First Case of H Syndrome in Northern Morocco: Clinical Features and Literature Overview.

H syndrome is a rare multisystem genodermatosis with an autosomal recessive inheritance, characterized by cutaneous hyperpigmentation, hypertrichosis, hepatosplenomegaly, hearing loss, heart anomalies, hypogonadism, short stature, hyperglycemia, hallux valgus and flexion contractures of fingers and toes. To date, 161 cases have been reported worldwide with limited data from North Africa. Herein, we report the case of a 17-year-old Moroccan girl diagnosed with H syndrome on the basis of clinical features and molecular testing. Clinical examination revealed dysmorphic features with insulin-depen

Plasma adrenocorticotropin concentrations vary with assay used in horses with and without pituitary pars intermedia dysfunction.

Plasma adrenocorticotropin (ACTH) concentration is measured to support a diagnosis of pituitary pars intermedia dysfunction (PPID). However, baseline ACTH and ACTH concentrations after IV administration of thyrotropin releasing hormone (TRH) vary with season and assay used. To compare two ACTH assays (Scantibodies immunoradiometric assay [IRMA] and Immulite 2000Xpi chemiluminescent immunometric assay [CIA]) for diagnosis of PPID, ACTH was measured before and 10 minutes after administration of TRH in three groups of horses: 1) four young horses (6 ± 3 years, no hypertrichosis and a pituitary gl

Porphyria cutanea tarda: cutaneous manifestations and management.

Porphyria cutanea tarda (PCT) is the most common porphyria and is caused by inhibition of hepatic uroporphyrinogen decarboxylase in the setting of iron overload and oxidative stress. Clinically, PCT presents with cutaneous photosensitivity, skin fragility, blistering and erosions on sun-exposed skin, hypertrichosis, dyspigmentation, milia, and dark urine. Because the skin findings may resemble other blistering disorders, including pseudoporphyria and other cutaneous porphyrias, recognition of the characteristic clinical pattern is important to prompt appropriate biochemical testing. Diagnosis

Gastrointestinal Side Effects of Low-Dose Oral Minoxidil in Patients with Alopecia: An Underrecognized Association.

INTRODUCTION: Low-dose oral minoxidil (LDOM) has become an increasingly used therapy for alopecia. While dose-dependent hypertrichosis is the most common side effect, gastrointestinal (GI) adverse effects are plausible due to smooth muscle relaxation, but they have not been systematically reported in clinical studies. OBJECTIVES: To investigate the frequency and characteristics of GI adverse effects in patients treated with LDOM for alopecia. METHODS: We conducted a prospective observational study at a trichology unit including patients attending their second consultation after initiating or

Tet2 deficiency alters CD4+ T cell function and promotes T cell lymphoma with a TFH cell immunophenotype.

TET2 mutations are frequent in TFH-derived lymphomas, but how epigenetic disruption initiates malignant T cell transformation is unclear. We generated Cd4cre;Tet2FL/FL mice, which developed aggressive T cell lymphoma (m-TCL) with a TFH cell-like immunophenotype. Genome-wide transcriptomics and epigenetic profiling of Tet2-/- CD4+ T cells prior to lymphoma development showed hyperactive TCR, PI3K signaling, and dysregulated TH differentiation program, with proliferation promoting signal transduction at the lymphoma stage. Tet2 loss promoted hyperplasticity under in vitro conditions but favored

Not quite naked: the bare necessities of human body hair evolution.

A striking characteristic that sets modern humans apart from their ancestors is the lack of a thick fur coat. Despite being popularly dubbed 'the naked ape', humans are covered in 2-3 million vellus hair follicles producing tiny hair fibres. This review explores why humans evolved thinner body hair, with a focus on the need for an effective cooling mechanism. We address anthropological evidence and traits that developed in parallel with humans losing their fur. We then discuss the differences between thick terminal hairs and thin vellus hairs, and the role of the dermal papilla in the regulati

The Voice of Cantú: Lower Voice Pitch Is a New Phenotypic Feature of Cantú Syndrome.

Cantú syndrome (CS) is a rare genetic condition caused by pathogenic variants in either ABCC9 or KCNJ8, leading to gain-of-function of KATP-channels. The main clinical features are hypertrichosis and cardiovascular abnormalities. This study investigates the voice characteristics in individuals with CS, an aspect that has received little attention in previous research. A total of 18 participants with molecular genetically confirmed CS were recruited, 11 females and 7 males, aged 9-63 years. Voice pitch analysis was conducted using the Voice Tools application, comparing these findings with norma

Chromosomal Rearrangements Identified in Three Additional Patients With Generalized Congenital Hypertrichosis With Gingival Hyperplasia Involving the 17q24.2-q24.3 Locus.

We describe three unrelated individuals with congenital generalized hypertrichosis with gingival hyperplasia (CGHGH), each carrying a distinct structural rearrangement (duplication, deletion, inversion) at 17q24.2-q24.3 identified by CMA and WGS. Despite differences in the type of rearrangement, all three patients seem to exhibit alterations affecting the genomic architecture of a cluster of genes, particularly involving the ABCA family (notably ABCA5, ABCA6, ABCA9, ABCA10), MAP2K6, and potassium channels (KCNJ16, KCNJ2). These findings suggest that disruption of the local chromatin organizati

Generalised Gingival Fibromatosis and Hypertrichosis: A Rare Case of Syndromic Presentation.

Gingival fibromatosis is a benign oral condition characterised by a slow progressive, non-haemorrhagic, non-painful, local or generalised enlargement of the gingival tissues. Gingival fibromatosis may have a genetic basis and is frequently associated with systemic or extraoral findings. Hypertrichosis is a clinical condition characterised by excessive hair growth without androgenic stimulation. The present case report describes a case of a 16-year-old girl who presented with severe generalised gingival fibromatosis along with hypertrichosis, which was treated by gingivectomy, and uneventful po

Expanding the therapeutic landscape of minoxidil for androgenetic alopecia: topical, oral and sublingual formulations.

Androgenetic alopecia (AGA) is the most common form of non-scarring alopecia, affecting up to 80% of men and 50% of women by the age of 70. Minoxidil, initially developed as an oral antihypertensive, has become one of the most widely used therapies for AGA due to its safety and efficacy. This review presents an overview of current evidence on topical, oral and sublingual minoxidil. Topical minoxidil, the only FDA-approved treatment for AGA in both men and women, promotes hair growth through several proposed mechanisms described in the article. Randomised controlled trials show that 5% formulat

Neuronal Heterotopy in a Patient with Wiedemann-Steiner Syndrome Caused by a Truncating KMT2A Variant: Clinical and Genetic Correlations.

Background and clinical significance: Wiedemann-Steiner syndrome (WSS) is a rare autosomal dominant neurodevelopmental disorder caused by heterozygous pathogenic variants in the KMT2A gene, which encodes a histone lysine methyltransferase essential for chromatin regulation. Affected individuals commonly present with developmental delay, intellectual disability, behavioral disturbances, short stature, characteristic facial features, and hypertrichosis, along with variable additional congenital anomalies. Emerging genotype-phenotype correlations suggest two functional classes of KMT2A variants:

Smoldering multiple myeloma in the United States: a population-based analysis.

INTRODUCTION: Primary cicatricial alopecias (PCAs) cause permanent hair loss and psychosocial distress. Management involves anti-inflammatory and hair growth-promoting therapies. A combination of tacrolimus, clobetasol, and minoxidil (TCM) may offer synergistic benefits by targeting distinct inflammatory pathways while increasing background hair density for improved camouflage. This study evaluates the safety and efficacy of TCM use in PCA patients. METHODS: We conducted a retrospective chart review of PCA patients aged 18+ years treated with TCM between January 1, 2009, and May 31, 2025. Out

Quality of life and patient-reported side effects of low-dose oral minoxidil in treating female pattern hair loss.

Female pattern hair loss (FPHL) significantly affects women's well-being and quality of life (QoL), and low-dose oral minoxidil (LDOM) has increasingly gained attention as a convenient and effective treatment option. Although hypertrichosis is reported more often with LDOM than with topical formulations, patient perceptions and tolerance of this side effect remain poorly understood. We conducted a telephone survey at a single institution among women with FPHL currently taking or previously treated with LDOM for at least one month to evaluate treatment effectiveness, QoL, and side effect burden
