Kindler epidermolysis bullosa (KEB), a rare genodermatosis caused by loss-of-function mutations in the FERMT1 gene (encoding kindlin-1), is clinically characterized by congenital blistering, skin atrophy, poikiloderma, photosensitivity, and an increased predisposition to cutaneous squamous cell carcinoma (cSCC). Hearing loss, a common sensory impairment with multifactorial origins, often stems from genetic or environmental factors. Here, we report a unique familial case in which one sibling has KEB complicated by cSCC, and the other has congenital autosomal recessive non-syndromic hearing loss

Parry-Romberg syndrome (PRS) is a rare, acquired neurocutaneous disorder characterized by progressive hemifacial atrophy, primarily affecting the skin, soft tissue, and underlying bone. The etiology remains unclear, though autoimmune, inflammatory, and genetic factors have been implicated. PRS frequently coexists with localized scleroderma (en coup de sabre), leading to diagnostic and therapeutic challenges. Here, we present two pediatric cases from tertiary care hospitals in Oman, emphasizing clinical presentation, diagnostic findings, and treatment response. The first patient, a 10-year-old

BACKGROUND: Infections have been associated with a greater risk of Alzheimer’s disease and related dementias (ADRD), but it is unclear how infections influence structural brain patterns over time, and whether post-infection brain atrophy can accelerate cognitive decline and molecular changes underlying dementia. METHODS: Using the Baltimore Longitudinal Study of Aging (BLSA; n = 793; mean age = 70.1), we examined how infections relate to longitudinal changes in machine learning-derived, 3 T-MRI neuroimaging signatures, and leveraged the UK Biobank (UKB; 1,120; mean age = 62.9 yrs) to externall

Sturge-Weber syndrome is a rare neurocutaneous disorder due to a somatic GNAQ mutation, which is characterised by facial capillary malformation, leptomeningeal angiomatosis, and possible ocular involvement. Identifying these faint birthmarks is important because even mild skin involvement may indicate significant underlying intracranial vascular malformations and early neurological complications. We report a seven-year-old boy with focal seizures beginning at two years of age, often progressing to generalised tonic-clonic events, and associated with intermittent left-sided headaches. Examinati

Sturge-Weber syndrome is a rare congenital disorder with vascular malformations affecting the brain, skin, and eyes. Type 3 Sturge-Weber syndrome, the rarest form, presents with isolated leptomeningeal angiomas, often causing diagnostic delays. This report highlights an unusual presentation where both recurrent focal seizures and migraine-like headaches were prominent, long-standing features. A 30-year-old man presented with two focal seizures affecting the right side of his body, preceded by aura and followed by postictal confusion, right-sided weakness, and migraine-like headache with photop

Anetoderma is a rare cutaneous disorder characterized by focal loss of dermal elastic tissue, often presenting as atrophic or herniated plaques. However, clinical presentations can vary, and atypical forms may resemble neoplastic or inflammatory lesions, complicating diagnosis. We present the case of a 78-year-old female who was evaluated for asymptomatic skin growths on the posterior neck, chest, and torso, reported to have been present for five years and slowly spreading. On physical examination, firm papules and subcutaneous nodules were noted on the left rib cage, left medial breast, and m

Spinal muscular atrophy (SMA) is a neurodegenerative disease caused by homozygous loss of the SMN1 gene. Copy number of the nearly identical paralog, SMN2, correlates with disease severity. SMN2 is the primary modifier of SMA, with only a few other modifiers reported. We reviewed the literature of rare siblings affected by SMA that show discordance in clinical presentation despite having the same number of SMN2 copies, which predicts the presence of genetic modifiers. We further recruited a sibling pair with discordant clinical presentations and performed detailed characterization. We utilized

Flegel disease, also known as hyperkeratosis lenticularis perstans, is a rare cutaneous disease. Here, we report a case of a 56-year-old man who presented with a history of asymptomatic skin lesions that had gradually increased in number over the past year, in the absence of any significant medical history or known predisposing factors for Flegel disease. Skin examination revealed multiple punctate, scaly, blanchable, erythematous papules measuring 1-2 mm in diameter and distributed symmetrically on both lower extremities. Scales were adherent to the peripheral edges of the papules. Palms, sol

Parry-Romberg syndrome (PRS), also known as progressive facial hemiatrophy, is a rare neurocutaneous disorder characterized by progressive, unilateral atrophy of the facial tissues. It usually appears during childhood or early adulthood; however, adult-onset cases are increasingly being recognized. PRS significantly overlaps with localized scleroderma, especially the en coup de sabre (ECDS) subtype, indicating a common pathophysiological mechanism. We present a 52-year-old female patient with adult-onset PRS associated with ECDS. The patient was first diagnosed with localized scleroderma in 19

Recent studies have identified phosphorylated α-synuclein in the skin of individuals with neurodegenerative disease. The levels of this protein in the skin have been correlated with disease severity. This protein has been specifically studied in alpha-synucleinopathies such as Parkinsons' Disease (PD) and Multiple System Atrophy (MSA). Given that skin biopsy studies have often shown high levels of phosphorylated α-synuclein in the neck area, and that other clinical studies have described easily identifiable changes in facial skin features, this systematic review explores whether age spots, whi

Progressive hemifacial atrophy, also known as Parry-Romberg syndrome (PRS), is an idiopathic disorder characterized by progressive unilateral facial depression caused by atrophy of the skin and subcutaneous tissue. We present a case of mandibular osteomyelitis occurring in a patient with this condition, together with a review of the literature. The clinical findings in this case suggest that autoimmune disease and trauma are involved in contributing factors to disease progression. Possible involvement of neurovascular and neuroskeletal dysregulations in the pathogenesis of PRS is also discusse

Necrobiosis lipoidica (NL) is a chronic granulomatous disease most commonly manifesting as yellow, atrophic plaques and ulcerations on the lower extremities. We present an exacerbation of NL in a patient with a known history of glucose-6-phosphate dehydrogenase deficiency, who, following a clinical and histopathologic diagnosis of NL, was found to have oropharyngeal Epstein-Barr virus-positive diffuse large B-cell lymphoma. After implementation of therapies for lymphoma, chemotherapeutic complications, and NL, the patient reported improvement in both pain and ulcerations. Though noninfectious

Mal de Meleda (MDM) is a rare hereditary skin disorder classified as an autosomal recessive palmoplantar keratoderma, with an estimated prevalence of 1 in 100,000 individuals. It is commonly linked to consanguineous parentage and typically presents during early childhood. The hallmark features of the condition include transient thickening of the palms and soles, skin sclerosis and atrophy, scleroatrophic erythematous lesions, pseudoainhum formation around the digits, and erythema around the mouth. Due to its rarity, many dermatologists may lack clinical familiarity with MDM. Moreover, there is

The mineralocorticoid receptor (MR) plays a pivotal role in skin homeostasis, inflammation, and repair, interacting closely with the glucocorticoid receptor (GR) to regulate various physiological and pathological processes. Dysregulation of MR signaling has been implicated in several skin disorders, including psoriasis, atopic dermatitis, and impaired wound healing. Furthermore, studies have shown that patients with primary hyperaldosteronism exhibit epidermal hyperplasia, impaired differentiation, increased immune cell infiltrates, and elevated pro-inflammatory cytokines due to MR overactivat

BACKGROUND: The nuclear-encoded enzyme polymerase gamma (Pol-γ) is crucial in the replication of the mitochondrial genome (mtDNA), which in turn is vital for mitochondria and hence numerous metabolic processes and energy production in eukaryotic cells. Variants in the POLG gene, which encodes the catalytic subunit of Pol-γ, can significantly impair Pol-γ enzyme function. Pol-γ-associated disorders are referred to as POLG-spectrum disorders (POLG-SDs) and are mainly autosomal-recessively inherited. Clinical manifestations include muscle weakness and fatigue, and severe forms of the disease can
