International modified Delphi consensus statement on visible light photoprotection: Effects, measurement, and recommendations.

Exposure to solar UVR is well-known to elicit photobiologic effects on the skin. However, exposure to visible light (VL) is now known to contribute to numerous cutaneous responses, such as post-inflammatory hyperpigmentation, and play a role in melasma and some photodermatoses. Unlike UVR, no standard methods have been agreed upon for assessing VL photoprotection. The aims of this modified Delphi consensus process were to convene an international panel of experts in VL photobiology to define the cutaneous effects of VL on the basis of current best evidence, determine the populations most in ne

Drug repurposing for skin pigmentation disorders: Discovery of novel tyrosinase inhibitors.

Skin pigmentation disorders present a significant therapeutic challenge, often arising from disturbances in melanin metabolism that lead to uneven pigmentation and skin damage. Tyrosinase (TYR), the key enzyme in melanin biosynthesis, serves as a crucial therapeutic target for managing hyperpigmentation and pigmentary skin diseases. In this study, we performed structure-based virtual screening of a comprehensive library of FDA-approved drugs and chemical active agents against tyrosinase and selected top-ranked compounds based on predicted binding affinities and favorable binding poses for expe

Nonapeptide AOP-P1 ameliorates UVB-induced hyperpigmentation by suppressing melanogenesis via targeting natriuretic peptide receptor 2.

Hyperpigmentation disorders remain challenging to treat because most available agents mainly act on downstream melanogenic enzymes rather than upstream regulatory mechanisms. In this study, we identified AOP-P1, an amphibian skin-derived nonapeptide, as a potent anti-melanogenic peptide with nanomolar activity and no detectable cytotoxicity. By integrating in vitro and in vivo functional assays with transcriptomic profiling, receptor-binding studies, genetic perturbation, and pathway validation, the natriuretic peptide receptor 2 (NPR2) was identified as a unrecognized upstream regulator of me

Report From the International Conference on Incontinentia Pigmenti: Translating Discovery to Therapy.

Incontinentia pigmenti (IP) is a rare, X-linked dominant condition characterized by a perinatal rash as well as abnormalities of the central nervous system (CNS), eyes, hair, nails, and teeth. The CNS and ophthalmic manifestations are potentially severe and may lead to neurologic impairment and vision loss. Due to the rarity of IP, there remains a limited understanding of its physiology, and the guidelines for screening, clinical staging, and management of complications remain controversial. To address these gaps, a multidisciplinary, international conference on IP was held in Charlotte, North

Acquired dermal macular hyperpigmentation: a unified spectrum of dermal pigmentary dermatoses.

Acquired dermal macular hyperpigmentation (ADMH) encompasses a group of dermal pigmentary disorders (lichen planus pigmentosus, erythema dyschromicum perstans and Riehl melanosis/pigmented cosmetic dermatitis) that present with insidious-onset grey-to-brown macules and patches due to dermal melanin deposition. Traditionally regarded as distinct entities, these disorders share overlapping clinical, dermoscopic and histopathological features, leading to the adoption of ADMH as a unifying term. In this focused review, we discuss the pathogenesis and clinical features of ADMH, as well as diagnosti

Efficacy of immune checkpoint inhibitors in patients with xeroderma pigmentosum and advanced cutaneous squamous-cell carcinoma and literature review.

AIMS: Xeroderma pigmentosum (XP) is a disorder characterized by defective DNA repair, leading to the early development of cutaneous malignancies, including cutaneous squamous cell carcinomas (cSCC). The management of locally advanced and metastatic cSCC in XP patients is particularly challenging, primarily due to the risk of major surgical sequelae and the hazardous use of radiotherapy. This study aims to evaluate the efficacy and safety of anti-PD1 immunotherapy in XP patients with unresectable cSCC in Mayotte and Réunion Island and to conduct a literature review. METHODS: This retrospective

Antibody Deficiency in Xeroderma Pigmentosum.

We describe a 3-year-old patient with xeroderma pigmentosum (XP) and genetically confirmed XPA deficiency who presented with recurrent infections in early childhood. Immunological assessment revealed mild hypogammaglobulinemia with IgG2 and IgG3 subclass deficiencies, as well as impaired humoral immunity demonstrated by a reduced antibody response to repeated vaccinations against bacterial antigens. Flow cytometric analysis further showed an altered distribution of peripheral T helper (TH) cell subsets. In addition, we report a second case: a 33-year-old XP patient with ERCC4 deficiency who al

Effectiveness of omega-3 poly unsaturated fatty acids on the treatment of gingival hyperpigmentation: a randomized controlled clinical trial.

BACKGROUND: In dentistry, gingival hyperpigmentation is a common aesthetic issue caused mainly by excessive melanin deposits. Traditional surgical methods for depigmentation, like surgical techniques, can be effective, but they often associated with pain after the procedure and longer healing times. Because of their anti-inflammatory and antioxidant qualities, omega-3 polyunsaturated fatty acids (PUFAs) have demonstrated potential benefits in tissue regeneration and dermatological depigmentation. OBJECTIVE: To investigate the effectiveness of locally and systemically administered Omega-3 (PUFA

Clinicopathological and molecular characterization of tumor-associated macrophages in sporadic and Xeroderma Pigmentosum-related cutaneous melanoma.

BACKGROUND: Xeroderma pigmentosum (XP) is a rare autosomal recessive disorder, resulting from genetic defects in the nucleotide excision repair (NER) pathway. It predisposes individuals to skin cancers, including melanoma. The melanoma microenvironment is enriched with inflammatory mediators, particularly macrophages, which may serve as prognostic biomarkers or therapeutic targets. While inflammation is linked to melanoma outcomes, its role in carcinogenesis among XP patients remains poorly understood. METHODS: We assessed the cytokine profiles in the sera of XP-melanoma patients, as well as t

Interleukin 32 Exacerbates Vitiligo by Promoting Inflammation and Oxidative Stress-induced Injury in Melanocytes.

Oxidative stress-mediated melanocyte injury and subsequent inflammation constitute essential pathogenic mechanisms underlying vitiligo. The secretion of diverse cytokines plays a crucial role in modifying the inflammatory environment characteristics of skin lesions. Interleukin 32 (IL32) is a multifunctional cytokine recognized for its potent pro-inflammatory effects. Here, we observed significantly enhanced IL32 expression in vitiligo skin lesions. The present study investigates the role of IL32 in oxidative stress-driven melanocyte inflammation and its contribution to vitiligo progression by

Therapeutic Potential of Cysteine and Its Derivatives in Dermatology.

Cysteine is a sulfur-containing amino acid that plays a central role in skin physiology through thiol-mediated redox regulation and glutathione (GSH) synthesis. It critically influences melanogenesis, collagen homeostasis, and wound healing. However, its clinical application is limited by poor stability and bioavailability. In this review, we provide a mechanistic and comparative analysis of cysteine and its derivatives, including N-acetylcysteine (NAC), cysteinamide (C-NH2), GSH, and related compounds. These derivatives regulate melanogenesis by modulating dopaquinone pathways and tyrosinase

Differentiating solar lentigines from post-inflammatory hyperpigmentation with hyperspectral imaging: A pilot study.

OBJECTIVE: Solar lentigines and post-inflammatory hyperpigmentation (PIH) are common pigmentation disorders, and they are usually differentiated by a trained dermatologist. Accurate identification of the spot type is essential for providing patients with the most appropriate skin treatment. The aim of this study was to evaluate the capacity of hyperspectral imaging (HSI) in characterizing and discriminating solar lentigines from PIH, that is, to identify the colorimetric features and chromophore concentrations associated with each pigmentation disorder. METHODS: Acquisition of HSI was perform

Reduced gene expression and missense mutations in the transporter protein SLC45A2 in a hypopigmented Egyptian rousette fruit bat (Rousettus aegyptiacus).

Oculocutaneous albinism-characterized by absent or decreased melanin in the skin, eyes and hair-often co-occurs with sensory, skin and immunity disorders. The genetic basis of albinism in humans is complex, with many loci implicated in multiple forms of the disorder. Less is known about the underlying genetic causes of albinism and leucism in other species, and cross-species studies of the molecular correlates of hypopigmentation could highlight common and conserved pathways underlying mammalian pigmentation disorders. We characterize the putative causal loci of reduced pigmentation in an Egyp

Genetic Features of Albinism: A Comprehensive Analysis in the Russian Population.

We present a comprehensive molecular and epidemiological analysis of albinism in a Russian cohort, comprising 177 probands with isolated (OCA1, OCA2, OCA3, OCA4, and OA) and syndromic (HPS-associated) forms of the disease. The comparative analysis of population frequencies between the National Genetic Initiative "100,000 + Me" (NGI) project and GnomAD (v3.1.2) revealed variants in the TYR gene (NM_000372.5):c.650G>A and c.1037-7 T>A, which are specifically prevalent in the Russian population. The functional analysis was provided for variants in the TYR gene, which can probably affect splicing.
