Genetic associations between polyunsaturated fatty acids and alopecia areata.

The influence of circulating polyunsaturated fatty acids (PUFAs) on alopecia areata (AA) development remains poorly understood, with inconsistent results reported. Our study aims to evaluate their causal associations using the bidirectional Mendelian randomization method. European ancestry genome-wide association studies were used to analyze the genetic associations between PUFAs and AA (n = 211,428). Sample sizes for PUFAs were 13,527, 13,506, 114,999, 7818, and 13,549 for plasma linoleic acid (LA), omega-6, omega-3, docosahexaenoic acid (DHA), and PUFAs other than 18:2 (otPUFA), respectively

Depression and anxiety in visible skin diseases affecting the hands and face: a systematic literature review.

BACKGROUND: Visible skin diseases affecting the face and hands-crucial for non-verbal communication and social interaction-can deeply impact psychological well-being. Beyond physical symptoms, their visibility often affects self-esteem and social functioning, contributing to high rates of depression and anxiety. While facial dermatoses have been extensively studied, few investigations have explored the psychological impact of visible localization in other dermatoses. OBJECTIVE: To estimate the prevalence of depression and anxiety in patients with visible skin diseases, identify associated fac

Latent Profiles of Perceived Stress and Their Predictors in Alopecia Areata: A Cross-Sectional Study.

BACKGROUND: Alopecia areata (AA) causes significant psychological distress. Standard assessments often classify patient stress as merely high or low, overlooking key differences in stress experiences, such as anxiety versus helplessness. This study employed a person-centered method to identify distinct subgroups of AA patients based on their unique patterns of anxiety-related stress. METHODS: A cross-sectional survey of adults with AA was conducted using the validated Chinese version of the 14-item Perceived Stress Scale (PSS-14). Latent profile analysis (LPA) was applied to the PSS-14 respon

The Role of Inflammasome in the Pathogenesis of Alopecia Areata: NLRP3 and Its Cascade Contribute to the Onset and Early Stage of Alopecia Areata.

BACKGROUND: The inflammasome, a multiprotein complex, is crucial in the pathogenesis of various autoinflammatory disorders. In particular, the NOD-like receptor family pyrin domain-containing 3 (NLRP3) inflammasome has recently been associated with diseases such as psoriasis, vitiligo, lupus, and alopecia areata (AA). OBJECTIVE: The objective of this study is to investigate the potential involvement of the inflammasome in the pathogenesis of AA and to determine its correlation with clinical manifestations. METHODS: A total of 144 patients with AA and 22 healthy controls were included. Scalp

Woodhouse-Sakati Syndrome Due to the Rare DCAF17 c.321+1G>A Mutation: The Second Case Report Worldwide.

Woodhouse-Sakati syndrome (WSS) is a rare autosomal recessive neuroendocrine disorder caused by pathogenic variants in the DDB1- and CUL4-associated factor 17 (DCAF17) gene. The c.321+1G>A splice-site mutation is an extremely rare variant with limited reports in the literature. Identification of such ultra-rare variants contributes to strengthening genotype-phenotype correlations. We describe a 33-year-old Kashmiri woman with classical features of WSS, including hypergonadotropic hypogonadism, diffuse alopecia, diabetes mellitus, characteristic facies, and intellectual disability. Whole-exome

Early Dissecting Cellulitis of the Scalp Following Repeated Microinfusion of Medication Into the Skin in a Patient With Androgenetic Alopecia and Long-Term Exogenous Testosterone Use: A Case Report.

Dissecting cellulitis of the scalp is an uncommon chronic inflammatory disorder within the follicular occlusion spectrum and may progress to permanent cicatricial alopecia if not recognized early. We report a case of a 34-year-old male with androgenetic alopecia, two years of exogenous testosterone use, and a recent history of six-monthly sessions of scalp microinfusion of medication into the skin, who developed painful and pruritic inflammatory scalp nodules approximately 45 days after the last procedure. The procedure was performed using a Cheyenne Unlimited 4 dermograph fitted with a 2710MG

Do Alopecia Areata and Hair Colour Have a Shared Genetic Component?

Alopecia areata (AA) is a common T-cell mediated autoimmune disorder, which leads to sudden, non-scarring hair loss. An association between hair pigmentation and AA pathogenesis has long been postulated. Recent epidemiological evidence supports this link, indicating that individuals with darker hair colours have a higher risk of AA, while those with lighter hair colours exhibit a lower risk. This study aimed to investigate whether a shared genetic basis between hair colour determination and AA risk underlies this observation. To explore this, we analysed data from our AA genome-wide associatio

Real-World Systemic Treatment Patterns and Outcomes of Children With Severe Alopecia Areata: A French Nationwide Study (ESTAPED Cohort).

BACKGROUND/OBJECTIVE: Severe forms of alopecia areata (AA) often require off-label systemic treatments (ST) for which data on effectiveness and safety are limited. This study aimed to provide a comprehensive real-world overview of ST patterns in children with severe AA. METHOD: We conducted a retrospective, longitudinal, multicenter study of children with AA (aged <18 years) receiving at least one ST (2010-2023), identified using a keyword search in Ouest Data Hub Warehouse (a hospital data network in France with a critical mass of 5.1 million patients) or by members of the Société Française

Microneedle technology integrated with diverse therapeutic modalities for hair regrowth in alopecia.

Alopecia profoundly impacts an individual's appearance, quality of life, and social well-being, with its prevalence increasing with age. Conventional treatments, such as topical minoxidil and oral finasteride, suffer from limitations like inefficient drug delivery, side effects, and inconsistent efficacy. Other therapies, like hair transplantation, biologics and low-level laser therapy (LLLT), also face some constraints in practical application. Microneedles (MNs), as an emerging transdermal drug delivery system (TDDS), enable efficient local delivery of therapeutic agents to hair follicles in

JAK-STAT pathway-associated skin diseases: a refined functional framework for inflammatory skin diseases.

The Janus kinase (JAK)/signal transducer and activator of transcription (STAT) signaling pathway is involved in the pathogenesis of a variety of inflammatory and autoimmune skin diseases, such as atopic dermatitis (AD), psoriasis, alopecia areata (AA), vitiligo, etc. Many cytokines are involved in the occurrence and development of these diseases through the JAK-STAT-mediated intracellular signaling. Small molecule JAK inhibitors (JAKi) have demonstrated substantial efficacy in the treatment of many diseases, especially in inflammatory skin diseases. For example, abrocitinib and upadacitinib ha

Localized follicular delivery of a JAK inhibitor using optimized PhosGlyNanoVes: a novel approach for alopecia areata treatment.

Localized functional therapy with fewer side effects for alopecia areata (AA) is the major leading problem in the current scenario. A Janus kinase inhibitor, Tofacitinib citrate (TFC), can be explored for treating AA directly at the site of action to avoid systemic toxicity. The current study was designed to formulate and optimize the TFC phospholipon nanovesicles enriched with glycerol (PhosGlyNanoVes) and incorporate it into a gel (GlyNanoGel) with a view to improving its topical delivery in the treatment of AA. PhosGlyNanoVes were prepared using the thin lipid film hydration technique, with

Delays in Diagnosis of Alopecia Areata.

INTRODUCTION: Diagnostic delays in alopecia areata (AA) are often preventable and may be compounded by factors that have not been fully elucidated. Our objective was to identify potential associations and to characterize diagnostic delay of AA that extends beyond 1 year. METHODS: Patients with AA were recruited via online survey. A multivariable linear regression was performed to analyze diagnostic delay greater than 1 year while controlling for age, sex at birth, race, income, health insurance, and employment. A sensitivity analysis was performed to compare delays of over 9 months versus 1 y

The Concurrence of Atopic Dermatitis and Alopecia Areata: How Common Is It? Why Does It Happen? How Is This Managed Optimally in Clinical Practice?

Atopic dermatitis (AD) and alopecia areata (AA) are chronic immune-mediated disorders that frequently coexist in the same patient, an observation noted in both pediatric and adult populations. The clinical implications of a patient experiencing both disease states simultaneously underscores the need for a deeper understanding of pathophysiology of both disease states, how to optimally utilize specific therapeutic options to achieve synergistic outcomes, suggest simplified rational approaches to treatment selection, how to integrate care with other specialists when needed, and hopefully reach t

Assessing Alopecia Areata Misinformation: Social Media Analysis.

BACKGROUND: Alopecia areata (AA) is an autoimmune, hair loss-inducing disease affecting individuals of all genders and ethnic backgrounds worldwide. As social media platforms and artificial intelligence chatbots increasingly influence patient behavior, individuals with AA may turn to these sources for treatment guidance. OBJECTIVE: This study evaluated the accuracy of AA-related information on social media and assessed whether patients were exposed to misinformation that could hinder appropriate care. Influencers may recommend treatments applicable to alopecia broadly or promote therapies lac
