BACKGROUND: Alopecia areata, a common autoimmune disease, is not fully understood in terms of its cause. However, research suggests that an imbalance in specific blood metabolites may trigger immune system dysfunction, leading to an attack on hair follicles and ultimately resulting in alopecia areata.
METHODS: Two-sample MR analysis was conducted to investigate the causal relationship between plasma metabolites and alopecia areata using various methods. Heterogeneity and pleiotropy were assessed, robustness of findings evaluated, and reverse MR performed for effect analysis.
RESULTS: The MR

Atopic dermatitis (AD) and alopecia areata (AA) represent chronic inflammatory diseases characterized by heterogeneous immune-mediated mechanisms, including subtypes that may interconnect the two diseases, as well as other comorbidities. AD is globally recognized as the most common inflammatory skin disease and AA is an autoimmune disease, causing non-scarring hair loss. In both diseases the quality of life (QoL) is decreased, out-of-pocket expenses on alternative therapies and camouflage endeavours is high, increased productivity loss/absenteeism at work or school, and high healthcare costs a

INTRODUCTION: Alopecia areata (AA) is an autoimmune disorder, with potential roles of both genetic and environmental factors. Although AA is a specific autoimmune disease targeting hair follicles, its frequent association with other autoimmune diseases supports the notion that the autoinflammation is not confined to the hair follicles but is systemic. Systemic immune-inflammation index (SII), systemic inflammation response index (SIRI), and CRP (C-reactive protein) albumin ratio (CAR) are elevated in various diseases and correlate with disease severity.
OBJECTIVES: The aim of this study was t

INTRODUCTION: Severity of alopecia areata (AA) has been commonly defined by clinician assessment of scalp hair loss. Our aim was to characterize patient-reported factors beyond scalp hair loss that contribute to AA severity while modifying for socioeconomic factors.
METHODS: Patients with AA were recruited to take a survey. Multivariable linear regression was performed to evaluate lifetime AA severity while adjusting for socioeconomic variables, current AA severity at the time of survey, and patient-reported disease burden.
RESULTS: Patient-reported AA severity was associated with scalp hair

Because aberrations of protein kinase activity play causal roles in several human diseases, this family of enzymes is one of the most important drug targets of the 21st century. Of the 88 protein kinase antagonists that are approved by the FDA, eleven of them form irreversible covalent complexes with their target enzymes. The clinical efficacy of ibrutinib, a Bruton tyrosine kinase blocker, in the treatment of mantle cell lymphoma following its 2013 approval helped to overcome a general bias against the development of irreversible drug inhibitors. Other approved targeted covalent inhibitors in

BACKGROUND: The existence of a definite direct causal relationship between vitiligo and diverse autoimmune disorders remains unknown due to the influence of confounding factors and potential reverse causality.
METHODS: Mendelian randomization (MR) is a technique employed to explore causal connections between two phenotypes. In our research, bidirectional two-sample MR analyses were utilized to evaluate the causal connections between vitiligo and multiple autoimmune diseases (systemic lupus erythematosus, Graves' disease, inflammatory bowel disease, alopecia areata [AA], type 1 diabetes mellit

Alopecia areata (AA) is a chronic disease that significantly impacts well-being and quality of life. Despite its burden, professional support and treatment utilization remain low. This study investigated the experiences of 226 individuals in Finland with AA through an online survey. Most (82.7%) had at least one comorbid condition, most commonly atopic dermatitis (45.1%) and allergic rhinitis (43.4%). Depression and anxiety were reported by 22.1% and 23.0% of respondents, respectively. Those with ≥50% hair loss were significantly less likely to report higher overall well-being (odds ratio 0.60

Alopecia areata (AA) is a chronic inflammatory disease distinguished by non-scarring alopecia. The latest evidence suggests it is a T cell-mediated autoimmune disease that manifests in genetically susceptible individuals. In contrast to adult-onset AA, clinical traits and treatment of pediatric AA have been investigated very little. We hereby report a pediatric case of recalcitrant AA treated with a 308 nm excimer lamp and topical tofacitinib 2% gel.

Separable and dissolvable microneedle array patch based primarily on hyaluronic acid is designed for the topical delivery of baricitinib in alopecia areata (AA) therapy. Locoregional application of a separable microneedle array patch (S-MAP) may avoid the systemic side effects of oral administration and can be administered without pain compared with typical intradermal injections. Baricitinib (which has poor aqueous solubility) is evenly suspended with sodium carboxymethylcellulose. Subsequently, the mixture is blended with other pharmaceutical excipients, such as hyaluronic acid, sucrose, and

INTRODUCTION: People with alopecia areata (AA) experience stigma, but little is known about how this affects healthcare interaction (HCI). We aim to explore the relationship between stigma and HCI in AA.
METHODS: A cross-sectional survey of 500 members of the National Alopecia Areata Foundation was performed. Survey items related to stigma and HCI were summed and sorted into low, medium, and high tertiles. Multinomial logistic regressions were performed to control for demographic and clinical factors.
RESULTS: Respondents (n = 500) were white (80.0%), female (86.2%), and insured (96.0%), wit

Introduction Alopecia areata (AA) is an autoimmune disorder causing non-scarring hair loss on the scalp, eyebrows, eyelashes, and other areas. Janus kinase (JAK) inhibitors have emerged as promising treatments, but data on their efficacy in Middle Eastern populations, including Jordanians, are limited. The Severity of Alopecia Tool (SALT) score is commonly used to assess disease severity, while Clinician-Reported Outcome (ClinRO) measures provide additional insights. Aim To evaluate the efficacy of JAK inhibitors in Jordanian AA patients using the SALT score as the primary outcome measure. Me

BACKGROUND: While Janus kinase inhibitors (JAKi) show excellent outcomes in Phase2b/3 trials for alopecia areata (AA), they do have potential side effects due to their immunosuppressive nature and other effects on the JAK-STAT pathway. These side effects can be mitigated by adherence to lab monitoring.
OBJECTIVE: The authors sought to analyze patients, using JAKi, to manage symptoms of AA, to determine frequency of lab monitoring and check for adherence to the recommended rate of every three months.
METHODS: A retrospective chart review was conducted by first identifying all patients diagnos

BACKGROUND: Alopecia areata (AA) is an autoimmune disorder mediated by T cells, resulting in hair loss on the scalp, eyebrows, and body. Conventional treatments for AA often exhibit high recurrence rates and various side effects. Recently, Janus kinase (JAK) inhibitors have emerged as promising therapeutic options for managing AA and several other autoimmune disorders.
CASE PRESENTATION: We present the case of a 26-year-old patient who initially responded well to Tofacitinib but subsequently experienced relapses during the treatment period. Corticosteroid therapy was effective in managing the

INTRODUCTION: Therapeutic burden (TB) has been proposed as a potential predictor of treatment outcomes in both dermatological and non-dermatological diseases. This study aims to introduce the concept in the context of alopecia areata (AA) and assess its potential value in supporting therapeutic decision-making in clinical practice.
METHODS: A prospective cohort study was conducted including patients with AA who started treatment with baricitinib between January 2022 and January 2025 at a third-level hospital center. The main variable was TB, defined as the cumulative sum of previous systemic

Patients with, survivors of, and women at increased risk of breast cancer may experience various hair loss disorders, including those related to cancer treatments (such as chemotherapy- or endocrine therapy-induced alopecia), alopecia areata, androgenetic alopecia, and cicatricial alopecias. In the USA, approximately 1 in 8 women (13.1%) will develop breast cancer during their lifetime, emphasizing the importance of understanding safe treatment options for this population. Management of scarring and nonscarring alopecias in patients with or those at high risk of breast cancer requires the sele

INTRODUCTION: Diphenylcyclopropenone (DPCP) is an effective treatment in severe alopecia areata (AA). Platelet-rich plasma (PRP) has been introduced as an effective modality in AA. We present the first study to evaluate and compare the efficacy and tolerability of DPCP alone and DPCP in combination with PRP in the treatment of severe AA.
METHODS: This is a comparative, randomized, double-blind study that included 14 patients with severe AA. Participants received weekly DPCP plus monthly PRP to treat alopecia patches on one side of their scalp (group 1) and DPCP plus placebo on the other side
