Pathogenesis of pathogenic Naegleria amoeba.

In brain sections of the Naegleria-caused cases of primary amoebic meningoencephalitis, extensive demyelinization was found in the white matter, besides the severe histopathological changes and large clusters of trophozoites in the grey matter. The myelinoclasis appeared to be a result of a specific phospholipolytic effect, unlike that in post-viral encephalomyelitis, which has been attributed to vascular blockade or hemorrhages. In monkey kidney cell cultures a very early cytopathic effect was observed and traced to the cytolytic property of the seeding culture fluid. Rat brain slices inocula

Consciousness and the brain: evolutionary aspects.

Self-reflective consciousness is distinguished from 'simple' awareness and from conscious awareness. Awareness is a fundamental property of living matter as it reacts to environmental stimuli. A hypothesis is advanced to account for the general phenomenon of decussation of nerve fibres in the central nervous system of vertebrates. This feature, the crossing over of nerve fibres to the opposite side of the brain, is interpreted as exemplifying the inherent caution of living forms. The evolution of increasing complexity of structure in spinal cord and brain is a prerequisite for increasing aware

Subarachnoid space of the CNS, nasal mucosa, and lymphatic system.

We have briefly reviewed the literature pertaining to the movement of tracer molecules and infectious organisms within the olfactory nerve. There is a body of evidence indicating that tracers placed in the CSF will quickly move via the olfactory nerve to the nasal mucosa and then to the cervical lymph nodes. Organic and inorganic tracer materials and organisms as diverse as viruses, a bacillus, and an amoeba, when placed in the nasal cavity, have been shown to move from the nasal mucosa via the olfactory nerve to the olfactory bulb and the CSF. We think that a portion of the data on tracer mov

Differences in virulence of Naegleria fowleri.

All pathogenic Naegleria fowleri isolated from the environment were highly virulent to mice when instilled intranasally. Axenic cultivation gradually decreased virulence of highly virulent strains. This decrease was most pronounced in environmental isolates and of minor importance in N. fowleri isolated from human cerebrospinal fluid. The low virulent strains obtained by continuous axenic cultivation appeared after clonation to consist of individuals with different virulence. Virulence could be enhanced in low virulent strains by brain passage and passages in Vero cell cultures, but could not

Primary amebic meningoencephalitis with cerebral and cerebellar abscesses: case report.

A case of fatal primary amebic meningoencephalitis is reported. We need to remind ourselves that, if the usual laboratory tests fail to show any microbial or fungal agents either in the CSF or in fluids or tissues obtained at operation, it is imperative that a warm wet slide preparation be made. Primary amebic meningoencephalitis is a very rare disease, and it is only by prompt recognition of the amebae that we can make an early diagnosis and institute the appropriate therapy.

Naegleria fowleri in chick embryos. Effects of embryo age and incubation temperature, and the infectivity of embryo-derived amebae for mice.

Chick embryos were infected with Naegleria fowleri which was initially isolated from an ultimately fatal human case. Following inoculation of equivalent numbers of amebae on the chorioallantoic membrane, younger embryos died earlier than older embryos infected at the same time. Incubation of infected embryos at 32 degrees C prolonged survival only slightly in comparison with those at 37 degrees C. N. fowleri maintained for more than 25 serial passages in chick embryos retained infectivity for mice and the ability to convert to the biflagellate form in vitro.

A case of primary amebic meningoencephalitis. Light and electron microscopy, and immunohistologic studies.

A 56-year-old alcoholic developed primary meningoencephalitis and died 3 weeks later. Autopsy revealed multiple areas of necrosis and acute and granulomatous inflammation in the brain and pancreas. Trophozoites and cysts were discovered in the brain and trophozoites alone in the pancreas. Morphologic studies revealed some parasitic features described as characteristic for Naegleria, but clinical, histological and immunohistological features favored in Acanthamoeba infection. This study calls for caution in the use of pure morphological criteria for the differential diagnosis of Naegleria and A

Primary amoebic meningoencephalitis.

As a serendipitous by-product of polio virus research, a highly fatal amoebic meningoencephalitis was recognized in animals. The causative microorganisms, contaminants of the viral cultures, were identified as small soil amoebae. These organisms, previously considered non-pathogenic, are prevalent throughout the world. Based on animal studies, the original investigators suggested the possibility of a similar disease in humans. Seven years later, human cases of amoebic meningoencephalitis were reported from widely separated areas of the world. Since 1965, a total of 79 cases have been reported.

Meningoencephalitis and brain abscess due to a free-living amoeba.

A 47-year-old diabetic woman developed fatal meningoencephalitis due to a free-living amoeba. The responsible organism appeared to be neither Naegleria nor Acanthamoeba-Hartmannella. Both acute and chronic (granulomatous) inflammatory reactions of the brain were present, and both cysts and trophozoite forms were readily visualized. The latest classification and methods of identification of such amoebae are reviewed and their threat to public health underscored. It should be realized that a variety of free-living amoebae exist in nature, which potentially can produce meningoencephalitis in huma

Retrospective identification of Acanthamoeba culbertsoni in a case of amoebic meningoencephalitis.

Acanthamoeba culbertsoni was identified retrospectively in a case of amoebic meningoencephalitis, previously reported by Jager and Stamm (Lancet, 2, 1343, 1972). This is the second report of this species causing secondary infection in man. Positive results were obtained only with anti-A. culbertsoni sera when the brain sections were stained by the indirect immunofluorescence antibody test with various antisera produced against different Acanthamoeba species. Antiserum raised against purified plasma membranes of A. culbertsoni showed once more its highly specific diagnostic value.

Free-living amebic meningoencephalitides: comparative study.

Primary amebic meningoencephalitis (PAM) is an infectious disease essentially confined to the central nervous system (CNS) and caused by free-living ameboflagellates of the genus Naegleria (N) and Acanthamoeba (A). N produces an acute fulminant, usually fatal hemorrhagic-necrotizing meningoencephalitis, associated with an inflammatory reaction composed of neutrophils, eosinophils, macrophages and a few lymphocytes. The incubation period is between 3-7 days. Typically, PAM occurs in healthy, young individuals, who frequently have a history of swimming or washing their face in infested waters. T

Meningoencephalitis due to Acanthamoeba SP. Pathogenesis and clinico-pathological study.

Amebic Meningoencephalitis (AM) and Primary Amebic Meningoencephalitis (PAM) are infectious diseases essentially confined to the Central Nervous System (CNS) and caused by free-living amebas of the genus Acanthamoeba (A.) and Naegleria (N.) respectively. AM due to A. sp. (Acanthamoeba castellanii and Acanthamoeba culbertsoni) have been reported in chronically ill debilitated individuals, some of them under immunosuppressive therapy, or in immunologically impaired patients without a history of recent swimming in contrast to cases due to N. sp. which usually occurs in healthy, young individuals

Amebiasis in goldfish.

Five cases of amebiasis were diagnosed in goldfish (Carassius auratus) from home aquariums and from a laboratory aquarium. Granulomas containing amoebae were in many organs but were most numerous in kidneys. Because there were pseudopods, food vacuoles, vesicular nucleoli and other ultrastructural characteristics of the organisms, we identified the organisms as amoebae. On the basis of mitotic stages it is possible they belong in the family Hartmannellidae.

[Amoebic meningoencephalitis by "Eaegleria" and "Acanthamoeba" (author's transl)].

Primary amoebic meningoencephalitis (ME) by Naegleria fowleri a free-living protozoon found in fresh, warm waters, is a well known fatal disease lasting less than one week. It affects sporadically swimmers and children playing in mud puddles. Less than 100 cases have been described. Recently a more rare, distinct amoebic meningoencephalitis due to some species of free-living Acanthamoeba was identified, lasting some weeks or more but still with a fatal evolution. In this case the amoebae do not always enter the brain directly through the cribrous membranes but cause mild, primary infections of
