Serial passaging in vitro generates a Vero cell-adapted coxsackievirus A6 strain with distinct phenotypic characteristics.

BACKGROUND: Coxsackievirus A6 (CVA6) has emerged as a major etiological agent of hand, foot, and mouth disease (HFMD), yet no commercial vaccine is available against CVA6. CVA6 exhibits poor adaptability to Vero cells, a WHO-approved substrate for human vaccine production, and the mechanisms underlying its infectivity and pathogenesis remain incompletely understood. METHODS: A Vero cell-adapted CVA6 strain was generated through serial passaging in vitro. Two recombinant viruses derived from the earlier and later generations, corresponding to passage 10 and passage 45, were generated and desig

Identification of key amino acids defining conformational neutralizing epitopes of coxsackievirus A5 using monoclonal antibody escape mutants.

BACKGROUND: Coxsackievirus A5 (CVA5) is an emerging pathogen associated with severe hand, foot, and mouth disease. Protective antibodies are critical for protection from CVA5 infection, yet the antigenic determinants of CVA5 remain poorly defined. In this study, we aimed to map the conformational neutralizing epitopes of CVA5 using monoclonal antibodies (mAbs) and escape mutant analysis. METHODS: Two IgG, one IgA, and five IgM neutralizing mAbs were purified and characterized for binding affinity and neutralization potency. Antibody pressure selection was applied to generate immune escape mut

Establishment of an oral enterovirus 71 (EV71) infection model in immunocompetent mice for antiviral therapy evaluation.

UNLABELLED: Enterovirus 71 (EV71) is one of the major pathogens responsible for hand-foot-and-mouth disease in children and is associated with severe neurological complications, including death. Despite the fact that EV71 naturally infects humans through the oral route, the absence of a simple and reliable oral infection model has significantly impeded investigations into its pathogenesis and the development of antiviral interventions. In this study, we established a robust oral EV71 infection model in widely used immunocompetent BALB/c mice, employing a clinical isolate devoid of adaptive mut

Global Evolution and Phylodynamics of Coxsackievirus A4.

Coxsackievirus A4 (CVA4) is an enterovirus associated with diverse clinical syndromes, yet its global evolution and transmission dynamics remain poorly defined. This study aimed to characterize the molecular epidemiology, evolutionary history, and phylodynamic patterns of CVA4 at both local and global scales. In 2024, 34 CVA4 infections were identified during a localized herpangina outbreak in Beijing, primarily affecting children under 5 years of age, with a pronounced peak in July. High detection and isolation rates, together with temporal clustering in mid-2024, indicate a genuine short-ter

Highly sensitive and rapid determination of Coxsackievirus A16 using restriction endonuclease-mediated reverse transcription multiple cross displacement amplification.

BACKGROUND: As a major etiological agent of hand, foot, and mouth disease in humans, Coxsackievirus A16 (CVA16) exerts a detrimental impact on the health of infants and toddlers. In the absence of effective antiviral drugs and vaccines targeting CVA16, a diagnostic strategy with sensitivity, specificity and rapid response is indispensable for the prevention and management of CVA16 infection. RESULTS: In this study, a novel molecular diagnostic approach, coupling real-time fluorescence technique with reverse transcription multiple cross-displacement amplification (RT-MCDA), was developed for t

Enterovirus Testing in Hand, Foot, and Mouth Disease and Herpangina: A Highly Sensitive Single-Round VP4-VP2 Reverse-Transcription Polymerase Chain Reaction Assay with a Redesigned Reverse Primer.

Molecular typing of enteroviruses (EVs) is essential for surveillance of hand, foot, and mouth disease (HFMD). Conventional reverse-transcription polymerase chain reaction (RT-PCR) targeting the VP4-VP2 region can be insufficiently sensitive, reducing the detectability of Enterovirus A (EV-A). We developed a single-round RT-PCR assay using a modified reverse primer design (C3R) for rapid EV detection and genotyping. Sensitivity was evaluated using EV-A71 and poliovirus type 1 reference strains, across 60 EV-positive clinical specimens. The C3R-based assay showed ~1000-fold higher sensitivity f

Modifying Effect of Wind Speed on the Temperature-humidity Interaction Impacting Hand, Foot, and Mouth Disease as Assessed by Years Lived with Disability in Western China.

OBJECTIVE: Hand, foot, and mouth disease (HFMD) transmission is sensitive to temperature-humidity interactions; however, the role of wind speed in modifying these effects remains unknown. This study investigated how wind speed modifies the combined effects of temperature and humidity on HFMD burden and identified subgroups of individuals with increased vulnerability to these climate exposures. METHODS: We analyzed data from 524,100 HFMD cases and daily meteorological measurements across Guizhou, China, between 2012 and 2019. Disease burden was quantified as the number of years lived with disa

Novel NSAID-Se derivative YSN-1-167 against Enterovirus 71 infection by inhibiting 3Dpol activity.

Hand, foot, and mouth disease (HFMD) is primarily caused by Enterovirus 71 (EV71) and Coxsackievirus A16 (CA16), while excessive inflammatory responses induced by viral infection are the main cause of severe conditions. Hence, it is important to develop agents with both antiviral and anti-inflammatory activities for clinical treatment. In this study, we synthesized 20 compounds based on the hybridization of nonsteroidal anti-inflammatory drugs (NSAIDs) and organoselenium and found that YSN-1-167 exhibited significant anti-EV71 function using an EV71-GFP infection system. Antigenic characterization and mapping of a conserved neutralizing epitope on VP1 of coxsackievirus A5.

BACKGROUND: Coxsackievirus A5 (CV-A5) has re-emerged as an important pathogen associated with hand, foot, and mouth disease (HFMD) in young children. However, the antigenic structure of CV-A5 and the key determinants responsible for neutralizing antibody responses remain largely undefined, hindering rational vaccine design and antibody-based intervention strategies. METHODS: To investigate the relationship between viral particle composition and humoral immune responses, three forms of CV-A5 particles, including empty particles (EPs), full particles (FPs), A-particles (APs) were purified from

EV-A71 induces GSDME-dependent T-cell pyroptosis: A novel mechanism and MeCbl therapeutic potential.

Enterovirus A71 (EV-A71) is the primary pathogen causing severe hand-foot-and-mouth disease (HFMD) in young children, with T-cell immune dysfunction closely linked to severe clinical outcomes. However, the molecular mechanisms underlying EV-A71-mediated T-cell impairment remain unclear, and no specific therapies are currently available. Here, we investigated the interaction between EV-A71 and T cells, and explored potential targeted therapeutic strategies. Our results showed that EV-A71 efficiently infects T cell lines (Jurkat, EL-4) and primary mouse CD3+ T cells in a dose- and time-dependent

Onychomadesis: A Rare Manifestation of Coxsackievirus A6 Infection in an Adult Patient.

Hand, foot, and mouth disease (HFMD) is a self-limiting viral illness predominantly affecting children under 10 years of age, with peak incidence in those aged one to four years. Recent outbreaks of Coxsackievirus A6 (CVA6) have increased its incidence across all age groups, including adults. Onychomadesis, a proximal nail plate detachment due to transient matrix arrest, is a recognized but underreported delayed complication of HFMD. We report a case of a 60-year-old man who developed onychomadesis three weeks after clinical recovery from CVA6-induced HFMD. Initial symptoms included fever, sor

Targeted sequence capture of coxsackievirus A6 using nanopore sequencing directly from clinical specimens.

Coxsackievirus A6 (CVA6) has emerged as a predominant causative agent of hand, foot, and mouth disease worldwide. Efficient genomic surveillance is crucial for understanding its evolution and spread. Nanopore sequencing presents a promising platform for rapid and cost-effective viral genome sequencing directly from clinical samples. We developed a tiling amplicon-based nanopore sequencing protocol for CVA6. Eight clinical CVA6-positive samples, spanning a wide range of Ct values (15.54-25.28), were each serially diluted to three concentrations (undiluted, 10⁻¹, and 10⁻²), generating a total of

Moracin M inhibits EV71 infection primarily by blocking viral attachment to host cells.

The global outbreak of hand, foot, and mouth disease (HFMD), mainly caused by enterovirus A71 (EV71) and coxsackievirus A16 (CA16) of the genus enterovirus, poses a serious threat to the health of young children, and severe neurological manifestations and associated symptoms are primarily attributed to EV71 infection. There is currently no specific antiviral drug available for HFMD. In this study, we identified Moracin M (MM), a flavonoid compound derived from Smilax china, as a potent inhibitor of EV71 and CA16 infection. Mechanistic investigation revealed that MM exerts its antiviral effects

Dairy Worker Infection Prevention Knowledge, Beliefs, and Behaviors.

INTRODUCTION: Recent outbreaks of infectious diseases on dairy farms, with workers becoming ill, have focused attention on the potential for zoonotic disease transmission of pathogens between animals and humans in dairy production. While many dairy farms have a biosecurity plan and protocols in place to reduce the possibility of cow-cow transmission of pathogens such as foot-and-mouth disease, previous surveys have indicated that few farms have organized ongoing infection‑prevention programs to prevent the spread of infection to workers. OBJECTIVE: The purpose of the current study is to descr

Impact of EV-A71 vaccination and non-pharmaceutical interventions on hand, foot, and mouth disease: A 14-year interrupted time series analysis.

BACKGROUND: Hand, foot, and mouth disease (HFMD), particularly EV-A71, has caused large-scale epidemics in the Asia-Pacific region, with China alone reporting over 2 million annual cases and causing a substantial morbidity and mortality. While EV-A71 vaccination was introduced in 2016 and COVID-19 non-pharmaceutical interventions (NPIs) were implemented from 2020-2022, comprehensive evidence on their independent and combined effects on HFMD epidemiology remains scarce. We aimed to quantify the impact of vaccination, NPIs, and post-pandemic policy relaxation on HFMD incidence and pathogen distr

Differential immune responses elicited by two tetravalent epitope-based DNA vaccines differently designed for human hand-foot-and-mouth disease.

Hand-foot-and-mouth disease (HFMD) remains a significant public health concern for children. Enterovirus A71 (EV71) and coxsackievirus A16 (CVA16) have been the main causes of HFMD; however, CVA10 and CVA6 have become more prevalent in recent years. A universally effective vaccine that covers all four major enteroviruses is of importance. In this study, we developed and assessed the immunogenicity of two differently designed tetravalent DNA-based HFMD vaccines in a murine model. The new immunogen, EVCVAme, was designed with a string of reported B-cell epitopes from the four enteroviruses, whil

TLR2-Driven NLRP3 Inflammasome Activation Mediates EV71-Induced Neuropathogenesis in Neonatal Mice.

EV71 is a leading cause of HFMD-associated neurological complications, yet its CNS pathogenic mechanisms remain unclear. Using neonatal BALB/c mice, we identified 10 days post-infection (dpi) as the disease peak, characterized by neuronal necrosis and robust NLRP3 inflammasome activation. We demonstrate that EV71 upregulates TLR2, triggering the NF-κB pathway to prime and activate the NLRP3 inflammasome within microglia. This process drives pro-inflammatory cytokine release and neuroinflammatory damage. Pharmacological blockade of TLR2 with C29 suppressed NF-κB phosphorylation and NLRP3 assemb
