Co-occurrence of retinoblastoma and oculocutaneous albinism in a Mexican patient: a case report.

INTRODUCTION: Retinoblastoma (RB) and oculocutaneous albinism (OCA) are genetically independent disorders whose coexistence is exceptionally rare, with an estimated theoretical frequency of 1 in 800 million births. METHODS: We report a 5-month-old Hispanic male with OCA who developed unilateral multifocal RB. Next-generation sequencing identified a de novo heterozygous pathogenic RB1 nonsense variant (c.1411C > T, p. Gln471*) and homozygous pathogenic OCA2 missense variant (c.2228C > T, p. Pro743Leu). RESULTS: The profound retinal hypopigmentation characteristic of OCA precluded standard pig

A homozygous variant in the GJB2 gene as a potential phenocopy of Waardenburg syndrome: case report.

Waardenburg syndrome (WS) is a neurocristopathy primarily characterized by congenital hearing loss and multisystem pigmentary abnormalities. We report a 12-year-old Han Chinese girl who presented with moderate-to-severe sensorineural hearing loss, a wide nasal root, diffuse hypopigmentation of the hair, and facial freckles-features partially similar to the WS phenotype. However, her calculated W-index was 1.87, below the diagnostic threshold for dystopia canthorum (>1.95), and her overall clinical presentation did not meet the diagnostic criteria for any WS subtype. Whole-exome sequencing (WES

The Safety of Melasma Treatment in Vitiligo Patients: An International E-Delphi Study.

Vitiligo and melasma frequently coexist, creating a treatment dilemma, since agents effective for one condition can destabilize the other, and no consensus currently exists on safe management. We conducted a scoping literature review (through June 2023) on treatment of patients with concomitant vitiligo and melasma, followed by a two-round international e-Delphi survey (22 experts from 14 countries; consensus defined as ≥ 70% agreement) and a final online consensus meeting. Only one study supported the safe use of oral tranexamic acid for melasma in vitiligo patients. Expert consensus favored

Prevalence and geographical distribution of persons with albinism (PWA) in Togo in 2024.

BACKGROUND: Persons with albinism (PWAs) in Africa represent a vulnerable population facing social discrimination, severe health issues such as skin cancers, and precarious socioeconomic conditions. The objective of this study was to estimate the prevalence of albinism and map the distribution of persons with albinism in Togo in 2024. METHOD: This cross-sectional descriptive study was conducted from November 2023 to October 2024 across all 39 health districts in Togo, and it included a telephone survey in which community health workers (CHWs) identified PWAs in their districts. CHWs were subs

Immune-mediated remodelling of tissue architecture in Post-Kala-Azar Dermal Leishmaniasis (PKDL).

BACKGROUND: Post-Kala-Azar Dermal Leishmaniasis (PKDL), a dermal sequel of apparently cured visceral leishmaniasis (VL), presents with macular or polymorphic lesions. A relationship, if any, between immune cell infiltration and cytokines potentially causing tissue-remodelling remains poorly defined, and was the focus of this study. METHODOLOGY/PRINCIPAL FINDINGS: Skin biopsies along with plasma from PKDL patients and healthy controls were analyzed. Tissue sections were assessed for immune cell subsets (CD8 ⁺ T cells, CD68 ⁺ macrophages, CD20 ⁺ B cells, CD66b⁺ neutrophils), associated cytokine

In Vitro Screening of Sargassum spp. Ethanolic Extracts for Antioxidant Activity and Inhibition of Disease-Associated Enzymatic Targets Related to Diabetes, Alzheimer's Disease, and Parkinson's Disease.

Sargassum is a large and diversified genus of marine brown algae that is distributed widely in tropical and subtropical areas. They have been used for centuries in Asian countries, and have demonstrated a variety of health advantages, including anti-cardiovascular disease, anti-osteoarthritis, anti-cancer, anti-inflammatory, antioxidant, and hypopigmentation effects. However, the putative anti-diabetic, anti-Alzheimer's, and anti-Parkinson's diseases activities of various Sargassum spp. have not been investigated and compared thoroughly. Thus, the purpose of this study was to examine the anti-

Spontaneous hypopigmentation mutant in threespine stickleback fish exhibits changes in the gut microbiome consistent with inflammatory bowel diseases.

Albinism is a large group of disorders characterized by a loss of pigment in ectoderm-derived tissues - most noticeably impacting the skin, hair, and eyes. Albinism is categorized as either syndromic or non-syndromic. Syndromic albinisms include a wide range of disorders with mutations impacting not just pigmentation but other cellular functions. These include, non-exclusively, melanogenesis disorders, endosome trafficking disorders like lysosomal storage disorders, and cell migration disorders. Mutations leading to syndromic albinisms can cause multi-organ system symptoms due to broad disrupt

Clinical significance of circulating regulatory T Cells, soluble CD25, and CXCL9 in assessing disease activity of vitiligo.

Vitiligo is an autoimmune skin disorder characterized by T-cell-mediated melanocyte destruction and loss of immune tolerance. Dysregulation of regulatory T cells and altered chemokine signaling have been implicated; however, their relationship with disease activity remains unclear. Therefore, this study aimed to evaluate peripheral immune cell subsets, particularly Tregs, sCD25, and CXCL9 biomarkers in patients with vitiligo and their relationship with disease activity. This study included 44 patients with non-segmental vitiligo and 44 matched healthy controls. Peripheral blood lymphocyte subs

Vitiligo and Its Mimics in Childhood: A Diagnostic Framework for Hypopigmented Disorders.

Hypopigmented disorders in childhood encompass a broad spectrum of presentations, including depigmentation, hypopigmentation, and pseudo-hypopigmentation arising from non-melanin mechanisms. In clinical practice, the primary challenge often lies in distinguishing these conditions from vitiligo, the most common acquired depigmentation disorder. Although vitiligo is categorized into nonsegmental and segmental subtypes, many children initially present with an undetermined or focal form before the clinical subtype is clearly established. A systematic diagnostic approach begins with Wood's lamp exa

Oxidative stress in refractory and recurrent vitiligo: from immune amplification mechanisms to potential stratified antioxidant combination therapy.

Vitiligo is an acquired depigmenting disorder characterized by destruction of epidermal melanocytes and skin depigmentation. Although treatments have developed substantially, there is still a considerable proportion of vitiligo patients fail to achieve satisfactory repigmentation or experience disease recurrence after treatment withdrawal. Vitiligo treatment failure involves heterogeneous clinical states, including persistent disease activity, treatment resistance, site-related limitations, and disease recurrence. Importantly, refractory and recurrent vitiligo are heterogeneous and partially o

UVB-Induced Repigmentation in Vitiligo: Hair Follicle Melanocyte Stem Cell Migration and Its Molecular Mechanisms.

Vitiligo is an acquired depigmentation disorder characterized by the loss of epidermal melanocytes and can markedly impair patients' quality of life. Because of its dual effects on immunosuppression and repigmentation, UVB phototherapy has become a first-line treatment for vitiligo. It is particularly valuable in inducing repigmentation of skin lesions. Since epidermal melanocytes are markedly reduced or even absent in vitiligo lesions, migration of melanocyte stem cells (McSCs) from the hair follicle bulge to the interfollicular epidermis is considered a critical step in UVB-induced repigment

MC1R variants in four Chinese children with red hair and hypopigmentation.

Loss-of-function variants in the melanocortin 1 receptor (MC1R) gene are a well-established cause of red hair in European populations. However, the genetic spectrum and phenotypic effects of MC1R variants in East Asians remain poorly defined, as red hair is exceedingly rare in this population. To characterize the clinical and genetic features of MC1R-associated red hair and hypopigmentation in Chinese pediatric patients and to expand the genotypic spectrum of MC1R in East Asian populations, four unrelated Chinese children with red or reddish-brown hair underwent comprehensive dermatologic and

Keratinocyte Autophagy in Inflammatory Dermatoses: Mechanisms, Disease-Specific Programs and Therapeutic Opportunities.

Autophagy is increasingly recognised as a regulator of epithelial stress adaptation in the skin, but its role in inflammatory dermatoses remains difficult to interpret because autophagy intersects with barrier biology, keratinocyte differentiation, immune signalling, intracellular trafficking and lysosomal degradation. Keratinocytes are immune-competent epithelial cells that integrate cytokine exposure, oxidative stress, microbial sensing and tissue injury into inflammatory output. In this review, we synthesize evidence on keratinocyte autophagy as a disease-dependent regulator of epidermal ho

Gut and skin microbiome-metabolome pathways in vitiligo: from dysbiosis to immune activation and melanocyte dysfunction.

Vitiligo is an autoimmune depigmenting disorder driven by genetic susceptibility, oxidative stress, immune dysregulation, and progressive melanocyte loss. Emerging microbiome-metabolome evidence suggests that gut and skin dysbiosis may contribute to disease progression through the gut-skin axis. This review integrates current evidence linking microbial imbalance with immune activation and melanocyte dysfunction in vitiligo. Studies in patients and experimental vitiligo models show altered gut and skin microbial diversity, enrichment of inflammation-associated taxa, depletion of commensal bacte
