Integrating Psychological Support and Topical Therapy for the Effective Management of Stress-Induced Alopecia Areata: A Case Report.

Alopecia areata is an autoimmune disorder characterized by hair loss in defined patches. Genetic factors are considered primary contributors to the onset and exacerbation of certain conditions. However, environmental triggers, particularly psychological stress, also play a significant role. This case report presents a 28-year-old woman with no prior history of autoimmune or immune-related conditions who developed severe alopecia areata following prolonged emotional stress. Despite the absence of systemic or nutritional deficiencies, she experienced significant hair loss, which improved with 5%

IL-1 signaling enrichment in inflammatory skin disease loci with higher-risk allele frequencies in African ancestry.

Inflammatory skin diseases (ISDs) exhibit varying prevalence across different ancestry background and geographical regions. Genetic research for complex ISDs has predominantly centered on European Ancestry (EurA) populations and genetic effects on immune cell responses but generally failed to consider contributions from other cell types in skin. Here, we utilized 273 genetic signals from seven different ISDs: acne, alopecia areata (AA), atopic dermatitis (AD), psoriasis, systemic lupus erythematosus (SLE), systemic sclerosis (SSc), and vitiligo, to demonstrate enriched IL1 signaling in keratin

Exploring the shared genetic mechanisms of atopic dermatitis and alopecia areata via bioinformatics approaches.

Atopic dermatitis (AD) and alopecia areata (AA) are chronic inflammatory skin diseases. While studies suggest a possible immune defense mechanism link between the two, the causal relationship remains unclear, and current methodologies have limitations. This study aims to investigate the causal relationship between AD and AA using genome-wide association study statistics from the Integrative Epidemiology Unit Open Genome-Wide Association Study (IEU Open Gwas) project. We utilized the inverse variance weighting (IVW) method as our primary analysis approach for assessing the causal association be

Photodynamic therapy: Clinical applications in dermatology.

Photodynamic therapy (PDT) is a noninvasive treatment modality that uses photosensitizers in conjunction with a light source to induce therapeutic effects mediated in part through reactive oxygen species. PDT can be used as monotherapy or in combination with other treatments. In the United States, PDT is approved by the Food and Drug Administration for the treatment of actinic keratoses and is utilized off-label for other dermatologic conditions. Studies show that PDT has a favorable safety profile and, in order of level of evidence, has been used to treat actinic keratoses, basal cell carcino

Alopecia Areata: Understanding the Pathophysiology and Advancements in Treatment Modalities.

Alopecia areata (AA) is an autoimmune condition that presents with non-scarring hair loss affecting multiple patients worldwide during their lifetime. It ranges from well-defined patchy to diffuse total hair loss, impacting all hair-bearing areas of the body. AA most commonly predominantly manifests on the scalp. The pathophysiology of AA is complex and multi-faceted. The findings of our review article were consistent with the recent literature, delineating autoimmunity, genetic susceptibility, and environmental aspects to be the contributing factors. One of the main causes of AA is believed t

Pathophysiology of Alopecia Areata in the Pediatric Patient.

Alopecia areata (AA) is an autoimmune non-scarring hair loss that arises in genetically susceptible individuals, potentially in combination with environmental triggers or inciting events, of which the exact mechanism is not yet fully understood. Genome wide association studies have demonstrated an association between AA and variants in HLA haplotypes on chromosome 6 which correlate with other autoimmune conditions as well as other gene variants. Familial and twin studies also confer additional evidence to a genetic component. AA pathogenesis relies on immune privilege collapse at the hair foll

Characteristics and predictive values of super-responders in alopecia areata under tofacitinib treatment: a single-centre retrospective study.

BACKGROUND: Alopecia areata (AA) is an autoimmune disorder characterized by nonscarring hair loss, significantly impacting patients' quality of life. Rapid hair regrowth is the primary goal of treatment, particularly for super-responders (SRs), who demonstrate remarkable and swift improvement. The advent of Janus kinase (JAK) inhibitor treatment has transformed AA management, yet predictors of SRs in AA remain underexplored. OBJECTIVES: This study aimed to characterize SRs among patients with AA treated with tofacitinib, identify clinical and demographic predictors of SRs, and analyse treatme

Key considerations of injectable minoxidil for alopecia.

Minoxidil has become an increasingly popular treatment option for hair loss disorders including androgenetic alopecia, cicatricial alopecia, and alopecia areata. There are various minoxidil formulations available including topical, oral, and injectable minoxidil. While injectable minoxidil may offer certain advantages, potential risks and costs should also be considered. First, we explore the enhanced bioavailability of injectable minoxidil in comparison to topical and oral formulations, as well as the variability in sulfotransferase enzyme levels and its role in the activation of minoxidil at

Investigating the Genetic Association of Selected Candidate Loci with Alopecia Areata Susceptibility in Jordanian Patients.

Background and Objectives: Alopecia areata (AA) is a common cell-mediated autoimmune disease of the hair follicle that results in hair loss patches, affecting males and females of all ages and ethnicities. Although its etiology is not fully understood, AA is hypothesized to have a multifactorial basis with a strong genetic association. This study aims to replicate the genetic association of several risk loci in the Jordanian population for the first time. Materials and Methods: Genomic DNA samples of 152 patients with AA and 150 control individuals were extracted from EDTA blood tubes collecte

Potential role of leptin in Janus kinase 2 inhibitor-associated weight gain: a monocentric retrospective study.

Janus kinase (JAK) inhibitors are widely used for treating atopic dermatitis (AD) and alopecia areata (AA). Weight gain is an emergent adverse event associated with JAK inhibitors, particularly those acting on the JAK2 pathway. We conducted a retrospective monocentric two-cohort study, including 226 patients, to evaluate the prevalence of weight gain in patients with AD and AA who were treated with JAK inhibitors for ≥ 1 year. We included 142 patients in the JAK1/2 cohort (receiving upadacitinib 30 mg or baricitinib 4 mg) and 84 in the JAK1 cohort (receiving upadacitinib 15 mg or abrocitinib 1

Iron Chelators in the Management of Autoimmune-Induced Alopecia: A Focus on Hypoxia-Inducible Factor 1 Modulation and Hair Restoration.

Autoimmune-induced alopecia, such as alopecia areata, involves immune-mediated damage to hair follicles, leading to significant hair loss. Emerging therapies that stabilize hypoxia-inducible factor 1-alpha (HIF-1α) show promise in counteracting follicular degradation and supporting hair regrowth. This communication highlights the potential of iron chelators, specifically deferoxamine (DFO) and deferiprone (DFP), to stabilize HIF-1α by reducing iron availability, thereby promoting vascularization, cellular proliferation, and a regenerative environment in the hair follicle niche. Clinical trials

Oral and Topical Janus Kinase Inhibitors in Patients With Cutaneous T-Cell Lymphoma: A Real-World Single-Center Experience.

The JAK/STAT pathway is implicated in the pathogenesis of cutaneous T-cell lymphoma (CTCL), with early studies suggesting that JAK inhibitors (JAKi) may be used for treating CTCL. Patients with CTCL may have other indications for JAKi use, but data on JAKi agents in CTCL is limited due to warnings against their use in malignancy. We retrospectively characterized the experience of our tertiary cancer center with JAKi in patients seen for CTCL between the years 2011 and 2024. We identified 26 CTCL patients who received a JAKi: 7 received a JAKi prior to their CTCL diagnosis (topical (n = 5) or o

Psychosocial burden and the impact of illness perceptions and stigma on quality of life, anxiety and depression in alopecia areata: results from the Alopecia + Me study.

BACKGROUND: Alopecia areata (AA) can significantly impact patients' quality of life (QoL) and mental health, leading to increased levels of anxiety and depression. It is unclear whether this impact is more strongly associated with disease severity or patients' disease perception, and which patients are more likely to have a greater psychological burden. OBJECTIVES: To examine the psychosocial impact of AA, while focusing on illness perceptions and stigma, aiming to identify high-risk subgroups and key perceptions linked to worse QoL, anxiety and depression. METHODS: This was a UK cross-secti
