Detection and identification of Naegleria species along with Naegleria fowleri in the tap water samples.

Naegleria fowleri, the causative agent of Primary Amoebic Meningoencephalitis (PAM), is commonly found in warm freshwater environments and can enter the brain through nasal passages during activities like swimming or ablution. PAM has a high fatality rate, raising concerns about its global health impact. In Pakistan, particularly in Karachi, a significant number of cases have been reported, often with no history of recreational water exposure, but with regular ablution using tap water. This study analyzed the physicochemical parameters, abundance of total and fecal coliforms, and detected N. f

An expanded phylogenomic analysis of Heterolobosea reveals the deep relationships, non-canonical genetic codes, and cryptic flagellate stages in the group.

The phylum Heterolobosea Page and Blanton, 1985 is a group of eukaryotes that contains heterotrophic flagellates, amoebae, and amoeboflagellates, including the infamous brain-eating amoeba Naegleria fowleri. In this study, we investigate the deep evolutionary history of Heterolobosea by generating and analyzing transcriptome data from 16 diverse isolates and combine this with previously published data in a comprehensive phylogenomic analysis. This dataset has representation of all but one of the major lineages classified here as orders. Our phylogenomic analyses recovered a robustly supported

Cryptic presentation: primary amoebic meningoencephalitis mimicking a high-grade malignancy resulting in surgical fatality within 48 hours. Illustrative case.

BACKGROUND: A 71-year-old male presented with weakness of the right upper limb and headache for the past 3 months. Brain magnetic resonance imaging (MRI) with contrast showed a left frontal space-occupying lesion, suggestive of a high-grade malignancy. Awake craniotomy with complete excision of the lesion was performed under immunofluorescence guidance. On postoperative day 2, the patient developed signs of meningoencephalitis and died. The histopathology report showed an abscess with parasitic infection, suggestive of primary amoebic meningoencephalitis (PAM), an infection caused by Naegleria

A Balamuthia amoebic encephalitis survivor in China, and literature review.

Balamuthia amoebic encephalitis (BAE) is a rare, fatal parasitic infection of the central nervous system, with a current mortality rate above 95%. The high fatality rate is largely attributed to atypical clinicopathological features, delayed diagnosis, and the absence of effective treatment methods, so quick recognition of this disease is vital. In this paper, we present a survivor of BAE, who was confirmed through histologic examination and metagenomic next-generation sequencing (mNGS) of brain lesions. This case, unlike most previous reports, was a successful survival case. It highlights the

Emerging patents versus brain eating amoebae, Naegleria fowleri.

Primary Amoebic Meningoencephalitis (PAM) is a severe and often fatal infection caused by the free-living amoebae Naegleria fowleri. This condition typically results from exposure to contaminated warm freshwater/inadequately treated recreational water/or ablution/nasal irrigation with contaminated water. The management of PAM is hindered by the absence of effective treatment coupled with challenges in early diagnosis. This review explores emerging patents that could be utilized for the treatment, diagnosis of PAM, as well as water treatment. Recent patents from the past five years, along with

Advancing the understanding of Naegleria fowleri: Global epidemiology, phylogenetic analysis, and strategies to combat a deadly pathogen.

Naegleria fowleri is a rare but deadly pathogen that has emerged as an important global public health concern. The pathogen induces primary amoebic meningoencephalitis (PAM), a rapidly progressive and almost always fatal life-threatening brain infection. The devastating impact of N. fowleri and the high mortality rate underscores a deeper understanding and the development of innovative strategies to tackle this issue. Despite various studies that have been conducted on N. fowleri, a comprehensive review that integrates recent findings and addresses critical gaps in understanding remains lackin

Exploring cotton plant compounds for novel treatments against brain-eating Naegleria fowleri: An In-silico approach.

To find potential inhibitors of Naegleria fowleri S-adenosyl-L-homocysteine hydrolase (NfSAHH), a brain-eating parasite, structure-based drug design was adopted. N. fowlerica causes primary amebic meningoencephalitis (PAM), a fatal central nervous system (CNS) disorder if untreated. NfSAHH protein (PDB ID: 5v96), involved in parasite growth and gene regulation, was targeted and screened against 163 metabolites from Gossypium hirsutum (cotton plant). With the aid of different software and web tools, the metabolites were subjected to several analyses. The RMSD was evaluated to validate our molec

Cloning of nf-profilin and intercellular interaction with nf-actin in Naegleria fowleri cysts.

Naegleria fowleri is a free-living amoeba found in lakes, soil, hot springs, and poorly chlorinated swimming pools. It is pathogenic to humans, causing a rare and fatal brain infection known as primary amoebic meningoencephalitis (PAM). A previous study utilized RNA-seq analysis to examine genes expressed in N. fowleri cysts and trophozoites, focusing on the nf-profilin gene, which showed high expression in cysts. Profilin is a small actin-binding protein that regulates nf-actin polymerization and cell movement. Sequence analysis revealed 83% similarity with non-pathogenic N. gruberi and 38% s

The role of plasma metagenomic sequencing in identification of Balamuthia mandrillaris encephalitis.

Balamuthia mandrillaris is a rare, free-living amoeba (FLA) that causes granulomatous amoebic encephalitis, a disease with close to 90% mortality. The geographical ranges of many FLA are expanding, potentially increasing human exposure to B. mandrillaris. Here, we report a case of a 58-year-old woman with progressive neurological symptoms, ultimately diagnosed postmortem with B. mandrillaris encephalitis through plasma metagenomic next-generation sequencing (mNGS) despite negative results on both cerebrospinal fluid (CSF) mNGS and CSF PCR testing. Histologic analysis and real-time PCR (qPCR) s

Amoebicidal thymol analogues against brain-eating amoeba, Naegleria fowleri.

Naegleria fowleri, known as the brain-eating amoeba, is the pathogen parasite that causes primary amoebic meningoencephalitis. None of the currently available therapies are fully effective, mainly due to the inefficacy of pharmacotherapy. In this regard, natural products and related compounds represent a promising strategy for amoebicidal drug discovery. Herein, a series of eight monoterpene phenol derivatives of thymol bearing ester, carbonate, or carbamate moieties were prepared, and screened as potential amoebicidal agents on N. fowleri. The cytotoxicity of these compounds on murine macroph

Balamuthia amoebic encephalitis directly causing intracranial infection: A case report.

Balamuthia amoebic encephalitis (BAE) is a rare and often fatal central nervous system (CNS) infection caused by Balamuthia mandrillaris, a free-living amoeba typically found in soil and water. This organism can invade the brain directly, bypassing other organs, making early diagnosis particularly challenging. Symptoms often do not appear as distinctive early warning signs, and many patients do not experience noticeable skin lesions or systemic symptoms before neurological manifestations emerge. Balamuthia can enter the body through various routes, including the respiratory tract, skin, or gas

Fatal amoebic meningoencephalitis caused by Balamuthia mandrillaris in Pongo pygmaeus and first case report in Pan troglodytes verus.

Balamuthia mandrillaris is an amoeba that can cause granulomatous amoebic encephalitis (GAE) as well as lung and skin infections in both humans and animals. Studies on B. mandrillaris-related GAE cases have increased in recent years. This amoeba has been identified as a cause of encephalitis and death in several non-human primates. In this study, we report a case of a 4-year-old female Bornean orangutan (Pongo pygmaeus) in a zoological center that exhibited neurological symptoms for several days. After unsuccessful treatments and a worsening in her condition, euthanasia was deemed necessary. A

[NAEGLERIA FOWLERI AMOEBA MENINGITIS - REVIEW AND UPDATE].

Naegleria fowleri is an amphizoic, thermophilic, flagellate amoeba, also known as the brain eating amoeba. It can survive in water, soil, and mostly in the human host's central nervous system. It thrives in fresh water and a hot climate. Naegleria fowleri causes infection in children as well as adults, following aspiration of fresh water into the nostrils. It causes a deadly infection called primary meningoencephalitis [PAM]. PAM is more common in developing countries; a few cases occurred in developed countries such as the USA, and even in Israel.

Unveiling the molecular architecture of the mitochondrial respiratory chain of Acanthamoeba castellanii.

Acanthamoeba castellanii is a ubiquitous free-living amoeba that can cause severe infections in humans. Unlike most other organisms, A. castellanii possesses a "complete" mitochondrial respiratory chain, meaning it con-tains several additional enzymes that contribute to its metabolic versa-tility and survival in diverse environments. This review provides a com-prehensive overview of the mitochondrial respiratory chain in A. castellanii, focusing on the key alternative components in-volved in oxidative phosphorylation and their roles in energy metabo-lism, stress response, and adaptation to var

A comprehensive review on the neurological impact of parasitic infections.

Parasitic infections continue to receive little attention in research and health care policy, despite their high prevalence and potential to cause serious neurological complications. For instance, neurocysticercosis, caused by the larval form of Taenia solium, is a leading cause of acquired epilepsy in endemic areas, affecting more than 2 million people worldwide. Toxoplasma gondii, a widespread protozoan parasite, has been linked to behavioral changes, mental health disorders, and an increased risk of schizophrenia, especially in individuals with prior exposure. Importantly, Toxoplasma infect
