Intracellular SCARB2 mimicry overcomes decoy failure to broadly inhibit enteroviruses.

Enterovirus A71 (EV-A71) and coxsackievirus A16 (CV-A16) are leading causes of severe hand, foot, and mouth disease (HFMD) with high childhood mortality and no specific antiviral therapy. SCARB2 is the essential uncoating receptor, yet extracellular SCARB2-Fc decoys fail to block infection, representing a decade-old paradox in enterovirus therapeutics. Here, we overcome this limitation by engineering RMP(R8), a cell-penetrating SCARB2 mimetic peptide comprising the minimal VP2-binding domain (W146-K166) fused to an octa-arginine transporter. RMP(R8) shows potent antiviral activity (EC50 = 0.15

High-throughput sequencing reveals the virome characteristics of wastewater in Gansu Province, China from 2024 to 2026.

Emerging and re-emerging infectious diseases continue to pose a persistent challenge to global public health security. As an unbiased and noninvasive population-level surveillance approach, wastewater-based epidemiology has emerged as an urban-scale monitoring tool for proactively capturing the community disease burden. In this study, metagenomic and targeted next-generation sequencing were employed to preliminarily characterize wastewater viral communities in Gansu Province from 2024 to 2026. Across the three annual sampling groups, the Simpson diversity index calculated from metagenomic taxo

Virological and epidemiological investigations of a severe hand foot and mouth disease outbreak, Vietnam, 2023.

We report on the virological and epidemiological features of a severe hand foot and mouth disease outbreak in Vietnam in 2023. Enterovirus A71 (EV-A71) subgenogroup B5, first emerging around November 2022, and imported into Vietnam through a single introduction, was the primary cause. Convalescent plasma collected from patients infected with subgenogroups B5 in 2014 and 2023 could neutralize both contemporary viruses with comparable titers, suggesting no evidence of immune evasion. However, at one and six months before the start of the outbreak, only 6.7% of 0- < 3 year-old children in Ho Chi

Long-term performance evaluation and seasonal field calibration of a low-cost particulate matter sensor.

UNLABELLED: Low-cost air quality sensors (LCS) provide high-resolution monitoring with affordability and real-time data but require performance evaluation and calibration due to probable lower accuracy compared to reference instruments and sensitivity to meteorological factors such as air relative humidity and temperature. Although most studies on low-cost sensors have been conducted in Europe and America, we did not find any study in regions like Iran, where air pollution levels and meteorological conditions differ significantly. In this study, performance evaluation and calibration of a low-

Exploratory effects of coffee-derived exosome-like nanoparticles in a fibrillar amyloid-beta 1-42-induced neurodegenerative rat model: evidence from fluorodeoxyglucose positron emission tomography and phosphorylated tau/amyloid precursor protein expression.

Enterovirus 71 (EV71) is now recognized as an emerging neurotropic virus in Asia and with Coxsackie virus (CV) it is the other major causative agent of hand-foot-mouth diseases (HFMD). Effective medications and/or prophylactic vaccines against HFMD are urgently needed. From a scientific (the feasibility of bioprocess, immunological responses and potency in animal challenge model) and business development (cost of goods) points of view, we in this review address and discuss the pros and cons of different EV71 vaccine candidates that have been produced and evaluated in animal models. Long Non-Coding RNA NEAT1 Inhibits Enterovirus 71 Replication by Enhancing IFN-β Transcription Through DDX60 Signaling: Pathological Insights and Clinical Implications.

Enterovirus 71 (EV71) is the principal pathogen linked to severe hand, foot, and mouth disease (HFMD), with its pathogenesis remaining poorly understood. Here, we found that EV71 infection dramatically increases the expression of NEAT1, resulting in the formation of paraspeckles. Notably, NEAT1 specifically enhances IFN-β transcription via the DDX60-IRF7 pathway, thereby promoting host resistance to EV71. Further experiments indicated that NEAT1 serves as a positive feedback for DDX60 signaling. In detail, NEAT1 facilitates the relocation of the paraspeckle protein SFPQ to the paraspeckle. Thi

Estimating the Uptake of and Factors Associated With a New Vaccine Outside of the National Immunisation Programme: A Case Study of the EV-A71 Vaccine Against Hand, Foot, and Mouth Disease.

The burden of hand, foot, and mouth disease (HFMD) in children under five is substantial, with the greatest burden in China. Most cases are mild, although some are severe and even fatal. A vaccine against EV-A71, the pathogen most commonly associated with severe HFMD, was licensed in China in Dec 2015 but not introduced into the National Immunisation Programme (NIP). It was hence not covered by routine national vaccine surveillance and its coverage remained unknown for the years following its initial licensure. Here we report the results of a novel data collection and analysis approach to addr

Spatial heterogeneity in the temperature-hand, foot, and mouth disease association among children: A multicounty time-series study in western China.

While meteorological and socioeconomic factors are well-documented modifiers of spatial heterogeneity in temperature-hand, foot, and mouth disease (HFMD) associations, substantial unexplained heterogeneity remains. This study investigates underexplored environmental modifiers-including extreme temperature events (heat waves and cold spells), air pollution, and normalized difference vegetation index (NDVI)-by analyzing 484,928 HFMD cases among children under six years old in western China (2013-2019) using distributed lag nonlinear models and meta-regression. We found that cold spells (accounte

A photonic crystal sensing array based on a tandem CRISPR/Cas13a system for ultra-sensitive and high-throughput detection of the CVA6 virus.

Coxsackievirus A6 (CVA6) has emerged as a major cause of hand-foot-mouth disease (HFMD), yet no standardized detection method for it is currently available. Developing a simple, sensitive, and specific CVA6 test is crucial for HFMD control and safeguarding the health of at-risk children. Herein, a photonic crystal (PC) sensing array based on a tandem CRISPR/Cas13a system has been proposed for highly specific and ultra-sensitive analysis of CVA6 RNA, without the need for reverse transcription and amplification procedures. In this strategy, two crRNAs targeting CVA 6 RNA were designed and screen

Mutation of the highly conserved amino acids in the N-terminal region of the EV-A71 VP1 protein impairs viral RNA release during virus entry.

UNLABELLED: Enterovirus A71 (EV-A71) causes hand-foot-mouth disease, which may progress to severe neurological and systemic complications in young children and infants. To deliver viral genomic RNA into the target cell for replication, the mature EV-A71 capsid particle undergoes drastic structural changes to become an expanded A particle, which is marked by the movement of the N-terminal region of viral protein VP1 from the interior to the capsid surface and the dislodgement of the pocket factor. It is not completely understood which amino acids support these key activities of VP1 during EV-A7

Shifting epidemiological patterns and strain replacement of hand, foot, and mouth disease in southern China, 2009-2022: A longitudinal study.

BACKGROUND: China launched a universal campaign of monovalent-inactivated EV-A71 vaccine in 2016. However, the monovalent-inactivated vaccine is ineffective against HFMD with other etiologies. To date, little is known about the epidemiological patterns and the replacement of the predominant pathogens that induce HFMD after the universal vaccination campaign. METHODS: Using HFMD surveillance and VP1 sequencing data in pre- and post-vaccination periods, time series analysis, and Wavelet approaches, we identified pathogen replacement, changes in the viral spectrum and epidemiological patterns of

[Hand-foot-mouth disease with atypical clinical presentation caused by Coxsackievirus A6].

Hand-foot-mouth disease is typically a mild, self-limiting enterovirus infection, most often caused by Coxsackievirus A16 and enterovirus A71. Over the past 15 years, however, Coxsackievirus A6 (CVA6) has emerged worldwide, including in Hungary, as the main pathogen responsible for atypical clinical forms. Atypical hand-foot-mouth disease differs from the classic presentation by widespread vesiculobullous and often painful skin eruptions, frequent perioral lesions and post-infectious nail changes. We present the case of a 2-year-old girl with generalized vesicobullous eruptions, in whom CVA6 (

A Meta-Analysis of Influencing Factors for Reinfection of Hand, Foot and Mouth Disease in China, Based on Adjusted Effect Estimates.

Background: Numerous studies have reported on the epidemiology of hand, foot and mouth disease (HFMD) reinfection and its potential influencing factors; however, findings regarding reinfection rates as well as determinants such as gender, age, residence, and pathogens remain inconsistent. Due to this heterogeneity in reported outcomes, a comprehensive systematic review and meta-analysis are warranted to consolidate existing evidence. Methods: Effect estimates were expressed as reinfection rates, odds ratio (OR)/hazard ratio (HR) and 95% confidence intervals (CI). When necessary, data were conv

Cost-Effectiveness Analysis of a Bivalent Vaccine for Hand, Foot, and Mouth Disease: A Simulation-Based Study in Beijing, China.

BACKGROUND: Hand, foot, and mouth disease (HFMD) remains a major public-health concern in China. While the monovalent EV-A71 vaccine has effectively reduced EV-A71-associated cases, it offers no protection against CV-A16. The introduction of a bivalent EV-A71/CV-A16 vaccine may offer broader protection, but its economic viability under different immunization strategies remains uncertain. METHODS: We developed a dynamic transmission model integrated with cost-effectiveness analysis to assess the epidemiological and economic impact of a hypothetical bivalent EV-A71/CV-A16 vaccine in China.
