Comparative Profile of Microbiome in Normal Skin and Acne Vulgaris Skin Patients.

INTRODUCTION: Changes in the skin microbiome are associated with acne vulgaris (AV), a condition characterized by comedones, papules, and pustules. While some bacteria have been studied, many others remain unexplored, highlighting the need to understand the microbiome differences between acne-prone and normal skin. This study aimed to compare skin microbiome profiles between AV patients and healthy individuals and to explore associations with microbial diversity and specific bacterial populations involved in AV pathogenesis. METHODS: A total of 144 participants were recruited, comprising 36 A

Antimicrobial photodynamic therapy using sodium iron chlorophyllin against drug-resistant Cutibacterium acnes from patients with acne vulgaris.

Antimicrobial resistance in Cutibacterium acnes is a growing concern, limiting treatment options for acne vulgaris (AV) and increasing the risk of opportunistic infections. Photodynamic therapy (PDT), using an effective photosensitizer (PS) and optimized light parameters, is a potent broad-spectrum antimicrobial strategy with minimal risk of resistance development. This study aimed to assess the efficacy of PDT using sodium iron chlorophyllin (CHL-Fe) as an in-house photosensitizer (CH-PDT) against 10 clinically isolated, drug-resistant C. acnes strains. The isolates, obtained from patients wi

Anti-Acne Potential of Quercus salicina Extract: Inhibition of Cutibacterium acnes Growth and Virulence.

Acne vulgaris is a prevalent inflammatory skin disorder mediated by Cutibacterium acnes, a key etiological factor. In this study, the anti-acne properties of the ethyl acetate fraction of Quercus salicina Blume (QsB-EE) against C. acnes were investigated. The QsB-EE fraction was separated into subfractions (E1-E6) using preparative HPLC. Antimicrobial and anti-virulence activities were evaluated via bacterial growth, biofilm, and lipase activity assays. Virulence gene expression was assessed using qRT-PCR, and bioactive compounds were identified by LC-MS/MS. The QsB-EE demonstrated significant

Near-infrared light-driven nanomotors-based microneedles for the active therapy of bacterial infected acne.

Acne, caused by Cutibacterium acnes, triggers inflammatory lesions. The hypoxic biofilm microenvironment exacerbates acne progression, while inadequate hydrogen peroxide and dense biofilm barriers hinder oxygen generation and nanomedicine penetration. Here, we develop microneedles patch loaded with near infrared-driven self-oxygenating Z@P-M nanomotors for photothermal therapy of acne. Zinc peroxide nanoparticles are asymmetrically modified with polydopamine, followed by in-situ manganese dioxide growth on polydopamine to form Z@P-M nanomotors. Z@P-M nanomotors loaded microneedles penetrate th

Non-optimal apparent temperatures can increase the risk of acne: a multi-center time-series study.

As extreme temperature events increase globally, the influences of non-optimal temperature on health outcomes are of increasing concern. However, whether there is a correlation between non-optimal apparent temperature (AT) and acne is unknown. To illustrate the association between non-optimal AT and acne, data of daily acne outpatient visits, meteorological factors, and air pollutants from 2015 to 2019 in Chongqing, China were obtained. A multi-center study using distributed lag non-linear model (DLNM) was employed to explore the possible association between short-term non-optimal AT and outpa

Substrate Specificity and Immunological Implications of Cutibacterium acnes Phage Endolysins.

Cutibacterium acnes resistance to antibiotics poses a significant challenge in treating acne vulgaris. Bacteriophages offer a promising alternative to overcome this challenge given their specificity. In the current study, 15 bacteriophages were isolated from acne affected volunteers and subjected to whole genome sequencing to characterize their genetic features, diversity and endolysins encoding genes for further structural and functional analysis. Five representative endolysins (CAP 1-1, 6-3, 7-1, 10-3, and 12-3) were chosen for structural and functional analysis after average nucleotide iden

Transparent, 3D-printed microneedle patches that co-deliver ALA and guide blue light for preclinical photodynamic therapy of acne.

Acne is a common chronic inflammatory skin disease associated with Cutibacterium acnes (C. acnes). Although photodynamic therapy (PDT) effectively improves acne, the transdermal delivery of photosensitizers and limited light penetration through the skin restrict its therapeutic efficacy. In this study, we developed a dual-functional flexible microneedle patch using 3D printing technology, capable of simultaneously delivering the photosensitizer 5-aminolevulinic acid (ALA) and blue light. The microneedle patch exhibits favorable mechanical properties (a fracture force of 2.47 N per patch and a

Ionic liquids in dermatological drug delivery: advances, mechanisms, and therapeutic potential.

Current pharmaceutical and biological techniques have been greatly enhanced by the use of ionic liquids (ILs) in healthcare. ILs may be precisely tailored at the molecular level due to their exceptional structural tunability, which makes it possible to build task-specific solutions for a variety of medical difficulties. This adaptability has resulted in advances in targeted administration, drug solubilisation, and the removal of physiological barriers that have historically restricted the effectiveness of treatments. The effective treatment of dermatological conditions is still constrained by

Genetic exploration of the relationship between liability to psychiatric disorders and acne vulgaris.

Observational epidemiology suggests a link between the dermatological disorder acne vulgaris and several psychiatric disorders. However, the biological mechanisms that underlie the relationship between acne and mental health are poorly characterised. Here, we employed a genetic approach using large-scale genome-wide association studies of acne and ten psychiatric disorders to both estimate causal effects and uncover potential shared genetic risk factors. Multiple psychiatric disorders displayed evidence of small-to-moderate genetic correlations with acne. However, only genetic liability to sch
