Primary amoebic meningoencephalitis (PAM) or "brain-eating amoeba disease" is a relatively rare but typically fatal infectious disease of increasing global concern. Discovery of PAM and that Naegleria fowleri is the causative agent responsible are frequently credited to a paper published by the Australian pathologists Malcolm Fowler and Rodney F. Carter in 1965. It was significant as confirmation of an earlier suspicion that free-living, non-parasitic amoebae may be opportunistic pathogens of the human central nervous system. Like many discoveries, however, the historical details are rather mo

The genus Naegleria is a taxonomic subfamily consisting of 47 free-living amoebae. The genus can be found in warm aqueous or soil habitats worldwide. The species Naegleria fowleri is probably the best-known species of this genus. As a facultative parasite, the protist is not dependent on hosts to complete its life cycle. However, it can infect humans by entering the nose during water contact, such as swimming, and travel along the olfactory nerve to the brain. There it causes a purulent meningitis (primary amoebic meningoencephalitis or PAME). Symptoms are severe and death usually occurs withi

BACKGROUND: Acanthamoeba is a free-living protozoan widely distributed in the environment and causes Acanthamoeba keratitis, skin, and brain disease. Acanthamoeba can exchange genes, potentially increasing antimicrobial resistance and virulence. Therefore, this systematic review aimed to summarize published studies on horizontal gene transfer (HGT) between Acanthamoeba and its intracellular microorganisms and to evaluate the impact of HGTs on the pathogenicity of Acanthamoeba.
METHODS: This systematic review was conducted following the recommended reporting guidelines of the Preferred Reporti

We discuss some of the major cerebral parasites responsible for neglected diseases affecting humanity, especially in low-income countries. The World Health Organization states that cerebral malaria caused by Plasmodium falciparum is responsible for over 20% of deaths, especially in paediatric age. In addition to microscopy and molecular techniques, diagnosis can also be made with fundus magnetic resonance imaging and retinal fluorescein angiography, which have recently proven useful for assessing and understanding the clinical status of brain malaria. Currently, the best available treatment is

Objective:Naegleria fowleri, known as the brain-eating amoeba, is a thermophilic, freshwater amoeba causing primary amoebic meningoencephalitis (PAM), a disease that progresses rapidly from symptom onset to death. Climate change is causing surface water temperatures to increase, providing a hospitable environment for N. fowleri, possibly increasing risk factors for PAM. This review synthesizes the peer-reviewed biomedical literature published between January 2012 and December 2025, examining the risk of N. fowleri infection in the context of a warming climate. Methods: A scoping review was con

Background and objectives Acanthamoeba spp. are free-living amoebae causing rare but frequently fatal, granulomatous amoebic encephalitis (GAE). Understanding their genotypic distribution is essential for defining epidemiology and identifying environmental sources. This study investigated Acanthamoeba in clinically suspected cases of GAE and in environmental water sources across India. Methods A total of 125 clinical specimens from patients with suspected GAE [cerebrospinal fluid (CSF), brain biopsy tissue, or pus] and 14 environmental water samples (pond, well, and household water) were analy

Brain infection by the pathogenic free-living amoebae Naegleria fowleri is a life-threatening illness, with a mortality rate >95% in the US. One of the challenges faced in treating these infections is the lack of efficacious drugs. Here, we further characterize two leads. The first, a N. fowleri enolase inhibitor (HEX ((1-hydroxy-2-oxopiperidin-3-yl) phosphonic acid)), emerged from a target-based approach while the other, BDGR-20191 (rac-(4aR,13bS)-10,11-dichloro-4-(oxetan-3-ylmethyl)-1,2,3,4,4a,5,6,13b-octahydro-8H-[1,6]naphthyridino[5,6-b]quinazolin-8-one), was identified in a phenotypic scr

BACKGROUND: Primary amoebic meningoencephalitis (PAM), caused by Naegleria fowleri, is a rare but often fatal central nervous system infection that mimics bacterial meningitis. This similarity can lead to misdiagnosis and delayed treatment, especially in neonates and infants, resulting in poor outcomes. This study evaluates the clinical and paraclinical features, diagnostic challenges, treatment strategies, and outcomes of PAM in this vulnerable population, emphasizing the importance of early and accurate diagnosis and treatment.
METHODS: This systematic review followed the PRISMA statement.

Neuroinvasive free-living amoebae (FLA), particularly Naegleria fowleri and Acanthamoeba spp., are responsible for rare but devastating infections of the central nervous system (CNS). Approximately 480 cases of primary amoebic meningoencephalitis (PAM) and fewer than 200 well-documented cases of Acanthamoeba-associated granulomatous amoebic encephalitis (GAE) have been reported worldwide. Mortality rates frequently exceed 90%. PAM typically develops following exposure to warm freshwater contaminated with N. fowleri and progresses rapidly in otherwise healthy individuals. In contrast, GAE usual

Free-living amebas such as Acanthamoeba spp., Balamuthia mandrillaris, and Naegleria fowleri can cause infections in humans and animals. Here, we describe 2 cases of meningoencephalitis and an outbreak of multisystemic disease caused by Acanthamoeba sp. in cattle from Central-Western Brazil. Within 3 d of onset, 2 adult cattle from 2 different herds developed acute neurologic signs, including incoordination progressing to flaccid paralysis, opisthotonos, recumbency, and spontaneous death (case 1), and blindness, behavioral alterations, and flaccid paralysis leading to euthanasia (case 2). Naegleria fowleri is a free-living amoeba that causes a rare but almost always fatal brain infection known as primary amoebic meningoencephalitis (PAM). Although amphotericin B (AmB) is considered the primary treatment for PAM, it is often recommended in combination with several other drugs due to side effects associated with high concentrations. In this study, we investigated whether histone deacetylase inhibitors (HDACis) enhance the amoebicidal activity of AmB against N. fowleri. Anti-proliferative and cytotoxic activities were assessed using the cell counting kit and lactate dehydrogenase

Granulomatous amoebic encephalitis (GAE) is a rare but highly fatal central nervous system infection caused primarily by Acanthamoeba spp. and Balamuthia mandrillaris. Delayed diagnosis and the absence of standardized treatment protocols contribute to mortality exceeding 90%. This review summarizes current knowledge regarding GAE pathogenesis, diagnosis, therapeutic challenges, and emerging treatment strategies. The pathogenesis involves amoebic adhesion, secretion of proteases and phospholipases, host inflammatory responses, and blood-brain barrier disruption. Recent advances in molecular dia

Balamuthia mandrillaris is a rare and difficult-to-diagnose infection with high mortality. We present the case of an immunocompetent toddler presenting with central nervous system (CNS) vasculitis. He was initially diagnosed with Takayasu arteritis and had an initial period of improvement following immunosuppressive therapy. Subsequently, he had rapid decompensation with diffuse intracranial lesions progressing to severe neurological injury and compassionate extubation. Metagenomic sequencing of the CSF and pathology and PCR from brain biopsy were positive for B. mandrillaris To our knowledge,

Naegleria fowleri is a free-living amoeba that causes primary amoebic meningoencephalitis (PAM) in healthy humans. Infection occurs through deep inhalation of contaminated water during water-related activities. Treatment of PAM is complicated due to the low efficacy and high toxicity of available drugs. Therefore, novel therapeutics and control agents are urgently needed. Since this amoeba shares an aquatic habitat with other microbes, it is logical to search for bioactive molecules produced by aquatic bacteria to find novel therapeutic and control agents to manage N. fowleri infection. The "brain-eating amoeba" Naegleria fowleri dwells in ponds where it normally feeds on bacteria, but if it enters the brain it can cause a deadly infection. To establish infection, N. fowleri must migrate through different environments-along olfactory axons, through openings in the cribriform plate, and within brain tissue-yet how it does so remains unknown. As a model for N. fowleri migration within these environments, we examine how its nonpathogenic relative, Naegleria gruberi, navigates environments of distinct geometries. We show that Naegleria uses both actin-rich protrusions and membran

Naegleria fowleri is the etiological agent of primary amoebic meningoencephalitis, a fulminant infection of the central nervous system that leads to death in most cases. Currently, no standardised therapy is available, and existing treatments are limited by inconsistent efficacy and significant toxicity. β-Hydroxy-γ-lactones previously described as trypanocidal and leishmanicidal agents share structural features, a reactive lactone carbonyl and a stereodefined hydroxyl group, that are commonly linked to disruption of redox and mitochondrial homeostasis in protozoan parasites. Based on this pre

Amoebiasis, a gastrointestinal infection caused by Entamoeba histolytica, is the third leading cause of mortality worldwide among parasitic infections with over 100,000 deaths annually. Apart from dysentery, it can manifest as extraintestinal disease, most commonly liver abscess, and rarely pulmonary, cardiac, and brain involvement. This case demonstrates the challenging management of a complicated amoebic liver abscess (ALA) with pulmonary involvement and venous thromboembolism. A 26-year-old male presenting with pleuritic right hypochondriac pain, fever, and dyspnoea underwent hepatobiliary

Amoebic infections of the brain can cause brain abscesses and meningoencephalitis. Brain abscess is a rare form of extraintestinal amoebiasis caused by hematogenous spread from intestinal infection with Entamoeba histolytica, which occurs through oral intake of contaminated water and food or fecal-oral transmission. It is mostly associated with amoebic liver abscesses and can be effectively treated with metronidazole. Meningoencephalitis caused by pathogenic free-living amoebae is extremely rare. Primary amoebic meningoencephalitis (PAM), caused by Naegleria fowleri infection of the olfactory

The month of March is dedicated to Colon Cancer Awareness. Worldwide, colorectal cancer (CRC) incidence has been on the rise. It is currently the third most common cancer in men (746,000 cases, 10.0% of the total) and the second in women (614,000 cases, 9.2% of the total).1 Arecent meta-analysis reported a 61% risk reduction in CRC incidence with colonoscopy.2 Unlike screening programs for breast and prostate cancers, not only has CRC screening reduced mortality from colon cancer and detected early CRC, it has also decreased the incidence of CRC through detection and removal of pre-cancerous l
