Phototherapy of granuloma annulare: a single center retrospective study.

BACKGROUND: Granuloma annulare is a chronic, benign skin condition characterized by small erythematous patches, plaques or papules in annular or disseminated order. Due to the cosmetic impact, many patients experience a significant level of distress. Nevertheless, selecting the optimal treatment method often poses a challenge due to the limited available data. A remission of skin lesions can be achieved through topical, systemic, or intralesional therapies, and various forms of phototherapy. PATIENTS AND METHODOLOGY: This retrospective study included 58 patients diagnosed with granuloma annul

Systemic Janus kinase inhibitors in the management of granuloma annulare.

Granuloma Annulare (GA) is an inflammatory granulomatous disorder that is typically localized to the skin. First line therapies for localized GA include topical and intralesional corticosteroids. Systemic corticosteroids have been used for generalized; however, rates of partial and complete resolution are disappointing. Recent advances in understanding the pathophysiology of macrophage activation and granuloma formation have led to Janus Kinase (JAK) inhibition as a therapeutic target for GA. The objectives of our study were to provide insight into the pathophysiology of GA and describe the cl

Clinicopathologic Analysis of Granuloma Annulare With Insights Into Its Incidence and Demographics in the United Arab Emirates From 2016 to 2023.

Granuloma annulare (GA) is a benign, often chronic dermatologic condition with varying clinical presentations and has been associated in the literature with a notable psychosocial burden, particularly in generalized cases. This retrospective cross-sectional study analyzed the epidemiological and clinicopathological features of GA and its variants in the United Arab Emirates (UAE) from 2016 to 2023. A total of 171 histopathologically confirmed GA cases were included, collected from a single dermatopathology center. The incidence of GA diagnoses showed temporal variation, peaking in 2020 at 0.67

SPP1hi macrophages, NKG7 T cells, CCL5hi fibroblasts, and IgM plasma cells are dominant features of necrobiosis.

Necrobiosis is a histologic term used to describe abnormal deposits of "degenerating" collagen within the skin. It can be found as an incidental finding in various granulomatous conditions, but is a hallmark of necrobiosis lipoidica (NL) and necrobiotic xanthogranuloma (NXG). There is limited prior research on necrobiosis. Here, we employed single-cell analysis of lesional and nonlesional skin to study the pathophysiology of necrobiosis. Our findings demonstrate that necrobiotic lesional skin is characterized by SPP1hi macrophages expressing MARCO; NKG7-expressing effector CD8+ T cells coexpre

Cutaneous manifestations of diabetes mellitus: a narrative review.

Diabetes mellitus is a highly prevalent human endocrine disorder. Skin lesions are reported in approximately one-third of all diabetes mellitus patients. The clinical presentation and frequency vary according to the subtype of diabetes mellitus, metabolic control, and clinical course, with certain skin diseases occurring before diagnosing hyperglycemia. In this regard, the correct definition of cutaneous manifestations associated with diabetes mellitus can help define the etiology of hyperglycemia as well as the need to optimize glycemic control. In this narrative review, the most common cutan

Psoriasis in Obese Adolescents with Diabetes-From Common Molecular Background to Vicious Circle of Metabolic Syndrome-Case Report and Review of Literature.

Psoriasis and type 1 diabetes mellitus (T1DM) are chronic autoimmune diseases sharing common immunological pathways, particularly the involvement of interleukin 17 (IL-17), driving Th17-mediated inflammation. This review explores the overlap between psoriasis, obesity, T1DM, and necrobiosis lipoidica (NL), a skin condition associated with diabetes. Obesity exacerbates inflammation through immune cell activation in adipose tissue and the release of proinflammatory adipokines, such as leptin, resistin, and IL-18, which enhance autoimmune responses and insulin resistance. Leptin promotes the diff

Thrombocytopenia in myelofibrosis is characterized by inflammatory megakaryocytes with reduced G6B expression.

The megakaryocytic (MK)-specific immunoreceptor G6b-B plays an essential role in MK development. Because germ line loss-of-function mutations of G6b-B in humans and its deletion in mouse models lead to thrombocytopenia and a myelofibrosis-like clinical phenotype (MF-MPIG6B), we explored the role of G6b-B in patients with myelofibrosis (MF) due to a myeloproliferative neoplasm (MPN) with thrombocytopenia (MPN-MF-T). We demonstrated that MKs generated from mononuclear cells (MNCs) from a patient with MF-MPIG6B as well as patients with MPN-MF-T failed to express GATA binding protein 1 and G6B and
