chronic recurrent multifocal osteomyelitis (CRMO) is a rare autoinflammatory bone disorder that predominantly affects children, and approximately 25% patients also present with an associated inflammatory condition. Most patients respond to nonsteroidal anti-inflammatory drugs, but refractory cases may need corticosteroids, bisphosphonate therapy, or disease-modifying antirheumatic drugs (DMARDs) like methotrexate, sulfasalazine, tumor necrosis factor-alpha (TNF-α) inhibitors, or even anti-interleukin-1 (anti-IL-1) blockers. We report the case of a previously healthy nine-year-old girl presenti

INTRODUCTION: Enthusiasm for manicures is at a high amongst young populations and has led to a growth in nail cosmetology in the last decade. Reviewing the known nail and cutaneous adverse outcomes associated with manicures can improve counseling for safe usage.
AIMS: To provide a comprehensive review of cutaneous disorders reported with various types of nail cosmetics, to allow consumers to make informed decisions when choosing a manicure type. In addition, this article promotes cognizance amongst dermatologists of potential causes of dermatologic conditions associated with nail cosmetics.

A man in his 50s develops signs of immune dysfunction (recurrent chest infections and new-onset chronic diarrhoea), alongside a history of multiple distinct malignancies and nail dystrophy. Immunological testing reveals T-cell lymphopenia and hypogammaglobulinaemia. Whole genome sequencing reveals a heterozygous c.1465del mutation in the FOXN1 (forkhead box N1) gene. This case suggests that previously healthy carriers of heterozygous FOXN1 mutations may be at risk of developing immune dysfunction in later life. Potential reasons for the severe and delayed phenotype include a dominant-negative

Dear Editor, Genodermatoses are usually diagnosed in early childhood but diagnosis may be delayed in rare cases. Schöpf-Schulz-Passarge syndrome (SSPS) first described in 1971 is a rare type of autosomal recessive ectodermal dysplasia characterized by palmoplantar keratoderma, periocular and eyelid apocrine hidrocystomas, hypodontia, hypotrichosis, and nail dystrophy (1). Fewer than 40 cases have been reported up to 2018 (2) and the number has slightly increased with subsequent case reports. We report here a 73-year-old male patient whose parents were third-degree cousins presented to dermatol

Stevens-Johnson syndrome (SJS) and toxic epidermal necrolysis (TEN) overlap syndrome, characterized by extensive epidermal necrosis, represents a life-threatening severe dermatological disorder.Therapeutic agents and regimens for this condition include high-dose glucocorticoids, intravenous immunoglobulin (IVIG), plasma exchange, immunosuppressants, and TNF-α inhibitors. A study demonstrates that JAK/STAT hyperactivation-characterized by interferon signature enrichment, STAT1 phosphorylation in keratinocytes/macrophages, and subsequent GBP1/WARS1-mediated cytotoxicity-drives epidermal detachme

Lichen planus pigmentosus (LPP), a rare variant of lichen planus, characterized by the insidious onset of dark brown to gray macules. In this report, we present a unique case of LPP in a 30-year-old female patient who developed diffuse hyperpigmented macules with nail dystrophy. Notably, histopathology revealed classic lichenoid interface dermatitis with predominant plasma cell infiltration in the papillary dermis, confirmed by strong CD38 and CD138 immunoreactivity. Laboratory evaluation revealed elevated serum Kappa light chain and thyroid stimulating hormone levels, raising the possibility

In laparoscopy, specimens have to be removed from the abdominal cavity. If the trocar opening or the vaginal outlet is insufficient to pass the specimen, the specimen needs to be reduced. The power morcellator is an instrument with a fast rotating cylindrical knife which aims to divide the tissue into smaller pieces or fragments. The Food and Drug Administration (FDA) issued a press release in April 2014 that discouraged the use of these power morcellators. This article has the objective to review the literature related to complications by power morcellation of uterine fibroids in laparoscopy

Leprosy is a chronic granulomatous infectious disease caused by Mycobacterium leprae (M. leprae) and/or M. lepromatosis that has a wide spectrum of clinical presentations and multi-organ involvement, primarily on the skin and peripheral nerves. Recent studies have addressed the possibility of nail involvement in leprosy, but to this day limited studies have provided histological evidence confirming the presence of acid-fast bacilli (AFB) in the nail apparatus. A case of lepromatous leprosy-related dystrophy in 20 nails in a 37-year-old male was reported. Physical examination revealed, on all 2
