The Dosage Effect of Manganese Superoxide Dismutase (MnSOD Val16Ala) Polymorphism in Acne Vulgaris and In Silico Identification Analysis of Biological Features of Variation.

The exact cause of acne vulgaris remains unclear. However, evidence suggests polymorphism in the manganese superoxide dismutase (MnSOD) gene increases acne vulgaris risk by altering the protein's secondary structure. This study aimed to perform in silico and in vitro analyses of the MnSOD Val16Ala substitution, involving 119 acne vulgaris patients and 96 controls. Bioinformatic tools, including SIFT, PolyPhen, SNPs&GO, Panther, PhD-SNP, SNAP2, I-mutant, and I-TASSER, were utilized for insilico analyses. Genotypes and allele frequencies were determined by real-time PCR. Odds ratios (OR) and 95%

A co-amorphous salt of ursodeoxycholic acid and matrine incorporated into dissolving microneedles for combined antibacterial and anti-inflammatory treatment of acne vulgaris.

Acne vulgaris is characterized by microbial dysbiosis and excessive inflammatory activation, whereas current topical therapies are often limited by poor skin penetration and increasing antibiotic resistance. Ursodeoxycholic acid (UDCA) has anti-inflammatory potential but suffers from low aqueous solubility and limited cutaneous permeability. In this study, a co-amorphous salt of UDCA and matrine (MAT) was developed to improve the pharmaceutical performance of UDCA and to combine the complementary activities of the two components. UDCA-MAT was further incorporated into hyaluronic acid-based dis

Dysbiosis in Acne Vulgaris and Hidradenitis Suppurativa: A Comparative Microbiome Analysis.

Acne and hidradenitis suppurativa (HS) are inflammatory disorders of the pilosebaceous unit that exhibit distinct clinical manifestations, indicating that they likely differ in their underlying pathophysiology. Microbial dysbiosis is implicated in both diseases, yet direct comparisons using unified methods and analyses incorporating the oral microbiome are lacking. In this study, we collected lesional and nonlesional skin, buccal mucosa and faecal samples from 28 HS patients, 29 acne patients and 40 healthy controls, and profiled microbial communities using 16S rRNA V3-V4 sequencing with qPCR

Growth-Inhibitory Activity of a Clerodane Diterpene from Callicarpa americana Leaf Extracts and Its Derivatives against Cutibacterium acnes.

Cutibacterium acnes is a Gram-positive bacterium implicated in acne vulgaris, a common inflammatory skin disease. Rising antibiotic resistance in C. acnes highlights the need for new therapeutic agents. Callicarpa americana L., a shrub native to the southeastern United States, has a history of use in Native American traditional medicine. Bioassay-guided fractionation of C. americana leaf extracts led to the isolation of 12(S),16ξ-dihydroxycleroda-3,13-dien-15,16-olide (compound 1), a clerodane diterpene. Compound 1 exhibited growth-inhibitory activity against C. acnes (MIC: 8 μg/mL). A route t

Removal Mechanism of Cutibacterium acnes Biofilms: A Comparison between Synthetic Surfactants and Rhamnolipid Biosurfactants.

Biofilms are composed of autogenously secreted extracellular polymeric substances (EPS) and affect nearly all aspects of human activity. Biofilms with negative health and economic effects can be controlled through the integrated use of surfactants and mechanical force. However, the relationships between the surfactant molecular structures and biofilm removal pathways have not been sufficiently elucidated. Additionally, Cutibacterium acnes (C. acnes), a skin commensal bacterium with biofilm-forming capabilities that can exacerbate acne vulgaris, has been rarely investigated. In this study, we e

Green-synthesized metal oxide nanoparticles enhance the antibacterial activity of Portulaca oleracea through foliar nano-elicitation.

Acne-associated and other skin-related bacterial infections are increasingly difficult to manage because of antimicrobial resistance and the need for safer therapeutic alternatives. Portulaca oleracea is a medicinal plant with recognized bioactive potential, yet its antibacterial activity can potentially be enhanced through nano-elicitation. Unlike studies that examine nanoparticles as direct antimicrobials, this work investigated green-synthesized metal oxide nanoparticles as foliar elicitors to improve the antibacterial potential of P. oleracea extracts. This approach links sustainable nanop

Traditional Chinese medicine in acne treatment: From classical formulas to bioactive phytoconstituents and mechanisms.

ETHNOPHARMACOLOGICAL RELEVANCE: Acne vulgaris is a multifactorial inflammatory skin condition that affects over 80% of adolescents worldwide. Conventional monotherapy often fails to address sebaceous hyperplasia, Cutibacterium acnes dysbiosis and epidermal barrier dysfunction concurrently. Traditional Chinese herbal medicine (CHM) has long been used to treat acne patterns such as 'damp-heat' or 'blood-stasis', but the mechanisms underlying its effectiveness require further substantiation within a modern translational framework. AIM OF THE REVIEW: To systematically evaluate the efficacy and sa

The shared genetic architecture between acne vulgaris and inflammatory bowel disease: A cross-trait analysis.

Acne vulgaris (AV) is clinically associated with inflammatory bowel disease, yet the biological mechanisms driving this association remain unclear. We investigated the shared genetic architecture of AV and inflammatory bowel disease (including ulcerative colitis and Crohn's disease) using data from GWASs. Although AV was genetically weakly correlated with inflammatory bowel disease, MiXeR analyses estimated that 28.2% of the causal variants for inflammatory bowel disease are also causal for AV, with distinct overlap for ulcerative colitis and Crohn's disease. Using the pleiotropic analysis und

Chemical Characteristics and Antimicrobial Activity of Arctostaphylos uva-ursi (L.) Spreng. Extracts Against Skin-Associated Bacteria.

The interest in the use of phytochemicals and herbal medicines for the treatment of acne vulgaris has grown steadily over recent decades. The research on the secondary metabolites and biological properties of bearberry (Arctostaphylos uva-ursi (L.) Spreng.) has been intensified in recent years, but the range of bacterial strains tested, many of which are highly relevant to human health, remains very limited. Therefore, the aim of this study was to evaluate the chemical composition and the antioxidant, antimicrobial, and cytotoxic activities of water and ethanolic bearberry leaf extracts. Compa

AcneFormer: A Lesion-Aware and Noise-Robust CNN-Transformer for Acne Image Classification.

Convolutional neural networks (CNNs) have been widely used for acne image classification due to their effectiveness in capturing local texture of skin lesions. However, the locality of convolution operations limits their ability to model long-range dependencies. Vision Transformer (ViT) methods address this issue to some extent but their high computational complexity and reliance on large-scale pre-training present challenges. Although CNN-Transformer architecture alleviates this conflict to some extent, acne images present task-specific challenges, including indistinct lesion boundaries, subt

Malassezia Folliculitis Presentation, Diagnosis, and Treatment: A Review of "Fungal Acne".

BACKGROUND: Malassezia folliculitis (MF), commonly referred to as "fungal acne" is a dermatological condition caused by colonization of the commensal fungus Malassezia, which turns pathogenic under certain conditions. MF presents as pruritic, erythematous papules and pustules and is often mistaken for other dermatological conditions, including acne vulgaris (AV). Interest in MF has intensified in recent years, likely due to social media trends, leading to concern regarding patients inappropriately applying antifungals without a confirmed diagnosis. METHODS: This narrative review summarizes th
