The IKAROS Transcription Factor Gene IKZF1 as a Critical Regulator in the Pathogenesis of Childhood Acute Lymphoblastic Leukemia: Insights from a Bangladeshi Population.

UNLABELLED: The IKZF1 transcription factor gene plays a pivotal role in the commitment of multipotent progenitors to common lymphoid progenitors during lymphopoiesis. Genome-wide association studies (GWASs) have revealed an intriguing link between childhood acute lymphoblastic leukemia (ALL) and IKZF1 polymorphisms. This study investigated whether two specific IKZF1 variants, rs10272724 (T > C) and rs4132601 (T > G), are potential risk factors for childhood ALL among Bangladeshi children. In this study, we found that ALL occurs with a significantly greater frequency in male children than in fe

Pharmacophore Establishment and Optimization of Saturated 1,6-Naphthyridine-Fused Quinazolinones that Inhibit Meningoencephalitis-Causing Naegleria fowleri.

Primary amoebic meningoencephalitis (PAM) is a human brain infection caused by Naegleria fowleri with a 97% mortality rate. Quinazolinones resulting from a Mannich-coupled domino rearrangement were recently identified as inhibitors of the amoeba. Herein, we resolved the effective concentrations for 25 pilot compounds and then, using the Mannich protocol and a key late-stage, N-demethylation/functionalization, we synthesized 53 additional analogs to improve potency, solubility and microsomal stability. We established an antiamoebic quinazolinone pharmacophore, culminating in (±)-trans-57b which

Drug modifications: graphene oxide-chitosan loading enhanced anti-amoebic effects of pentamidine and doxycycline.

Acanthamoeba castellanii is the causative pathogen of a severe eye infection, known as Acanthamoeba keratitis and a life-threatening brain infection, named granulomatous amoebic encephalitis. Current treatments are problematic and costly and exhibit limited efficacy against Acanthamoeba parasite, especially the cyst stage. In parallel to drug discovery and drug repurposing efforts, drug modification is also an important approach to tackle infections, especially against neglected parasites such as free-living amoebae: Acanthamoeba. In this study, we determined whether modifying pentamidine and

Understanding the pathogenicity of Naegleria fowleri in association with N. fowleri antigen-1 (Nfa1).

Naegleria fowleri, a brain-eating amoeba, thrives in lakes and rivers with aquatic vegetation and causes primary amoebic meningoencephalitis (PAM) in humans. Most recently, it has become such a serious problem that N. fowleri was detected in tap water in Houston, USA. Several pathogenic factors are considered very important to destroy target cells in the brain. In particular, the food-cup where N. fowleri antigen-1 (Nfa1) is located, is strongly expressed in pseudopodia involved in the movement of N. fowleri, and is involved in phagocytosis by attaching to target cells. In this article, we rev

Application of ventriculoscopy in granulomatous amoebic encephalitis: a case report in China and literature review.

Balamuthia mandrillaris is a kind of brain-eating amoeba leading to granulomatous amoebic encephalitis (GAE) with a high mortality rate and limited effective treatment options. Its underlying pathological mechanisms are still poorly understood. Here, we presented a 58-year-old man with Balamuthia mandrillaris meningoencephalitis, who died 33 days after onset. In this unique case, we introduced the use of ventriculoscopy in the treatment of B. mandrillaris meningoencephalitis. Through ventriculoscopy, we observed that the inflammatory response triggered by the disease was diffuse and extensive

Balamuthia Mandrillaris Central Nervous System Vasculitis in an Immunocompetent Child: Case Report.

Balamuthia mandrillaris granulomatous amebic encephalitis is a rare disease that is associated with a high rate of mortality. Delays in diagnosis and treatment are common because of limited information on the organism in addition to its nonspecific clinical presentation. Prior reports have demonstrated that the encephalitis presents as multifocal lesions throughout the central nervous system with enhancement and edema. Here we report a case involving a 4-year-old previously healthy female child with a novel pathologic presentation of B mandrillaris infection, including vasculitis involving mul

Enlightening the promising role of nanoparticle-based treatments against Naegleria fowleri-induced primary amoebic meningoencephalitis: A brain-eating disease.

Naegleria fowleri, is the causative agent of Primary Amoebic Meningoencephalitis (PAM), a lethal acute brain inflammation with high mortality. The virulent and reproductively active trophozoite stage of N. fowleri migrates to central nervous system (CNS) by entering through nasal passage and causes severe neural infection, brain disease and inflammation with high mortality. In this review we present the current available information about N. fowleri, including its case reports, pathogenesis and the mechanism of host neuroinflammation associated with PAM. Various case reports reveal that the su

Transcriptomic profiling of "brain-eating amoeba" Naegleria fowleri infection in mice: the host and the protozoa perspectives.

The free-living amoeba Naegleria fowleri (NF) causes a rare but lethal parasitic meningoencephalitis (PAM) in humans. Currently, this disease lacks effective treatments and the specific molecular mechanisms that govern NF pathogenesis and host brain response remain unknown. To address some of these issues, we sought to explore naturally existing virulence diversity within environmental NF isolates. Herein, we purified two new NF environmental isolates (NF45 and NF1) and tested their in vivo virulence using experimental infection in mice. We found that NF45 was highly virulent (NF45_HV) compare

Brainstorm: A Case of Granulomatous Encephalitis.

BACKGROUND: Free-living amoebas (FLAs) can cause severe and fatal central nervous system infections that are difficult to diagnose. METHODS: We present the case of a 74-year-old immunocompetent woman admitted for focal neurological symptoms with enhancing lesions in the right cerebellar hemisphere. A first cerebral biopsy showed granulomatous inflammation, but no microorganisms were identified. After transient clinical improvement, she eventually deteriorated 4 months after initial presentation, with an MRI confirming multiple new masses affecting all cerebral lobes. RESULTS: A second brain

Experimental Granulomatous Amebic Encephalitis Caused by Acanthamoeba castellanii.

Acanthamoeba genus can affect humans with diseases such as granulomatous amebic encephalitis (GAE), a highly lethal neuroinfection. Several aspects of the disease still need to be elucidated. Animal models of GAE have advanced our knowledge of the disease. This work tested Wistar rats (Rattus norvegicus albinus) as an animal model of GAE. For this, 32 animals were infected with 1 × 106A. castellanii trophozoites of the T4 genotype. Ameba recovery tests were carried out using agar plates, vascular extravasation assays, behavioral tests, and histopathological technique with H/E staining. Data we

Amoebae: beyond pathogens- exploring their benefits and future potential.

Amoebae, fascinatingly diverse protists, showcase a dual nature that positions them as both friends and foes in our world. These organisms, defined by their distinctive pseudopodia, span a spectrum from harmful to helpful. On the darker side, species like Entamoeba histolytica pose serious health risks, causing intestinal and liver diseases, while the infamous "brain-eating" Naegleria fowleri leads to fatal primary amoebic meningoencephalitis (PAM), with a daunting 97% mortality rate. Other free-living amoebae, including Acanthamoeba castellanii and Balamuthia mandrillaris, also threaten the h

Laparoscopic segmental colectomy for a rare case of extraluminal amoeboma masquerading as stromal tumour of the colon.

Entamoeba histolytica is known to cause infectious colitis that presents with diarrhoea or dysentery. Extra intestinal manifestations result in amoebic abscesses to involve organs such as the liver, lungs or brain. Very rarely, it may present like a pseudotumour due to a mass formation if the primary infection was untreated or inadequately treated. We, herein, report a rare case of extraluminal amoebic pseudotumour in a young female with a diagnosis of a gastrointestinal stromal tumour of hepatic flexure of the transverse colon on pre-operative imaging and per operative findings, but final his

Similar evolutionary trajectories in an environmental Cryptococcus neoformans isolate after human and murine infection.

A pet cockatoo was the suspected source of Cryptococcus neoformans recovered from an immunocompromised patient with cryptococcosis based on molecular analyses available in 2000. Here, we report whole genome sequence analysis of the clinical and cockatoo strains. Both are closely related MATα strains belonging to the VNII lineage, confirming that the human infection likely originated from pet bird exposure. The two strains differ by 61 single nucleotide polymorphisms, including eight nonsynonymous changes involving seven genes. To ascertain whether changes in these genes are selected for during

Anti-amoebic effects of synthetic acridine-9(10H)-one against brain-eating amoebae.

Pathogenic A. castellanii and N. fowleri are opportunistic free-living amoebae. Acanthamoeba spp. are the causative agents of granulomatous amebic encephalitis (GAE) and amebic keratitis (AK), whereas Naegleria fowleri causes a very rare but severe brain infection called primary amebic meningoencephalitis (PAM). Acridinone is an important heterocyclic scaffold and both synthetic and naturally occurring derivatives have shown various valuable biological properties. In the present study, ten synthetic Acridinone derivatives (I-X) were synthesized and assessed against both amoebae for anti-amoebi

Laboratory Diagnosis of Primary Amoebic Meningoencephalitis.

Primary amebic meningoencephalitis (PAM) is a fulminant fatal human disease caused by the free-living amoeba Naegleria fowleri. Infection occurs after inhalation of water containing the amoeba, typically after swimming in bodies of warm freshwater. N. fowleri migrates to the brain where it incites meningoencephalitis and cerebral edema leading to death of the patient 7 to 10 days postinfection. Although the disease is rare, it is almost always fatal and believed to be underreported. The incidence of PAM in countries other than the United States is unclear and possibly on track to being an emer
