A one compartment model to assess skin lightening rate of cosmetic formulae in UV induced clinical studies.

Ultraviolet-induced skin darkening is often used as a clinical model to assess the efficacy of active ingredients aimed at modulating skin pigmentation ; targeting pigmentary skin disorders. Studying Ultraviolet (UV)-induced skin pigmentation therefore presents challenges due to the complex pigmentation kinetics involved. This work aimed to develop a population pharmacokinetic (popPK) model to describe the UV pigmentation kinetics, compare products efficacy with their corresponding vehicle in terms of elimination, i.e., the efficacy to decrease pigmentation, and conduct a meta-analysis on the

Neuropathic Pain and Enlarged Nerves in Adult Noonan Syndrome and Noonan Syndrome With Multiple Lentigines: Health-Related Quality of Life and Neurologic Symptoms.

Noonan syndrome (NS) and the clinically related Noonan syndrome with multiple lentigines (NSML) belong to the group of RASopathies. Although pain is not mentioned as a characteristic feature, it has recently been reported as a clinically significant problem. This pain is likely multifactorial in origin, with a significant contribution from neuropathic mechanisms. Patients with NS also have a high chance of having multifocally enlarged nerves, and sometimes show clinical features of neuropathy. The relationship between these nerve findings, pain, health-related quality of life and neurological

Cockayne syndrome mutation in XPG activate the integrated stress response.

Xeroderma pigmentosum (XP) and Cockayne syndrome (CS) are diseases provoked by mutations in multifunctional proteins that are involved in DNA repair. DNA-repair deficiency explains the high cancer incidence of XP, whereas cancer-free CS, characterized by growth retardation, neurological degeneration, and premature aging does not present as a classical DNA-repair deficiency disorder. Here, we compared a severe combined XP/CS case provoked by XPG-mutation with an XP "only" patient cell line caused by mutation in the same XPG gene to carve out the pathogenic cellular disturbances that provoke CS.

Nelumbo nucifera extract alleviates UVB-induced hyperpigmentation via NOX4-mediated AMPK-YAP-ATG5 signaling pathway.

BACKGROUND: Hyperpigmentation is primarily caused by ultraviolet (UV) radiation, which triggers oxidative stress and melanin accumulation in the skin. Moderate UVB exposure activates autophagy, which facilitates cell repair, while prolonged UVB exposure disrupts this process, worsening skin damage and pigmentation. Nelumbo nucifera extract (NnE) drew our attention in our preliminary screening experiments for its potential to regulate keratinocyte autophagy. Nevertheless, the molecular mechanisms underlying the extract of NnE's therapeutic effects and its bioactive constituents remain incomplet

Biological Roles of Melanin and Natural Product-Derived Approaches for Its Modulation.

Melanin produced in melanocytes contributes to photoprotection, oxidative stress reduction, immune regulation, and epidermal homeostasis, while its dysregulation underlies diverse pigmentary disorders. Natural products modulate melanogenesis by regulating tyrosinase activity, intracellular signaling pathways such as extracellular signal-regulated kinase/mitogen-activated protein kinase (ERK/MAPK) and cyclicAMP/protein kinase A/cAMP response element-binding protein (cAMP/PKA/CREB), and cellular redox balance. Anti-melanogenic effects have been reported for various fruit-derived phytochemicals,

Monopolar Radiofrequency for Facial Hyperpigmentation Treatment: An Integrated Retrospective Clinical Trial and Ex Vivo Study.

Aging-associated facial hyperpigmentation is driven not only by enhanced melanogenesis but also by dermal senescence and deterioration of the dermal-epidermal junction. The purpose of this study was to evaluate whether monopolar radiofrequency (MRF) monotherapy can improve aging-related facial hyperpigmentation by simultaneously suppressing melanogenic signaling and restoring senescence-associated dermal alterations. We assumed that deep dermal heating induced by MRF would modulate fibroblast senescence and basement membrane integrity, thereby indirectly regulating melanocyte activity. In a re

Potential Bioactive Function of Microbial Metabolites as Inhibitors of Tyrosinase: A Systematic Review.

Tyrosinase (EC 1.14.18.1) is a binuclear copper enzyme responsible for the rate-limiting steps of melanogenesis, catalyzing the hydroxylation of L-tyrosine and oxidation of L-DOPA into o-quinones that polymerize melanin. Beyond its physiological role in pigmentation, tyrosinase is also implicated in food browning and oxidative stress-related disorders, making it a key target in cosmetic, food, and biomedical industries. This systematic review, conducted following PRISMA guidelines, aimed to identify and analyze microbial metabolites with tyrosinase inhibitory potential as sustainable alternati

Solvent-free green preparation of niacinamide niosomes for targeting UVB-induced hyperpigmentation: QbD-based optimisation and in vivo assessment.

Hyperpigmentation is often driven by ultraviolet (UV)-induced inflammation and melanogenesis. This study aimed to develop a novel, eco-friendly niacinamide (NIA) nano-delivery system to overcome the skin barrier and effectively target both inflammation and pigmentation associated with ultraviolet B (UVB) damage. Employing a Quality-by-Design (QbD) approach, we utilised a D-optimal response surface design to fabricate and optimise solvent-free niacinamide niosomes (NIA-NIs). Particle size (PS) was minimised and zeta potential (ZP) maximised for enhanced stability and skin penetration using nove

Type I interferon signaling defines a novel disease signature in xeroderma pigmentosum C human keratinocytes.

Xeroderma Pigmentosum C (XPC) is a DNA damage recognition protein central to the global genome nucleotide excision repair (GG-NER) pathway, where it acts as a primary sensor of UV-induced DNA lesions. Loss-of-function mutations in the XPC gene lead to a photosensitive phenotype, with marked accumulation of unrepaired DNA damage and a dramatically elevated risk (10,000-fold) of skin cancer. However, understanding the molecular signaling mechanisms associated with XP-C has been hindered by the lack of reproducible disease models. Here, we overcome this challenge using our genetically engineered

Pigmentation disorders in three specimens of spiny dogfish, Squalus acanthias Linnaeus, 1758, from the Gulf of Venice, northern Adriatic Sea.

This study reports three cases of pigmentation disorders in Squalus acanthias from the Gulf of Venice. Two specimens exhibited spotless phenotypes, whereas one exhibited hypermelanism. Morphological assessments were conducted for two individuals, alongside genetic analysis of mitochondrial genes to determine phylogeographic relationships. Both specimens belonged to the South Pacific/Atlantic/Mediterranean haplogroup, with the first one associated with the main haplotype and the second one linked to a rarer haplotype. This study emphasizes the need for proper identification based on morphology

Xeroderma pigmentosum with multiple skin carcinoma and a homogenous XPC mutation: A case report from China and literature review.

Xeroderma pigmentosum is a rare autosomal recessive genetic disorder characterized by hypersensitivity to ultraviolet radiation and increased risk of skin cancer. Impaired DNA repair mechanisms are considered to be involved in the occurrence and development of this distinct disorder. We present the case of a 48-year-old Chinese woman with facial and chest tumors; these lesions had been rapidly growing over the past 6 months. Pathological biopsy and immunohistology indicated malignant melanoma in facial and chest tumors and squamous cell carcinoma in chest tumors. Using whole-exome sequencing,

Efficacy and Safety of Cysteamine-Isobionic Amide Complex in the Treatment of Periorbital Hyperpigmentation.

Periorbital hyperpigmentation is a common yet challenging cosmetic concern due to the thin, vascular, and pigment-prone nature of periocular skin, as well as its multifactorial etiology. Discoloration in this area can be driven by genetics, photoaging, dermal thinning, and pigmentary disorders, making treatment outcomes often unpredictable and temporary. The cysteamine–isobionic amide complex offers a novel, non-hydroquinone and non-retinoid approach, combining the multi-pathway pigment reduction of cysteamine with the barrier-supporting and pigment-evening properties of isobionic amide.
