Causal Association Between Ankylosing Spondylitis and Hidradenitis Suppurativa: A Twosample Bidirectional Mendelian Randomization Study.

BACKGROUND: Ankylosing spondylitis (AS) and hidradenitis suppurativa (HS) represent two chronic inflammatory diseases that share similar underlying immunological characteristics. Observational studies suggest a potential association, but the causal relationship remains underexplored. OBJECTIVE: To evaluate bidirectional causal links between AS and HS using Mendelian randomization (MR). METHODS: We implemented two-sample bidirectional MR analyses, utilizing summary statistics obtained from genome-wide association study (GWAS) datasets. Genetic instruments for AS (ncase=9069, ncontrol=1550) an

The Role of GLP1 Receptor Agonists and Multi-agonist Incretin Therapies for Specific Obesity-related Health Conditions: Evidence and Rationale for Prioritisation.

PURPOSE OF REVIEW: This review evaluates the strength and scope of clinical evidence for GLP-1RAs—liraglutide, semaglutide, and tirzepatide—across obesity and associated complications, providing a structured rationale for prioritisation in clinical practice and policy. RECENT FINDINGS: GLP-1RAs demonstrate significant and reproducible weight loss—approximately 15% with semaglutide and 20% with tirzepatide at one year. Their efficacy in obesity-related health conditions has been best demonstrated through large clinical trials in cardiovascular disease, type 2 diabetes, obstructive sleep apnoea

Management of Giant Keloids Using Full-Thickness Skin Grafts: A Case Series From a Low-Resource Region.

BACKGROUND: Management of giant keloids in the submandibular and cervical regions remains challenging due to high recurrence rates, functional impairment, and limited access to adjuvant therapies in low-resource settings. Chronic inflammation, mechanical tension, and adnexal involvement may contribute to aggressive keloid behavior. Optimal reconstructive strategies in such cases remain controversial, particularly when postoperative radiotherapy is unavailable. METHODS: The authors present a case series of three male patients with giant submandibular and anterior cervical keloids associated wi

Long-term Natural History and Elosulfase Alfa Treatment for Mucopolysaccharidosis Type IVA: A Single-Center Study in the Czech Republic.

BACKGROUND: Mucopolysaccharidosis type IVA (MPS IVA, Morquio A syndrome) is a rare lysosomal storage disease primarily characterized by severe skeletal dysplasia. Clinical and laboratory data, including treatment responses, are reported for a cohort of Czech patients with MPS IVA. RESULTS: Nineteen patients with MPS IVA from 17 families (13M/6F) were included; only one patient exhibited a mild form. The median age at disease onset was 3.0 ± 1.4 years, whereas the median age at confirmed diagnosis was 5.0 ± 3.5 years. At the time of diagnosis, all patients had skeletal symptoms (pectus carinat

Disease context dictates the cellular targets of IL-17 in inflammatory skin disease.

Therapeutic blockade of IL-17 and TNF can effectively treat inflammatory skin diseases such as hidradenitis suppurativa and psoriasis, yet the relative importance of the different cell types that respond to IL-17 and TNF remains unresolved. Keratinocytes are viewed as the dominant effector cells, whereas fibroblasts have recently emerged as important contributors. In mice, topical imiquimod induces IL-17- and TNF-dependent skin inflammation and is frequently used to model psoriasis. Here, we demonstrate that intradermal injection of recombinant IL-17 and TNF elicits skin inflammation with feat

Association between hidradenitis suppurativa and peripheral arterial occlusion disease: a propensity-score-matched cohort study.

BACKGROUND: There are plausible mechanistic associations between hidradenitis suppurativa (HS) and peripheral arterial occlusive disease (PAOD), which has significant clinical implications. OBJECTIVES: This study investigated the risk of incident PAOD in patients with HS. METHODS: A retrospective cohort study using the TriNetX research network was conducted. Patients with HS were matched 1 : 1 with an HS-free control group based on demographic and clinical factors. The primary outcome was the incidence of PAOD, analysed using hazard ratios (HRs) and Kaplan-Meier curves over a 15-year follow-
