The Genetics of Acne.

This review addresses the genetics of acne vulgaris, the most common skin disease. It is characterized by the presence of comedones (blackheads), papules, and pustules. The condition is associated with sebaceous glands in the face and chest, which produce an oily substance called sebum. In developed nations, acne affects over 80% of adolescents. Mild disease usually resolves spontaneously. More severe acne can leave permanent, disfiguring scarring and strongly affects quality of life. In those cases, medical intervention is warranted. To date, antibiotics and retinoids (synthetic vitamin A der

Fucoidan treatment reverses hair loss and inhibits inflammatory responses in a mouse model of androgenetic alopecia.

Androgenetic alopecia (AGA), a prevalent form of hair loss, is typically treated with minoxidil and finasteride, but their efficacy and safety are somewhat constrained. Previous studies have shown that fucoidan can regulate VEGF and Wnt signaling pathways, which are critical for hair growth. Based on bioactivity screening protocols for natural product, we hypothesized that fucoidan may exert beneficial effects on AGA, potentially through modulation of the Wnt pathway and other molecular mechanisms. This study aimed to investigate the effect of fucoidan on testosterone propionate-induced AGA in

3C3 alleviates atopic dermatitis-like phenotypes through cyclophilin A and B-mediated inhibition of calcineurin-NFAT pathway.

Atopic dermatitis (AD) disrupts a patients' quality of life and is associated with other atopic disorders, such as food allergy, allergic rhinitis, and asthma. Although topical medications are widely used, they have undesirable side effects, suggesting the need for treatments with improved efficacy and safety. In this study, we identified 3C3 as a candidate compound for AD treatment using in vitro TNFα-treated HaCaT keratinocytes and a DNFB-induced AD mouse model. 3C3 treatment alleviated AD-like phenotypes and suppressed the gene activation of pro-inflammatory cytokine. We further found that

Linear Focal Elastosis: A Peculiar Dermatosis.

Linear focal elastosis (LFE), also known as elastotic striae, is a rare cutaneous condition characterized by abnormal or increased deposition of elastic fibers in the dermis. It typically presents as asymptomatic, atrophic, or hyperpigmented linear bands, most commonly on the back. We report a case of LFE in a 15-year-old healthy male who presented with multiple asymptomatic, parallel hypopigmented linear bands with hyperpigmented borders on his back. A skin biopsy revealed characteristic thickened and fragmented elastic fibers in the reticular dermis, confirming the diagnosis. The pathogenesi

Cardiovascular autonomic failure correlates with cutaneous autonomic denervation in PD and MSA.

PURPOSE: Cardiovascular autonomic failure and neurogenic orthostatic hypotension (nOH) are common and disabling in Parkinson's disease (PD) and multiple system atrophy (MSA). Recent studies have shown evidence of postganglionic autonomic denervation in MSA as well as PD. We aimed to characterise the relationship between nOH, autonomic failure and postganglionic denervation in PD and MSA. We hypothesised that postganglionic autonomic denervation contributes to the development of nOH and correlates with the severity of cardiovascular autonomic failure. METHODS: We assessed 57 patients (37 PD, 2

Efficacy and Safety of Runzaozhiyang Lotion for Chronic Hand Eczema: Protocol for a Randomized Controlled Trial.

BACKGROUND: Chronic hand eczema (CHE) is a prevalent inflammatory skin disease in clinical practice. Guidelines recommend the use of topical medications, including corticosteroids and calcineurin inhibitors; however, prolonged use of these drugs may result in adverse effects, such as skin atrophy, drug tolerance, and rebound phenomena. Moreover, these medications exhibit limited efficacy in managing chronic hypertrophic hand eczema. Traditional Chinese medicine (TCM) has a long history of treating patients. The Runzaozhiyang lotion (RZZYL) is often used in the Department of Dermatology of Guan

Assessing the link between cerebellar volume and cognitive function in Alzheimer's disease: a pilot study.

The cerebellum, long recognized for its role in motor functions, has become increasingly acknowledged for its involvement in a broader spectrum of cognitive functions. Given the cerebellum's strong neuronal connections with cerebral regions affected by neurodegenerative diseases, this study investigated the cerebellar contribution to cognitive decline in Alzheimer's disease (AD). This study retrospectively analysed 127 individuals with subjective cognitive decline (SCD, n = 46), mild cognitive impairment due to AD (MCI, n = 24), and AD dementia (ADD, n = 57). Grey and white matter volumes of t

Parry-Romberg Syndrome: A Rare Case with Diagnostic Challenges and Orthodontic Implications.

Parry-Romberg syndrome (PRS) is a rare acquired disorder characterized by progressive hemifacial atrophy involving the skin, subcutaneous tissue, muscles, cartilage, and bone. The disease usually progresses for two to 20 years before stabilizing, with extracranial involvement of the trunk and extremities occasionally reported. The etiology remains uncertain, with proposed immunological, genetic, and neurovascular mechanisms. We present the case of a 24-year-old female with PRS who exhibited intraoral and extraoral soft tissue atrophy, localized alopecia, ocular changes, and otologic involvemen

Co-occurrence of two monogenic diseases within a single family.

Kindler epidermolysis bullosa (KEB), a rare genodermatosis caused by loss-of-function mutations in the FERMT1 gene (encoding kindlin-1), is clinically characterized by congenital blistering, skin atrophy, poikiloderma, photosensitivity, and an increased predisposition to cutaneous squamous cell carcinoma (cSCC). Hearing loss, a common sensory impairment with multifactorial origins, often stems from genetic or environmental factors. Here, we report a unique familial case in which one sibling has KEB complicated by cSCC, and the other has congenital autosomal recessive non-syndromic hearing loss

Post-infection brain atrophy accelerates cognitive and molecular changes underlying dementia.

BACKGROUND: Infections have been associated with a greater risk of Alzheimer’s disease and related dementias (ADRD), but it is unclear how infections influence structural brain patterns over time, and whether post-infection brain atrophy can accelerate cognitive decline and molecular changes underlying dementia. METHODS: Using the Baltimore Longitudinal Study of Aging (BLSA; n = 793; mean age = 70.1), we examined how infections relate to longitudinal changes in machine learning-derived, 3 T-MRI neuroimaging signatures, and leveraged the UK Biobank (UKB; 1,120; mean age = 62.9 yrs) to externall
